Information on which management practices can enhance soil organic matter (SOM) content and quality can be useful for developing sustainable crop production systems. We tested the influence of 12 years of no-till (NT) versus conventional tillage (CT), and four crop sequences on the organic C pools of a Grey Luvisolic sandy loam soil in northwestern Alberta, Canada. The crop sequences were: continuous wheat (Triticum aestivum L.), field pea (Pisum sativum L.)–wheat–canola (Brassica rapa L.)–wheat, red clover (Trifolium pratense L.) green manure–wheat–canola–wheat/red clover and fallow–wheat–canola–wheat. Soil samples from 1992, when the study was initiated, and 1996, 2000 and 2004 were analyzed for total organic C (TOC), the light fraction (LF) and its C content, and water-soluble and mineralizable C. Total organic C in the top 15 cm of soil was higher in the red clover rotation than either the pea or fallow rotation by 1996. The tillage effect became significant only in 2004 with NT having a higher TOC than CT. The LF dry matter (DM) increased from 6.9 g kg−1 soil in 1992 to a range of 10–13 g kg−1 in 2000 and 2004. It was higher under NT than CT in 2 of 3 years and in the red clover rotation than the pea or fallow rotation in 1 of 3 years. The LF C content exhibited a similar trend as LF DM. The water-soluble and mineralizable C pools were not affected by tillage but decreased with time. Among crop rotations, the red clover rotation tended to result in higher levels of hot water-soluble and mineralizable C. It is concluded that tillage had a greater influence than crop rotation on the LF DM and LF C (as indicators of C storage), whereas the converse effect applied to mineralizable C and, to a lesser degree, hot water-soluble C (as indicators of SOM quality).  相似文献

Several fungi were isolated from rhizosphere soils of various crops through enrichment on l-methionine (l-MET) as a sole C and N source. These fungi were examined for their C2H4 production potential in vitro. GC-FID analysis indicated that 78%, 83%, 89% and 72% of fungi isolated from rhizosphere soil of wheat, maize, potato and tomato, respectively, produced C2H4 from substrate l-MET. Eight of the most efficient C2H4-producing fungal isolates (two from each crop) were further tested for their ability to produce C2H4 from 2-keto-4-methylthiobutyric acid (KMBA) and α-ketoglutaric acid (α-KGA). Interestingly, all eight fungi produced C2H4 from KMBA, but none used α-KGA as C2H4 precursor. This result implies that the MET→KMBA→C2H4 pathway was most likely operating in these fungi. The most efficient C2H4 producer fungus (isolated from maize rhizosphere soil) was identified as Aspergillus terreus; this isolate was further studied to determine the optimal conditions for C2H4 production from l-MET. It was observed that a substrate (l-MET) concentration of 10 mmol l−1, a glucose (C-source) concentration of 6.0 g L−1, no nitrogen, a pH of 6.0, an incubation temperature of 30 °C and an incubation time of 72 h were optimal conditions for l-MET-derived C2H4 biosynthesis by Aspergillus terreus. In addition, C2H4 released from precursor (l-MET) by Aspergillus terreus significantly affected the seedling length and stem diameter of etiolated pea seedlings in both sterilized and non-sterilized soils compared to a control. C2H4-producing microflora may be ubiquitous in soil, and the availability of a substrate like l-MET could enhance C2H4 synthesis in the rhizosphere of a plant, in turn evoking a physiological response. As far as we know, this study represents the first comprehensive screening of rhizosphere fungi for their ability to produce C2H4 from l-MET and KMBA.  相似文献

Insight into nutrient cycling is gained by understanding the dynamics and quantifying nutrient mineralization from decomposing crop residues. Since wheat (Triticum aestivum L.), canola (Brassica napus L.) and pulse crops such as pea (Pisum sativum L.) are commonly grown in rotation, our objectives were to: (1) compare, using the mesh bag technique, the dry matter (DM) loss and release of N and P of straw and root residues of those crops in the 10-11 months following harvest, and (2) determine the influence of N fertilizer on residue decomposition and nutrient release. The no-tillage study started in autumn 1997 when straw residues were placed on the soil surface and root residues were buried in the soil, and sampled periodically through the 1998 growing season. Wheat was grown in 1998 and received 0 or 60 kg N ha-1. The study was repeated in 1998/1999. Wheat straw decomposed more slowly than canola or pea straw (losing an average of 12%, 24% and 25%, respectively, of initial DM in 10-11 months), however, the converse was noted for root residues (42%, 26% and 19% of initial DM). Average net N mineralization from wheat, canola and pea straw was essentially 0, 0.7 and 5.6 kg N ha-1, respectively. Phosphorus released from straw ranged from 0.5 kg ha-1 for pea to 0.75 kg ha-1 for canola. Net N and P mineralization from root varied little between crop species: 0.9-1.6 kg N ha-1 and 0.1-0.3 kg P ha-1. Nitrogen fertilization increased DM loss, and N and P release from straw residues.  相似文献

The current study was planned to develop an efficient vaccine against hydropericardium syndrome virus (HSV). Currently, formalin-inactivated liver organ vaccines failed to protect the Pakistan broiler industry from this destructive disease of economic importance. A field isolate of the pathogenic hydropericardium syndrome virus was adapted to chicken embryos after four blind passages. The chicken embryo-adapted virus was further serially passaged (12 times) to get complete attenuation. Groups of broiler chickens free from maternal antibodies against HSV at the age of 14 days were immunized either with 16th passage attenuated HSV vaccine or commercially formalized liver organ vaccine. The antibody response, measured by enzyme-linked immunosorbent assay was significantly higher (P < 0.05) in the group immunized with the 16th passage attenuated HSV vaccine compared to the group immunized with liver organ vaccine at 7, 14, and 21 days post-immunization. At 24 days of age, the broiler chickens in each group were challenged with 103.83 embryo infectious dose50 of pathogenic HSV and were observed for 7 days post-challenge. Vaccination with the 16th passage attenuated HSV gave 94.73% protection as validated on the basis of clinical signs (5.26%), gross lesions in the liver and heart (5.26%), histopathological lesions in the liver (1.5 ± 0.20), and mortality (5.26%). The birds inoculated with liver organ vaccine showed significantly low (p < 0.05; 55%) protection estimated on the basis of clinical signs (40%), gross lesions in the liver and heart (45%), histopathological lesions in the liver (2.7 ± 0.72), and mortality (35%). Birds in the unvaccinated control group showed high morbidity (84%), mortality (70%), gross (85%), and histopathological lesions (3.79 ± 0.14) with only 10% protection. In conclusion, this newly developed HSV vaccine proved to be immunogenic and has potential for controlling HSV infections in chickens.  相似文献

Genetic parameters for average daily gain from birth to weaning (ADGa), birth to 6 months (ADGb), weaning to 6 months (ADGc), weaning to yearling age (ADGd), and corresponding Kleiber ratios (KRa, KRb, KRc, and KRd) were estimated by using records of 3,533 Zandi lambs, descendent of 163 sires and 1265 dams, born between 1991 and 2005 at the Zandi Sheep Breeding Station at Khojir National Park, Tehran, Iran. A derivative-free algorithm combined with a series of six single-trait linear animal models was used to estimate phenotypic variance and its direct, maternal, and residual components. In addition, bivariate analyses were done to estimate (co)variance components between traits. Estimates of direct heritability (h 2 ) were 0.11, 0.15, 0.09, 0.10, 0.10, 0.10, 0.06, and 0.07 for ADGa, ADGb, ADGc, ADGd, KRa, KRb, KRc, and KRd, respectively, thereby indicating the presence of low additive genetic variation for growth rate and Kleiber ratio in this population of Zandi sheep. Maternal genetic component was found to be significant on ADGa and KRa and contributed 3% and 5%, respectively, in total phenotypic variance of ADGa and KRa. A widespread range of genetic correlations among traits studied was observed. Except for negative genetic correlations between ADGa and KRc, ADGa and KRd, and between KRa and KRc, in other cases, genetic correlations were positive and moderate to very high. Phenotypic correlations ranged from −0.49 (ADGa/KRd) to 0.94 (ADGc/KRc). These results indicate that selecting for improved growth rate or Kleiber ratio in Zandi sheep would generate a relatively slow genetic progress.  相似文献

Case 1: A tumor mass involving the rostral part of left cerebrum was found in a two-year-old female budgerigar (Melopsittacus undulatus) at necropsy. Histologically, the neoplastic cells were arranged in sheets or cords and occasionally showed nest growth patterns. These uniform tumor cells had a little cytoplasm and ovoid or round basophilic nuclei with clearly distinct cytoplasmic membranes. The tumor cells were strong diffusely immunostained with both neuron-specific enolase (NSE) and neurofilament protein and partially for synaptophysin. They lacked chromogranin A, glial fibrillary acidic protein (GFAP), vimentin, S-100, and cytokeratin antigen expression. Moreover, they had no reaction to antibodies against pituitary hormones, such as adrenocorticotrophic hormone (ACTH), growth hormone, and prolactin. The histological and immunohistochemical examination determined the tumor as neuroblastoma. Case 2: An extremely enlarged pituitary mass was found above the sella turcica of a male budgerigar. It was soft and well delineated from the adjacent structures. On histological examination, this tumor consisted of a sheet of large closely packed polyhedral cells that had scant to a large amount of pale to strongly eosinophilic cytoplasm. The pleomorphic nuclei were apparently variable in shape, from small round hyperchromatic to very large vesicular forms. The cell boundaries were not clearly distinct. The multifocal immunolabelling of neoplastic cells for NSE, synaptophysin, GFAP, and ACTH appeared, whereas a few cells reacted with vimentin and S-100 and stained negative for other markers, which were also utilized for case 1. Histological and immunohistochemical findings led to identification of corticotroph adenocarcinoma in the pituitary gland.  相似文献

A rust fungus was found causing stem cankers on 1‐ to 5‐year‐old stems of Salix elbursensis in the north west of Iran. The rust also forms uredinia on leaves and flowers of the host willow. Light and scanning electron microscopy revealed that the new rust is morphologically distinct from several Melampsora species occurring on the willows taxonomically close to S. elbursensis, but indistinguishable from Melampsora larici‐epitea. Examination of the internal transcribed spacer (ITS) region of the ribosomal DNA suggested that the rust fungus is phylogenetically close to Melampsora allii‐populina and Melampsora pruinosae on Populus spp. Based on both the morphological characteristics and the ITS sequence data, the rust is described as a new species –Melampsora iranica sp. nov.  相似文献
