Maize is categorized as a salt-sensitive crop and identification of fairly salt-tolerant lines is of paramount importance for increasing its production on saline soils. Experiments were conducted in randomized block design with three replications to identify maize accessions showing response to saline water irrigation, traits imparting tolerance, and their effect on yield attributes of maize. Significant variation was present among genotypes for specific leaf area (SLA), potassium (K) content, cob characteristics, yield, biomass, and harvest index. High amount of heritability with large genetic advance indicated the presence of additive gene action for traits like leaf water potential and leaf dry weight. Association analysis revealed high correlation between key traits and direct as well as positive effect of these traits on yield. Principal component analysis resolved three principal components, and high leaf area and water potential were conferring salt tolerance and thus higher yield.  相似文献

Heterogeneous amniotic fluid contains various cell types. The aim of this study was to characterize and differentiate some of the key stemness attributes of the amniotic fluid-derived cells in buffalo (Bubalus bubalis). The amniotic fluid (AF) cells were cultured without feeder cells, in DMEM containing 15% FBS, 1% non-essential amino acids, 1% penicillin/streptomycin/ampicillin, 1% vitamin solution, and 1% l-glutamine in 5% CO(2) in humidified air at 38.5±0.5 °C. After 6 days of culture different types of cells viz., star shaped (62.7%), spherical without nucleus (1.9%), spherical with nucleus (26.4%), pentagonal (0.4%), and free floating/rounded cells (8.3%) were observed. Most of the cells started anchorage-dependent growth after day 7 of the culture. Expression of alkaline phosphatase (AP) and Oct-4, Nestin and FGF-5 were observed from the AF cells at different passages. Using species-specific primers, a PCR amplicon of 200, 296 and 210 bp were observed for Oct-4, Nestin and FGF-5, respectively. The cells were found to have a normal karyotype at different passages. These results may contribute towards establishing non-embryonic pluripotent stem cells for various therapeutic and reproductive biotechnological applications in the species.  相似文献

Thirty-two water buffalo (Bubalus bubalis) calves aged 6?C10?months were used to evaluate serological responses to Brucella abortus strain RB51 (RB51) vaccination in a dose?Cresponse study and to compare the use of two selective media for the isolation of RB51. The animals were randomly divided into three treatment groups. Groups I?CIII received the recommended vaccine dose (RD) twice 4?weeks apart, RD twice 18?weeks apart and saline once, respectively. Lymph nodes were excised from the three groups and subjected to bacteriological examination to determine the frequency of detection of RB51. Pre- and post-vaccination blood samples were collected and tested for B. abortus antibodies using the buffered plate agglutination test (BPAT), complement fixation test (CFT), and dot-blot assay. Sera taken at all post-inoculation weeks (PIW) were negative for field strain B. abortus using the BPAT. Antibody responses to RB51 were demonstrated in all vaccinates but not in controls by CFT and dot-blot assay from 1 PIW up to 16?weeks following booster vaccination. The agreement for both assays was 80.7% and there was a linear interdependence with a Pearson??s correlation coefficient value of 0.578. The frequency of isolation of RB51 from the two selective media used was not significantly different (P?>?0.05).  相似文献

The segmentation of symptoms during image analysis of diseased plant leaves is an essential process for detection and classification of diseases. However, there are challenges involved in the task, many of them related to the variability of image and host/symptom characteristics and conditions. As a result of those challenges, the methods proposed in the literature so far focus on a specific problem and are usually bounded by tight constraints regarding image capture conditions. This research explores a new automatic method for segmenting disease symptoms on plant leaves that was designed to be applicable in a wide range of situations. The proposed technique employs only color channel manipulations and Boolean operations applied on binary masks, thus being simpler and more robust compared to many previously described automatic methods. Its effectiveness is demonstrated by tests performed over a large database containing images of 77 different diseases of 11 plant species. A comparison with manual segmentation is also presented, further reinforcing the advantages of the proposed approach.  相似文献

A field experimentation was conducted during 2009-2011 at CSK Himachal Pradesh Agricultural University, Palampur, India characterized with wet-temperate climate and acid Alfisol soil having medium available phosphorus content. The study aimed at bio-fortification and quality enhancement of okra (Abelmoschus esculentus)–pea (Pisum sativum) cropping system through arbuscular mycorrhizal fungi (AMF) (Glomus mosseae) at varying inorganic phosphorus (50, 75, and 100% soil-test-based recommended P dose) and irrigation regimes (40 and 80% available water capacity) in a Himalayan acid Alfisol. The results revealed that AMF and inorganic P significantly enhanced the concentrations and uptake of various primary [nitrogen (N), phosphorus (P), and potassium (K)]; secondary [calcium (Ca)]; and micronutrients [iron (Fe), zinc (Zn), copper (Cu), manganese (Mn), boron (B), and molybdenum (Mo)] in okra and pea crops. However, effects of varying irrigation regimes were found to be nominal. In okra, AMF inoculation considerably enhanced N, P, K, B, and Mo uptake by 5, 19, 3, 4, and 15%, respectively, over their non-AMF counterparts. Likewise in pea, a higher amount of N (10%), P (26%), K (7%), Fe (7%), Cu (38%), Zn (20%), Mn (4%), B (7%), and Mo (13%) uptake was registered through AMF inoculation over their non-AMF counterparts. Application of soil-test-based P dose from 50% to 100% P also resulted in significant and consistent improvement in N, P, K, B, and Mo uptake both in okra and pea and in Zn, Cu, Mn, and Fe uptake in pea crop. Magnitude of increase in Ca content was to the tune of 13 and 4%, respectively, in okra fruits and pea pods following AMF inoculation, whereas crude protein content enhanced by 4% each in both the crops. Overall, the current study demonstrates the important role of AMF in nutrient enrichment and quality enhancement of okra and pea crops in acid Alfisol, besides considerable reduction in investment on chemical fertilizers. Results of current study suggest that AMF use in Himalayan production systems is of tremendous significance to harvest nutritionally-rich farm produce for Himalayan communities suffering from malnutrition especially anemia and Zn deficiency, and equally to resource-poor Himalayan farmers who ill-afford expensive external inputs.  相似文献

A set of 50 rice genotypes comprising landraces, local selections, and improved varieties were characterized using simple sequence repeat(SSR) and inter simple sequence repeat(ISSR) markers to study genetic diversity and population structure. Following unweighted pair group method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 5 clusters and 11 sub-clusters, whereas population structure analysis separated 50 rice genotypes into 5 sub-populations. Grouping of rice genotypes showed better resemblance with the pedigree information of the genotypes. Both genetic diversity and population structure analysis separated majority of the improved varieties from landraces and local selections. Some of the SSR markers amplified unique alleles which were specific to a particular genotype and could distinguish them from the rest. The results indicate that these rice genotypes exhibit a higher genetic diversity and can be very useful in rice improvement program.  相似文献

A study was conducted to investigate the effects of paclobutrazol (PBZ) on ion leakage (IL), proline content and activities of antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) of 1-year-old ‘Olour’ mango plants subjected to NaCl stress. Plants were treated with two levels of salt, i.e., 0.0 g NaCl (control) and 25 g NaCl/25 kg soil and three levels of paclobutrazol (PBZ) solution (0.0 (control), 750 and 1500 mg/l). Ion leakage, proline content and activities of antioxidant enzymes were significantly altered by both salinity and PBZ treatments. Results indicated that PBZ (1500 mg/l) mitigated the salinity stress and reduced ion leakage of mango seedlings by 64% over non-PBZ-treated salinised plants. PBZ upregulated the endogenous proline content and salinised plants treated with PBZ (1500 mg/l) had 17% higher proline content than salinised plants without PBZ treatment. Higher antioxidant enzyme activity was also observed in salinised plants treated by PBZ than salinised plants without PBZ treatments. Moreover, higher dose of PBZ (1500 mg/l) resulted in higher activity of these enzymes in mango leaves. In comparison to salinised plants without PBZ treatment, salinised plants treated with PBZ (1500 mg/l) had higher SOD (24%), CAT (46%) and POD (163%) activities. Our results suggest that PBZ application under salt stress conditions alters the equilibrium between free radical production and enzymatic defense reactions in mango by enhancing the proline content and free radical scavenging capacity.  相似文献
