An inoculation method for Exobasidium splendidum and Exobasidium vaccinii was developed on the dwarf shrub Vaccinium vitis-idaea. Using inoculated ramets, we investigated whether there are differences between V. vitis-idaea populations in the susceptibility to Exobasidium infections and whether the defence reaction of V. vitis-idaea is visible at a molecular level. Sixteen V. vitis-idaea clones from four populations were propagated in tissue cultures and the ramets were inoculated with E. splendidum or E. vaccinii fungi. The expression of three flavonoid biosynthetic genes (chalcone synthase, dihydroflavonol 4-reductase and anthocyanidin synthase) and the accumulation of flavonoids and hydroxycinnamic acids were determined in response to E. splendidum infection. A pathogenesis-related (PR 4) gene was isolated and its expression was studied in host ramet leaves. To our knowledge, this was the first successful artificial infection reported with E. splendidum. Disease frequencies of the inoculated ramets were between 32% and 47% for E. splendidum and 33% for E. vaccinii, but below 10% in uninoculated control ramets. There were no differences in disease frequencies between V. vitis-idaea populations. Both symptomatic leaves and healthy leaves of diseased ramets showed activation of flavonoid biosynthesis at the gene level, whereas expression of PR 4 was observed only in symptomatic leaves. The increase of flavonoid biosynthesis in healthy leaves of diseased ramets may represent a general response to stress or a role in defence against the pathogen E. splendidium. Ability of V. vitis-idaea to defend chemically against Exobasidium fungi and the heterogeneity of genotypes, age, size, and growth rates in host plant populations might be reasons for the low infection incidence of Exobasidia in nature. 相似文献

This study was designed to examine whether or not specific tree species (Picea glauca, Picea mariana, Pinus banksiana, Populus tremuloides), their post-fire stand age, or their position in a successional pathway had any significant effect on the functional diversity of associated soil microbial communities in a typical mixed boreal forest ecosystem (Duck Mountain Provincial Forest, Manitoba, Canada). Multivariate analyses designed to identify significant biotic and/or abiotic variables associated with patterns of organic substrate utilization (assessed using the BIOLOG™ System) revealed the overall similarity in substrate utilization by the soil microbial communities. The five clusters identified differed mainly by their substrate-utilization value rather than by specific substrate utilization. Variability in community functional diversity was not strongly associated to tree species or post-fire stand age; however, redundancy analysis indicated a stronger association between substrate utilization and successional pathway and soil pH. For example, microbial communities associated with the relatively high pH soils of the P. tremuloides-P. glauca successional pathway, exhibited a greater degree of substrate utilization than those associated with the P. banksiana-P. mariana successional pathway and more acidic soils. Differences in functional diversity specific to tree species were not observed and this may have reflected the mixed nature of the forest stands and of their heterogeneous forest floor. In a densely treed, mixed boreal forest ecosystem, great overlap in tree and understory species occur making it difficult to assign a definitive microbial community to any particular tree species. The presence of P. tremuloides in all stand types and post fire stand ages has probably contributed to the large amount of overlap in utilization profiles among soil samples. 相似文献

N-linked protein glycosylation is the most abundant posttranslation modification of secretory proteins in eukaryotes. A wide range of functions are attributed to glycan structures covalently linked to asparagine residues within the asparagine-X-serine/threonine consensus sequence (Asn-Xaa-Ser/Thr). We found an N-linked glycosylation system in the bacterium Campylobacter jejuni and demonstrate that a functional N-linked glycosylation pathway could be transferred into Escherichia coli. Although the bacterial N-glycan differs structurally from its eukaryotic counterparts, the cloning of a universal N-linked glycosylation cassette in E. coli opens up the possibility of engineering permutations of recombinant glycan structures for research and industrial applications. 相似文献

The green-revolution Rht-B1b and Rht-D1b dwarfing alleles are usually associated with increased wheat yields but are linked to reduced early growth and poor emergence if sowing conditions are unfavourable. Other dwarfing genes are available but not used in commercial breeding. The Rht13 bread wheat donor, Magnif M1, produces uniquely short peduncle and penultimate internodes to reduce plant height. A set of near-isogenic (NILs) and recombinant inbred (RILs) lines varying for height were developed from the cross of Magnif M1 and the Rht8c-containing Chuan-mai 18, and evaluated for a range of agronomic characteristics across favourable environments. Reductions in plant height were associated with increased grain number (r2 = 0.35**) and harvest index (r2 = 0.62**) in the NILs. Reduced-height RILs containing the Rht13-linked, Xgwm577M microsatellite marker were significantly shorter, produced greater biomass, yield and harvest index, and increased spike and grain number than lines without the marker. Approximately 74 and 7% of the total phenotypic variance in plant height was accounted for by allelic differences in Xgwm577 and Rht8 loci, respectively. The peduncle and penultimate peduncle internodes of Rht13-containing lines were proportionately shorter than Rht8c-containing sibs and lines containing the Rht-B1b dwarfing allele. The unique height-reducing phenotype, increased grain number and yield associated with Rht13 indicate considerable potential for use of this dwarfing allele for improving wheat performance. 相似文献

BACKGROUND: Positive secular trends in adolescent obesity and an increased prevalence of fear of fatness, particularly among girls, have been documented world-wide. There is a lack of consensus about assessment criteria for childhood obesity and no standard exists for assessing Irish children. In 1990, the Irish National Nutrition Survey used body mass index (BMI) > or =26 kg m(-2) to describe the prevalence of overweight among Irish adolescents. OBJECTIVES: (1) To examine the range in classification of Dublin schoolchildren as overweight according to four standard assessment methods; (2) to assess changes in weight status, prevalence of fear of fatness and accompanying slimming practices in a one-year follow-up; and (3) to compare the prevalence of overweight with that documented in 1990 among adolescents of similar age. DESIGN: A one-year follow-up study of 199 healthy schoolchildren (90 boys and 109 girls; mean age of 11 years at baseline) attending seven fee-paying (six single- and one mixed-sex) and eight non-fee-paying (four single- and four mixed-sex) primary schools in Dublin city centre. MEASUREMENTS: Weight, height, waist circumference and triceps skinfold were measured and used in five definitions of overweight, including published cut-off points of BMI-for-age (Centers for Disease Control and Prevention BMI-for-age charts for boys and girls; BMI reference curves for the UK 1990; International Obesity Task Force age- and sex-specific BMI cut-offs), actual relative weight and BM > or =26 kg m(-2). Assessment of body image perceptions and satisfaction (using figure line drawings) was reported in a questionnaire specifically designed for this study. RESULTS: The prevalence of overweight within the total group differed between the four standard definitions of weight status, by 9% at baseline and 8% at follow-up. Accordingly, increasing trends over the year ranged from zero to 3%. Using the criterion BMI > or =26 kg m(-2), 6% of Dublin schoolchildren were overweight, compared with 1.9% of schoolchildren in 1990. Significantly more girls than boys were affected by fear of fatness and were trying to lose weight. CONCLUSION: A standard method for assessment of weight status is urgently needed for the evaluation of obesity prevention initiatives among Irish schoolchildren. Ventricular bands are strand‐like intraventricular structures extending between adjacent papillary muscles; a papillary muscle and the ventricular wall or the interventricular septum and ventricular wall. Examination of the domestic dog right ventricle documented six ventricular band types. Eighty‐five of the 89 examined hearts had at least one right ventricular band. In the current study, the right ventricle of the 85 dog hearts was re‐examined for more than one ventricular band type. Seven patterns of multiple different bands were identified in the 24 dog hearts. The patterns (i.e., combinations) of different ventricular bands were grouped into three categories. Category 1 had five different patterns of bands. Each pattern had two different band types. Category 2 had one combination of three different bands. Category 3 had one combination of four different bands. The presence of right ventricular bands around the trabecula septomarginalis dextra was also documented. All 24 dog hearts with multiple ventricular bands had a trabecula septomarginalis dextra. The main goal of this study was documentation of multiple right ventricular band patterns. A secondary goal was documentation of the combined presence of these bands and a trabecula septomarginalis dextra. In the dog, the ventricular bands and trabecula are both thin, strand‐like intraventricular structures with variable branching patterns before blending into the ventricular wall. The gross similarity of these structures and lack of information on their combined presence could precipitate their misidentification. 相似文献

Physical properties of resistant starch (RS) were examined in a range of barley genotypes to determine the contribution of starch and seed physical characteristics to the RS component. Thirty‐three barley genotypes were studied, which varied significantly in their RS, amylose, and starch contents and grain yield. From 33 genotypes, 13 exhibiting high RS were selected for detailed physicochemical analysis of starch. In high‐RS varieties, granule size and number were unimodal, compared with normal starches from a reference genotype, which showed a bimodal distribution. Principal component analysis (PCA) showed that a higher content of granules <15 µm was positively correlated with RS and amylose content, whereas the proportion of granules 15–45 µm was negatively correlated with the RS and amylose contents. Physical fractionation of starches by centrifugation into different population sizes demonstrated that size alone is not an accurate indicator of the population of A‐type and B‐type granules within a given genotype. PCA also showed that large 15–45 µm granules were positively correlated with seed thickness and that thousand grain weight was positively correlated with seed width. High‐RS and high‐amylose genotypes showed variation in overall yield and starch content, with some genotypes showing yield comparable to the reference genotype. Analysis of amylopectin chain length distribution showed that high amylose or RS content was not associated with a higher proportion of amylopectin long chains when compared with either waxy or reference (normal) barley genotypes. This study highlights useful markers for screening barley genotypes with favorable starch characteristics. 相似文献
