Symptomatically distinct isolates of potato leaf roll virus were transmitted from White Rose to Russet Burbank potato by aphids and by grafting, from White Rose potato toP. floridana by aphids, and fromP. floridana toP. floridana by aphids. Isolates from White Rose potato generally produced stable and characteristic secondary leaf roll symptoms (but not primary leaf roll symptoms) when transmitted to Russet Burbank potato but variable symptoms when transmitted toP. floridana. Isolates from two of the latter plants with moderate and severe symptoms generally incited stable and characteristic symptoms when transmitted to otherP. floridana plants. The stable symptoms produced by each of these isolates were not modified by infestingP. floridana plants with varying numbers of viruliferous aphids. The results obtained in this study corroborate previous evidence for the existence of strains of potato leaf roll virus. It is suggested that certain White Rose potato plants contained mixtures of leaf roll virus strains. Furthermore, it is postulated that these mixtures were at least partially separated by “natural” selection following transmission toP. floridana. This mechanism provides an explanation for the stable symptoms produced by isolates transmitted fromP. floridana toP. floridana.  相似文献

In young grass plants it is possible to relate the weight of the root to that of the shoot through the allometric formula y = bx k . Using this formula it was found that flower formation was associated with an increase in shoot growth relative to that of the root. Within varieties of Lolium , the greater the intensity of heading the more was the growth of the shoot relative to that of the root. Perenniality was associated with a high rate of root growth relative to that of the shoot. A decrease in the light intensity or soil water reduced the growth of the plant as a whole, but light decreased root growth more than that of the shoot while water supply had the opposite effect.  相似文献

A naturally new cyclopeptide, clavatustide C, was produced as a stress metabolite in response to abiotic stress elicitation by one of the hydrothermal vent fluid components Zn in the cultured mycelia of Aspergillus clavatus C2WU, which were isolated from Xenograpsus testudinatus. X. testudinatus lives at extreme, toxic habitat around the sulphur-rich hydrothermal vents in Taiwan Kueishantao. The known compound clavatustide B was also isolated and purified. This is the first example of a new hydrothermal vent microbial secondary metabolite produced in response to abiotic Zn treatment. The structures were established by spectroscopic means. The regulation of G1-S transition in hepatocellular carcinoma cell lines by clavatustide B was observed in our previous study. The purpose of the present study was to verify these results in other types of cancer cell lines and elucidate the possible molecular mechanism for the anti-cancer activities of clavatustide B. In different human cancer cell lines, including pancreatic cancer (Panc-1), gastric cancer (MGC-803), colorectal cancer (SW-480), retinoblastoma (WERI-Rb-1) and prostate cancer (PC3), clavatustide B efficiently suppressed cell proliferations in a dose-dependent manner. Although different cancer cell lines presented variety in Max effect dose and IC50 dose, all cancer cell lines showed a lower Max effect dose and IC50 dose compared with human fibroblasts (hFB) (p < 0.05). Moreover, significant accumulations in G1 phases and a reduction in S phases (p < 0.05) were observed under clavatustide B treatment. The expression levels of 2622 genes including 39 cell cycle-associated genes in HepG2 cells were significantly altered by the treatment with 15 μg/mL clavatustide B after 48 h. CCNE2 (cyclin E2) was proved to be the key regulator of clavatustide B-induced G1-S transition blocking in several cancer cell lines by using real-time PCR.  相似文献

A method of regiospecific analysis of triacylglycerols (TAGs) in vegetable oils and animal fats is reported here using the electrospray ionization-mass spectrometry (MS(3)) of TAG-lithiated adducts. The fragment ions of the MS(3) from the loss of fatty acids at the sn-2 position as alpha,beta-unsaturated fatty acids were used for regiospecific identification and quantification. The ratio of the regiospecific TAGs, ABA and AAB, in an oil sample usually fraction collected by high-performance liquid chromatography can be determined by the abundance of the fragment ions of [ABA + Li-ACOOH-B'CH=CHCOOH]+ and [AAB + Li-ACOOH-A'CH=CHCOOH]+. The method was used to analyze regiospecific TAGs in extra virgin olive oil. The results showed that the saturated fatty acids, palmitic and stearic acids, were mostly located at the sn-1,3 positions and unsaturated fatty acids, oleic and linoleic acids, were mostly located at the sn-2 position.  相似文献

A semi-annual eelgrass (Zostera marina L.) population became extinct in 2004. It had flourished for many decades at Terschelling in the western Wadden Sea, one of the most eutrophied locations where seagrass growth has been recorded. Semi-annual populations survive the winter season by seed (annual), and by incidental plant survival (semi-annual). We compared seed bank dynamics and fate of plants between this impacted site and a reference site in the winter of 1990-1991. Seed bank density at Terschelling was extremely low (5-35 seeds m−2) in comparison to the reference site (>60 seeds m−2) and also in comparison to seed bank densities of (semi-)annual eelgrass populations in other parts of the world. Plant survival during winter was nil. Nevertheless, the population more than doubled its area in 1991, implying maximum germination and seedling survival rates. However, from 1992 onwards the decline set in and continued - while the nutrient levels decreased. To establish the cause of the low seed bank density, we conducted a transplantation experiment in 2004 to study the relationship between seed production and macro-algal cover. The transplantation experiment showed a negative relationship between the survival of seed producing shoots and suffocation by macro-algae, which is associated with light limitation and unfavourable biogeochemical conditions. The plants died before they had started to produce seeds. Thus, it is likely that macro-algal cover was responsible for the low seed bank density found in Terschelling in 1990-1991. Both the recorded low seed bank density and absence of incidental plant survival during winter were related to eutrophication. These parameters must have been a severe bottleneck in the life history of the extinct population at the impacted site, particularly as Z. marina seed banks are transient. Therefore we deduce that this population had survived at the edge of collapse, and became extinct after a small, haphazard environmental change. We argue that its resilience during these years must have been due to (i) maximum germination and seedling survival rates and (ii) spatial spreading of risks: parts of the population may have survived at locally macro-algae-free spots from where the area could be recolonised. As a consequence, the timing of the collapse was unpredictable and did not synchronise with the eutrophication process. The lesson learnt for conservation is to recognise that eutrophication may be a cause for seagrass population collapse and its eventual extinction, even years after nutrient levels stabilised, or even decreased.  相似文献

The belief that high income is associated with good mood is widespread but mostly illusory. People with above-average income are relatively satisfied with their lives but are barely happier than others in moment-to-moment experience, tend to be more tense, and do not spend more time in particularly enjoyable activities. Moreover, the effect of income on life satisfaction seems to be transient. We argue that people exaggerate the contribution of income to happiness because they focus, in part, on conventional achievements when evaluating their life or the lives of others.  相似文献

The insecticidal effect of low temperatures for the control of all life stages of two stored-product insects, Plodia interpunctella and Liposcelis bostrychophila, was evaluated under laboratory conditions. The temperatures tested were 0, ??5, ??10 and ??15 °C, and, at these temperatures, the insects were exposed for 2, 4 and 8 h and also for 1, 2, 3 and 7 days. Regarding P. interpunctella, the most cold-tolerant life stage was larvae, given that 2 days of exposure were needed for complete (100%) mortality at ??10 °C. Moreover, all larvae were dead at ??15 °C even after 2 h of exposure. Eggs of P. interpunctella were susceptible to cold, as mortality was complete after 7, 1 day, 2 and 2 h at 0, ??5, ??10 and ??15 °C, respectively. L. bostrychophila was by far less susceptible than P. interpunctella. For this species, adults were still alive even after 7 days at ??10 °C, while complete mortality was achieved only after 1 day at ??15 °C. Eggs of L. bostrychophila were the most cold-tolerant life stage, as survival was observed even after 3 days at ??15 °C. The results of the present study provide the first data set on which all life stages of P. interpunctella and L. bostrychophila are evaluated for their susceptibility to cold treatments in a standardized series of exposures and temperatures. These results are expected to further encourage the “real world” application of cold treatments for the disinfestation of durable stored-products.  相似文献

Recently, the eastern region of the Maranhão state (Northeastern Brazil) became a hotspot of land-use change (LUC) directly from native vegetation to soybean cultivation, but due to the soil characteristics, LUC has caused substantial soil organic carbon (SOC) and nitrogen depletion. Therefore, we quantified these impacts arising from two factors: (i) different timeframes after LUC and (ii) contrasting soil management practices. For the first study, soil samples (0–30 cm; six replicates) were taken on soybean fields year one, year eight and 15 years after LUC. It the second study, another area was sampled, of which part was managed under no-tillage (NT) and the other using a mouldboard plough (MP). For both studies, native vegetation (NV) was sampled as the control. NV stored about 50 Mg of carbon (C) ha−1; but LUC reduced C stocks by 35% (after 8 and 15 years); moreover, labile-C decreased between 20% and 45%, while, microbial-C decreased between 20% and 60%, considering the interval between year one and 15 years. Regarding soil management, the MP did not cause differences on C stock (24 Mg C ha−1) in comparison to NT; however, both labile-C and microbial-C decreased by 15% to NT, while, decreased by 40% to MP. These results lead us to believe that, since LUC is inevitable, we suggested the adoption of the best agricultural management practices, in order to preserve/increase the SOC, reducing the impacts on GHG emissions and, thus, achieving sustainability and profitability.  相似文献
