The presence of illicit drugs and their metabolites in surface waters has to be considered a new type of hazard, still unknown, for the aquatic ecosystem, due to the potent pharmacological activities of all the illicit drugs. Our research was therefore aimed at evaluating the impact of illicit drugs on the aquatic fauna, till now still undervalued. To this aim, we verified the ability of the European eel (Anguilla anguilla), a well-known biomonitor of environmental contamination, to bioaccumulate cocaine, one of the most abundant illicit drugs found in surface waters. Silver eels were exposed to a nominal cocaine concentration of 20?ng/l for 1?month; at the same time, control, carrier, and post-exposure recovery groups were made. Brains, gills, liver, kidney, muscle, gonads, spleen, digestive tract, and sections of dorsal skin were assayed by high-pressure liquid chromatography. Cocaine was found in the tissues of the treated eels and, at low concentrations, in almost all tissues of post-exposure recovery eels. These results indicate that cocaine is able to accumulate into the eel tissues; its presence suggests potential risks for eels since cocaine could affect their physiology and contribute to their decline, and for humans consuming contaminated fish.  相似文献

The study of influenza type A (IA) infections in wild mammals populations is a critical gap in our knowledge of how IA viruses evolve in novel hosts that could be in close contact with avian reservoir species and other wild animals. The aim of this study was to evaluate the susceptibility to infection, the nasal shedding and the transmissibility of the H7N1 and H5N1 highly pathogenic avian influenza (HPAI) viruses in the bank vole (Myodes glareolus), a wild rodent common throughout Europe and Asia. Two out of 24 H5N1-infected voles displayed evident respiratory distress, while H7N1-infected voles remained asymptomatic. Viable virus was isolated from nasal washes collected from animals infected with both HPAI viruses, and extra-pulmonary infection was confirmed in both experimental groups. Histopathological lesions were evident in the respiratory tract of infected animals, although immunohistochemistry positivity was only detected in lungs and trachea of two H7N1-infected voles. Both HPAI viruses were transmitted by direct contact, and seroconversion was confirmed in 50% and 12.5% of the asymptomatic sentinels in the H7N1 and H5N1 groups, respectively. Interestingly, viable virus was isolated from lungs and nasal washes collected from contact sentinels of both groups. The present study demonstrated that two non-rodent adapted HPAI viruses caused asymptomatic infection in bank voles, which shed high amounts of the viruses and were able to infect contact voles. Further investigations are needed to determine whether bank voles could be involved as silent hosts in the transmission of HPAI viruses to other mammals and domestic poultry.  相似文献

The present study describes antimicrobial resistance patterns of Salmonella enterica subspecies enterica serovar Dublin (S. Dublin) in clinical submissions from calves and temporal and farm-type trends in antimicrobial resistance patterns of the isolates. A total of 300 isolates of S. Dublin were obtained from fecal or internal organs of calves fewer than 120 days of age originating from 84 dairies and 18 calf ranches from July 1998 to December 2002. The isolates were susceptibility tested to a panel of 10 antimicrobials using the disk diffusion assay. Temporal and farm-type trends in individual antimicrobial inhibition zone sizes were assessed and antimicrobial resistance patterns were described using cluster analysis. Isolates obtained from calf ranches compared with dairies exhibited decreased susceptibility to florfenicol, gentamicin, neomycin, sulfisoxazole, sulfamethoxazole/trimethoprim, and tetracycline. During the years 1998-2002, decreasing susceptibility was seen for ceftiofur, enrofloxacin, and sulfamethoxazole/trimethoprim. There were 20 different antimicrobial resistance patterns in the isolate set, indicating that S. Dublin has the ability to transfer and pick up resistance genes with relative ease. The trends seen in antimicrobial resistance in S. Dublin may likely be linked to antimicrobial drug use in young calves.  相似文献

A pioneering boat-based survey was conducted in 2019, to gather baseline information regarding the presence, composition, relative abundance, and spatial distribution of deep-diving and off-shore cetaceans in the northern South China Sea (SCS). A total of 27 sightings comprising at least 8 cetacean species were recorded during the 13-day survey, including 5 deep-diving species (i.e. Risso's dolphin [Grampus griseus], short-finned pilot whale [Globicephala macrorhynchus], sperm whale [Physeter macrocephalus], Cuvier's beaked whale [Ziphius cavirostris], and an unidentified beaked whale [either the ginkgo-toothed beaked whale, Mesoplodon ginkgodens, or Deraniyagala's beaked whale, Mesoplodon hotaula]), as well as 3 off-shore dolphins (i.e. pantropical spotted dolphin [Stenella attenuate], striped dolphin [Stenella coeruleoalba], and Fraser's dolphin [Lagenodelphis hosei]). With the exception of pantropical spotted dolphins, all other species were sighted and recorded at sea in the northern SCS for the first time. The pantropical spotted dolphin was the most frequently sighted species, comprising 30% of the total sightings. Deep-diving cetaceans were mainly sighted in the northern Xisha Archipelago, whereas off-shore dolphins were distributed across the survey area. The pantropical spotted dolphin was observed in aggregations of more than 100 individuals and nearly all encountered species included calves; these findings suggested that the survey area functions as an important feeding and calving ground for various cetacean species. This pioneering survey provides fundamental information regarding cetacean fauna in the northern SCS and highlights the need to strengthen research and conservation efforts concerning these species.  相似文献

Recently, significant progress has been achieved in improving the yield of good quality embryos in vitro. However, efforts are still required to recognize the factors and understand the mechanisms of oocyte maturation, which are essential for subsequent embryo development. The aims of the present study were to determine the frequency of apoptosis in oocytes recovered from slaughterhouse ovaries and to investigate whether insulin-like growth factor (IGF)-I action during oocyte maturation in vitro may withhold apoptosis and improve oocyte quality. Only oocytes of proper morphology with homogenous ooplasm and compact cumulus cells were selected for this study. All oocytes recovered from the slaughterhouse ovaries were divided into two groups. One group of oocytes, chosen for apoptosis detection, was examined immediately after recovery. The other group of oocytes was maturated in vitro. Oocytes were maturated with IGF-I supplementation (100 ng/ml). Oocytes without supplementation were used as a control. Apoptosis in oocytes was determined by positive results of TUNEL assay and active caspase labeling. The percentage of apoptotic oocytes detected by TUNEL fell to zero when the maturation medium was supplemented with IGF-I in comparison to the control matured oocytes (0 vs. 9.87%; P<0.05). However, active caspase labeling was only slightly decreased in the IGF-I matured oocytes compared with the control matured oocytes (1.13 vs. 2.08%; P<0.05). The results indicate that IGF-I may serve as an anti-apoptotic factor during oocyte maturation. We suggest that IGF-I may inhibit apoptosis in oocytes at the stage of caspase activation and may prevent further advancement of oocyte apoptosis.  相似文献

Spermatozoa are unique cells because of their morphological and physiological characteristics. They are produced during the process called spermatogenesis. Spermatogenesis consists of three phases: spermatocytogenesis, spermiogenesis and spermiation, during which spermatozoa undergo several changes. Spermatogenesis takes place within the seminiferous tubules containing two types of cells—the germ cells and the Sertoli cells—that alongside the Leydig cells, which play an important role when it comes to normal fertility. Everything is regulated by the hypothalamic–pituitary–gonadal axis and specific hormones due to multi-hormonal feedback systems. Spermatozoa possess morphological and physiological features, which are sometimes completely different from what is observed in various somatic cells. What is more, canine spermatozoa have specific characteristics making them special compared to the spermatozoa of other mammalian species. The metabolic energy production, which is crucial for the appropriate functioning of spermatozoa, can be fuelled by different metabolic pathways utilizing different chemical substrates. Inseparable from the oxidative phosphorylation process is the production of reactive oxygen species, which are both essential and toxic to spermatozoa. Furthermore, epididymis is a very important structure, responsible for the transport and maturation of spermatozoa, which are then stored in the last segment of epididymis—the epididymal cauda. Moreover, the retrieval of spermatozoa from the epididymides is crucial for the development of assisted reproduction techniques and sperm cryopreservation methods. The information gained from the research on domestic dogs might be transferred to their wild relatives, especially those species categorized as endangered.  相似文献

The activity of glycosidases is crucial for the function and biological activity of proteins conjugated with sugar moieties, which play an important role in adhesion of cells during attachment and detachment of the foetal membranes. The aim of study was to describe the ability of bovine placental tissues to break down O-glycosidic bonds in different glycoproteins by the determination of activity of β-galactosidase, α-l-fucosidase, β-N-acetyl-hexosaminidase and sialidase in early–mid-pregnancy as well as at parturition with released and retained foetal membranes. Moreover, the availability of substrates for these glycosidases in placental homogenates was evaluated. Placental samples were collected from pregnant (2–4 months) cows in slaughterhouse (n = 8) as well as during Caesarean section and divided into released foetal membranes (n = 8) and retained foetal membranes (n = 8). Tissue homogenates were subjected to spectrofluorimetric and spectrophotometric determinations of enzyme activities as well as electrophoretic separations. Enzyme activities expressed changes within examined time with significant (p < .05) differences between pregnancy and physiological parturition in β-N-acetyl-hexosaminidase and α-l-fucosidase in foetal part of placenta while in maternal part only in the latter one. Decreasing tendency in enzyme activity was noticed in foetal part of retained samples in comparison with released ones with significant (p < .05) differences in α-l-fucosidase activity. The analysis of staining of sugar moieties attached to selected proteins depicted availability of sugar molecules in examined tissues, but their patterns differed between samples. In conclusion, sugar moieties in conjugated proteins express changes in the course of pregnancy which is reflected by the alterations in activities of placental glycosidases.  相似文献

A 5-minute contact of nursing mothers of the Levant vole,Microtus guentheri, with paper impregnated with a DES suspension in soybean oil (125 μg/cm2 ) caused sterilization of 60% of the newborn female offspring. A 1-hour contact was sufficient to sterilize more than 90% of them. When the DES concentration was decreased to 1 μg/cm2 only 44% of the newborn female offspring were sterile although their nursing mothers had been exposed to contact with the treated paper for a period of 2 hours. The concentration of 0.1 μg/cm2 was totally ineffective in causing sterilization. Treatment of nesting material with DES offers a promising means of reproductive suppression in Levant voles.  相似文献
