As the sophistication and sensitivity of chemical instrumentation increase, so does the number of applications. Correspondingly, new questions and opportunities for systems previously studied also arise. As with most plants, the emission of volatiles from agricultural products is complex and varies among commodities. Volatiles are indicative of characteristics such as food quality, cultivar type, phenological stage, and biotic and abiotic stressors; thus, their systematic and accurate evaluation is important. Early volatile analyses entailed removal of the sample matrix in question, transport to the laboratory, and subsequent investigation. More recently, scientists are moving the laboratory to the field to obtain realistic emission patterns of the sample in its natural environment. This perspective proposes that a methodical and collaborative approach to the complex relationship between volatiles and agricultural commodities and their various phenological stages, oxidative degradation products, and fungal contamination is needed in order to fully comprehend the sample and associated relationships as a whole. These methodical approaches should incorporate both in situ and ex situ investigations of the sample. Ultimately, there exists an opportunity for development of methodologies that integrate both laboratory- and field-based collection of volatiles to explore and address the complex biological interactions of agricultural systems. 相似文献

Two complementary experiments were completed to assess short-term foraging dynamics, diet quality, and ruminal degradation kinetics of herbage consumed by steers with 3 levels of herbage depletion. Experiment 1 was a behavioral study in which 2 ruminally cannulated steers were allocated to grazing scenarios simulating 3 levels of herbage depletion. These treatments included an ungrazed sward (control), as well as medium and high levels of herbage depletion. Grazing scenarios were sampled for sward surface height and amount of green leaf and stem before being grazed. Foraging dynamics were determined through measurements of bite rate, bite depth, eating step rate, eating distance, potential area consumed while grazing, and bites and intake per eating step. Also, quality of potential herbage consumed was estimated from hand-plucked herbage. In Exp. 2, ruminal degradation kinetics of DM for samples of herbage consumed (masticate) by steers during Exp. 1 were assessed in situ using 5 ruminally cannulated steers. The immediately soluble, degraded, and undegraded DM fractions were determined. The DM disappearance rate and lag times were determined from a nonlinear regression model, and the effective degradability of DM was calculated. Herbage depletion resulted in increased eating steps/minute, as well as the potential area harvested while grazing (P < 0.05) and reduced herbage intake/eating step (P < 0.05). Neither the herbage potentially consumed nor the ruminal degradation kinetics was affected by extent of depletion (P > 0.05). Under these experimental conditions, steers adapted their foraging dynamic and were able to sustain diet quality in the short term. These results imply that behavioral adaptations would make diet quality less sensitive to certain levels of herbage depletion. 相似文献

Methicillin-resistant Staphylococcus aureus (MRSA) have emerged worldwide and have become resistant to a variety of antibiotics. MRSA colonisation in pigs was first reported from the Netherlands in 2005, where pigs were implicated as a source of human MRSA infections (Voss et al., 2005). This paper presents the first report on the presence of MRSA on large pig breeding farms in Croatia, together with the determination of the mecA gene, the results of spa typing and susceptibility to commonly used antimicrobials. Dust samples (7-11 per farm) were collected from eight large pig farms in Croatia. Of the total 68 swabs, the mecA gene was detected in 24 isolates growing on the MRSA agar. All isolates were resistant to oxacillin, tetracycline and streptomycin, and susceptible only to vancomycin, while 92% of the strains were susceptible to ciprofloxacin. Genotyping of the MRSA strains was performed by spa typing, and revealed t011 (n = 17), t034 (n = 5) and t1451 (n = 2). The impact of manipulating ruminal fill (RF) on intake rate of herbage and grazing dynamics was measured with three rumen‐cannulated beef heifers grazing Bermudagrass pastures individually. The treatments compared were removal of proportions of rumen contents of 0 (treatment RF0), 0·33 (treatment RF33), 0·66 (treatment RF66) and 1·00 (treatment RF100). Treatments were randomly applied in a 3 × 4 Youden‐square design. The rumens were emptied before and after planned grazing sessions (30 min) to set up the treatments, and to estimate intake rate and bite mass, respectively. Measurements were made of bite rate, bites per feeding station, feeding stations per minute, intake per feeding station, time per feeding station, eating and searching step rates and times. Apparent bite area and area grazed per feeding station were calculated. Ruminal fill affected short‐term intake rate and changed grazing dynamics. As RF increased, step rates, searching times, bite mass, apparent bite area, bites per feeding station, area grazed per feeding station, time per feeding station and intake per feeding station decreased (P < 0·01) while step times, eating step rates and bite depth increased (P < 0·01). The results of the present study indicate that RF is an important factor governing the intake characteristics and behaviour of grazing beef heifers. 相似文献

A mucosal lesion was created in the center of each test sinus of 6 mature, healthy, nonlactating Holstein cows by resecting a circumferential band of mucosa. Each lesion was then treated by implantation of strip grafts of autogenous oral mucosa, temporary silastic tube implant, or a combination of strip grafts and temporary silastic tube implant. All teats were evaluated for patency 6 weeks after treatment, and tube implants were removed through a second thelotomy incision. All teats were reevaluated for gross and radiographic patency 12 weeks after treatment, and teats were collected for histologic evaluation of lesions. All 4 teats treated with grafts only were obstructed at 6 and 12 weeks after treatment. Incomplete coverage of the lesion with mucosa was observed in all 4 teats. The major source of obstruction was proliferation of epithelium and keratin into the lumen. All 8 teats treated with temporary silastic tube implants alone were patent at 6 weeks after treatment, but were obstructed at 12 weeks after treatment. Foci of mucosa at the lesion site were detected in only 2 of the 8 teats. Obstruction resulted from proliferation of granulation tissue into the lumen. All 12 teats treated with grafts and a temporary tube implant were patent at 6 weeks after treatment and 11 of 12 were patent at 12 weeks after treatment, although marked luminal narrowing was evident in 9 of 11 teats. Partial to complete coverage of the lesion with mucosa was seen in all teats. Proliferative granulation tissue, epithelium, and keratin contributed to luminal narrowing in 10 of 11 patent teats. Bacteriologic culture of quarters from 6 of the 11 teats patent at the final evaluation yielded pathogens. 相似文献

Four cows with clinically intact udders in high lactation were intracisternally infected by a pathogenic field strain of Escherichia coli. Infections were induced by quarters and with time intervals. Hence, when the animals were slaughtered six hours from beginning of the experiment, epithelial samples could be collected from cisternal walls and lactiferous ducts which, by the time of sampling, had been in contact with pathogens for one, four, and six hours. Highly acute mastitis began to develop two hours from experimental infection and exhibited typical clinical symptoms, including general disorders, unambiguous alterations to the white blood count, and changes in milk composition. These changes were typical of acute toxin action on the udder quarters. Concomitant reactions of non-infected control quarters, detectable by clinical manifestations or by laboratory diagnosis, were not observed. 相似文献
