A high-resolution paleoclimate record spanning the past 25,000 years in southern East Africa

High-resolution profiles of the mass accumulation rate of biogenic silica and other geochemical proxies in two piston cores from northern Lake Malawi provide a climate signal for this part of tropical Africa spanning the past 25,000 years. The biogenic silica mass accumulation rate was low during the relatively dry late Pleistocene, when the river flux of silica to the lake was suppressed. Millennial-scale fluctuations, due to upwelling intensity, in the late Pleistocene climate of the Lake Malawi basin appear to have been closely linked to the Northern Hemisphere climate until 11 thousand years ago. Relatively cold conditions in the Northern Hemisphere coincided with more frequent north winds over the Malawi basin, perhaps resulting from a more southward migration of the Intertropical Convergence Zone.     

Self-Assembled Smectic Phases in Rod-Coil Block Copolymers

Rod-coil block copolymers are self-assembling polymers that combine the physics of orientational ordering of rodlike polymers and the microphase separation of coil-coil block copolymers. Several new solid-state morphologies were observed in a series of anionically synthesized model poly(hexyl isocyanate-b-styrene) rod-coil diblock copolymers examined by transmission electron microscopy and selected-area electron diffraction. The rod-coils formed smectic C-like and O-like morphologies with domain sizes ranging from tens of nanometers to almost 1 micrometer. Both structural and orientational changes were found for increasing rod volume fractions. In addition, some morphologies exhibited spontaneous long-range orientational order over many tens of micrometers.     

The deglacial evolution of North Atlantic deep convection

Deepwater formation in the North Atlantic by open-ocean convection is an essential component of the overturning circulation of the Atlantic Ocean, which helps regulate global climate. We use water-column radiocarbon reconstructions to examine changes in northeast Atlantic convection since the Last Glacial Maximum. During cold intervals, we infer a reduction in open-ocean convection and an associated incursion of an extremely radiocarbon ((14)C)-depleted water mass, interpreted to be Antarctic Intermediate Water. Comparing the timing of deep convection changes in the northeast and northwest Atlantic, we suggest that, despite a strong control on Greenland temperature by northeast Atlantic convection, reduced open-ocean convection in both the northwest and northeast Atlantic is necessary to account for contemporaneous perturbations in atmospheric circulation.     

Chromosome 17 deletions and p53 gene mutations in colorectal carcinomas

Previous studies have demonstrated that allelic deletions of the short arm of chromosome 17 occur in over 75% of colorectal carcinomas. Twenty chromosome 17p markers were used to localize the common region of deletion in these tumors to a region contained within bands 17p12 to 17p13.3. This region contains the gene for the transformation-associated protein p53. Southern and Northern blot hybridization experiments provided no evidence for gross alterations of the p53 gene or surrounding sequences. As a more rigorous test of the possibility that p53 was a target of the deletions, the p53 coding regions from two tumors were analyzed; these two tumors, like most colorectal carcinomas, had allelic deletions of chromosome 17p and expressed considerable amounts of p53 messenger RNA from the remaining allele. The remaining p53 allele was mutated in both tumors, with an alanine substituted for valine at codon 143 of one tumor and a histidine substituted for arginine at codon 175 of the second tumor. Both mutations occurred in a highly conserved region of the p53 gene that was previously found to be mutated in murine p53 oncogenes. The data suggest that p53 gene mutations may be involved in colorectal neoplasia, perhaps through inactivation of a tumor suppressor function of the wild-type p53 gene.     

Supraopticneurosecretory cells: autonomic modulation

Neurosecretory cells in the supraoptic nuclei of anesthetized cats were antidromically identified by electrical stimulation of the posterior pituitary. The responses of these units to afferent volleys from vagal and carotid sinus nerves were examined with a computer of average transients. Synaptic excitation of neurosecretory cells by stimulation of these pathways demonstrates the existence of excitatory inputs from vagal and carotid sinus nerve afferents. Since these pathways are involved in the release of antidiuretic hormone, the results support the hypothesis that its release is related to an increase in discharge frequency of supraoptic neurosecretory cells.     

Analysis of fenbendazole residues in bovine milk by ELISA

Fenbendazole residues in bovine milk were analyzed by ELISAs using two monoclonal antibodies. One monoclonal antibody (MAb 587) bound the major benzimidazole anthelmintic drugs, including fenbendazole, oxfendazole, and fenbendazole sulfone. The other (MAb 591) was more specific for fenbendazole, with 13% cross-reactivity with the sulfone and no significant binding to the sulfoxide metabolite. The limit of detection of the ELISA method in the milk matrix was 7 ppb for MAb 587 and 3 ppb for MAb 591. Fenbendazole was administered in feed, drench, and paste form to three groups of dairy cattle. Milk was collected immediately before dosing and then every 12 h for 5 days. The ELISA indicated that residue levels varied widely among individual cows in each group. Fenbendazole levels peaked at approximately 12-24 h and declined rapidly thereafter. Metabolites were detected at much higher levels than the parent compound, peaked at approximately 24-36 h, and declined gradually. Residue levels were undetectable by 72 h. The ELISA data correlated well with the total residues determined by chromatographic analysis, but the use of the two separate ELISAs did not afford an advantage over ELISA with the single, broadly reactive MAb 587. The ELISA method could be used to flag high-residue samples in on-site monitoring of fenbendazole in milk and is a potential tool for studying drug pharmacokinetics.     

Quantitative analysis of production traits in saltwater crocodiles (Crocodylus porosus): II. age at slaughter

Crocodile morphometric (head, snout‐vent and total length) measurements were recorded at three stages during the production chain: hatching, inventory [average age (±SE) is 265.1 ± 0.4 days] and slaughter (average age is 1037.8 ± 0.4 days). Crocodile skins are used for the manufacture of exclusive leather products, with the most common‐sized skin sold having 35–45 cm in belly width. One of the breeding objectives for inclusion into a multitrait genetic improvement programme for saltwater crocodiles is the time taken for a juvenile to reach this size or age at slaughter. A multivariate restricted maximum likelihood analysis provided (co)variance components for estimating the first published genetic parameter estimates for these traits. Heritability (±SE) estimates for the traits hatchling snout‐vent length, inventory head length and age at slaughter were 0.60 (0.15), 0.59 (0.12) and 0.40 (0.10) respectively. There were strong negative genetic (?0.81 ± 0.08) and phenotypic (?0.82 ± 0.02) correlations between age at slaughter and inventory head length.
