The response of the ocean redox state to the rise of atmospheric oxygen about 2.3 billion years ago (Ga) is a matter of controversy. Here we provide iron isotope evidence that the change in the ocean iron cycle occurred at the same time as the change in the atmospheric redox state. Variable and negative iron isotope values in pyrites older than about 2.3 Ga suggest that an iron-rich global ocean was strongly affected by the deposition of iron oxides. Between 2.3 and 1.8 Ga, positive iron isotope values of pyrite likely reflect an increase in the precipitation of iron sulfides relative to iron oxides in a redox stratified ocean.  相似文献

The Eastern oyster (Crassostrea virginica) is a useful, robust model marine organism for tissue metabolism studies. Its relatively few organs are easily delineated and there is sufficient understanding of their functions based on classical assays to support interpretation of advanced spectroscopic approaches. Here we apply high-resolution proton nuclear magnetic resonance (1H NMR)-based metabolomic analysis to C. virginica to investigate the differences in the metabolic profile of different organ groups, and magnetic resonance imaging (MRI) to non-invasively identify the well separated organs. Metabolites were identified in perchloric acid extracts of three portions of the oyster containing: (1) adductor muscle, (2) stomach and digestive gland, and (3) mantle and gills. Osmolytes dominated the metabolome in all three organ blocks with decreasing concentration as follows: betaine > taurine > proline > glycine > ß-alanine > hypotaurine. Mitochondrial metabolism appeared most pronounced in the adductor muscle with elevated levels of carnitine facilitating ß-oxidation, and ATP, and phosphoarginine synthesis, while glycogen was elevated in the mantle/gills and stomach/digestive gland. A biochemical schematic is presented that relates metabolites to biochemical pathways correlated with physiological organ functions. This study identifies metabolites and corresponding 1H NMR peak assignments for future NMR-based metabolomic studies in oysters.  相似文献

Purpose

Detonation synthesis nanodiamonds (ND) are among the most widely applied nanoparticles due to their low cost of production and broad scope of applications. However, the fate and behavior of NDs in the environment are largely unknown. The behavior of NDs is greatly affected by humic substances (HSs), which comprise 50 to 80 % of natural organic matter in water and soil ecosystems. The uptake of detonation NDs by wheat seedlings and its toxicity were evaluated in the presence of seven HSs of different origins, including humic acids (HA, HS fraction soluble in alkali and insoluble in acid) and fulvic acids (FA, soluble in both alkali and acid).

Materials and methods

To monitor the uptake of NDs by plants, tritium-labeled NDs were produced. Liquid scintillation spectrometry and autoradiography were used to determine the amount of NDs absorbed by plants. The photosynthetic activity of the plants was measured using light response curves.

Results and discussion

After a 24-h exposure period, the ND content in the plant roots was 1720 μg g?1. The introduction of HSs decreased the ND contents in the plant roots to 680–1570 μg g?1 (except for peat FA, for which the ND content did not differ from the blank value). The observed phenomenon was probably related mainly to the influence of HSs on the zeta potential of the NDs, which shifted from positive to negative. Based on chlorophyll fluorescence evaluation, the toxicity of NDs did not inhibit photosynthesis during illumination in the physiological range. However, NDs were slightly toxic to wheat plants under excessive light, likely due to the inhibition of electron transport between Q A and Q B and the disruption of the formation of a thylakoid transmembrane potential.

Conclusions

The introduction of HA in a suspension of NDs obviously reduced the inhibiting effect of the NDs; however, the mitigating activities of FA were not so apparent. Our results demonstrate the urgent need for further studies of the influences of NDs on plant growth and development.

This study aimed to assess the sex differences in the feeding behaviour of non-obese diabetic severe combined immunodeficient (NOD SCID) mice in a pharmacological model of type 1 diabetes mellitus (T1Dm). In our study, we chose NOD SCID mice of both sexes and assessed their feeding behaviour, body weight, body fat and water content under identical experimental conditions and diets. After 1 month of diabetes mellitus in mice in the experimental group, males and females did not show any increase in body weight, and they weighed significantly less than the control group. However, compared with the control group, in females with a background of T1Dm, there was a significant decrease in body fat. The amount of water consumed in the experimental groups was higher than that in the control groups. The amount of food consumed by males increased when they increased their water consumption, whereas food consumption in females decreased significantly with an increase in water consumption. Thus, we discovered sex differences in the feeding behaviour, body weight and body fat and water content in the pharmacological model of T1Dm after 1 month in NOD SCID mice.  相似文献

The area of contact between a T cell and an antigen-presenting cell (APC) is known as the immunological synapse. Although its exact function is unknown, one model suggests that it allows for T cell receptor (TCR) clustering and for sustained signaling in T cells for many hours. Here we demonstrate that TCR-mediated tyrosine kinase signaling in na?ve T cells occurred primarily at the periphery of the synapse and was largely abated before mature immunological synapses had formed. These data suggest that many hours of TCR signaling are not required for T cell activation. These observations challenge current ideas about the role of immunological synapses in T cell activation.  相似文献
