Polybrominated diphenyl ethers (PBDEs) are a class of global flame retardants whose residues have markedly increased in fish and human tissues during the last decade. They belong to persistent and toxic contaminants which need more attention and toxicological study as their degradation in the environment is not well understood. This study characterizes 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) bioconcentration, elimination and biotransformation in juvenile turbot Psetta maxima, in order to evaluate the risk of its bioaccumulation in the marine environment. During this study, juvenile turbot were exposed to environmentally relevant BDE-47 concentrations from 0.001 to 1 μg/L for 16 days via aqueous exposure. This study found that juvenile turbot bioconcentrated and biotransformed BDE-47. There was no difference in standard length or weight between control and exposed fish. Uptake and elimination of BDE-47 by the turbot during exposure were examined in controlled laboratory experiments. Bioconcentration of BDE-47 was similar in fish from all treatment groups. Bioconcentration was rapid and increased with exposure time, since the bioconcentration factor (BCF) was higher. However, elimination was slower in comparison and low elimination was detected after 10 days in clean water. BDE-47 concentration in water influences the BCF. Half-life ranged between 37 and 108 days and theoretical times t90 could range from 120 to 358 days. The present study demonstrates a stepwise debromination of BDE-47 to BDE-28.
A rapid and sensitive automated method for glucose monitoring that might be employed during wine fermentation and processing was developed. A flow injection (FI) system coupled with an automated dilutor and the "redox-versatile" modified electrode were used to directly measure glucose in wine. To avoid interferences during wine analysis, different formulations of enzymatically modified carbon paste electrodes (CPE) were used and evaluated in oxidation and reduction mode. The best selectivity and sensitivity for glucose monitoring in real samples was obtained in cathodic mode at a fixed potential of 0 V versus Ag/AgCl using a CPE modified with glucose oxidase, horseradish peroxidase, and ferrocene as redox mediator. A total linear range of 0.02-50 g/L glucose was covered using this automated system and allowed the measurement of glucose in dry, medium, and sweet white or red wines without any sample pretreatment. The results showed a good correlation with the standard method, and the proposed method is very rapid, simple, and reliable and does not need skilled operators. 相似文献
The use of treated industrial effluents to irrigate plants is an alternative, because of nutrients that can increase yield of the agricultural crops. This study was conducted to determine irrigation with treated effluent and gypsum application, which changes the chemical and physical characteristics of soils and the growth and nutrition of millet (Pennisetum glaucum). Thus, an experiment was conducted on PVC columns with three soil classes, Typic Hapludox, Typic Hapludult, and Arenic Hapludult. Nutrient and Na+ concentrations in the millet biomass reflected concentrations of elements in the effluent and soil. In the control, low N levels were found in the biomass, while higher leaf N concentrations were observed, due to irrigation with treated effluent. In the short term, irrigation with treated industrial effluent by controlled application could be an alternative and a complementary source of nutrients for plants, reducing the volume of nutrients and organic materials discharged into water bodies. 相似文献
Astaxanthin (ASTA) is a fat-soluble xanthophyll with powerful antioxidant functions. It is extracted from e.g. salmon, an important food source for certain human populations known to have a reduced risk of tumor development. It is possible that ASTA plays a role in cancer chemoprevention in such populations. The purpose of this study was to investigate the effects of dietary ASTA on chemically induced mammary tumorigenesis using N-methyl-N-nitroso-urea (MNU) in immature Wistar rats.
Methods
Thirty-six 37 days old juvenile female Wistar rats were at random allocated to 4 groups of which Groups 1 and 2 received a single dose of 55 mg MNU/kg body weight. The effects of ASTA was evaluated by giving rats of Groups 2 and 4 a dose of 50 mg ASTA/kg/day for the entire duration of the study. Group 3 rats received feed added alimentary oil.Necropsy and histopathological examinations were carried out on each rat 14 months after the administration of MNU. Haematological values and antioxidative status were determined. Oxidative stress was evaluated by monitoring superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities in hepatic tissue. Lipid peroxidation and carbonylation of proteins was determined in protein extracts from the liver.
Results
Tumor development occurred only in rats of Groups 1 and 2, i.e. MNU exposed animals. Frequency of tumor development in general and average number of tumors per animal were insignificant between these two groups. Mammary gland tumors developed in equal frequencies in Group 1 and 2 rats, respectively. Although only rather few tumors were found in the mammary glands, a substantial number of other tumors were found in Group 1 and 2 rats, but at equal rates.Biochemical analyses showed significant higher levels of GPx, malondialdehyde and dinitrophenylhydrazine in Group 1 rats that for rats in all other groups thus indicating protective effects of ASTA on MNU induced hepatic oxidative stress.
Conclusions
Supplementation with ASTA did not reduce tumorigenesis induced by MNU in Wistar rats. However, supplementation with ASTA seemed to have anti-inflammatory effects. 相似文献
The objective of the present work was to develop species-specific microsatellite markers for P. scalare and to analyze the diversity and genetic structure of a wild population, from the Amazon River, and three commercial stocks (common, marble, and clown morphological variants), from farmers in Vieras-Minas Gerais. Through microsatellite-enriched genetic libraries, 11 microsatellite markers with adequate amplification patterns were characterized. Population genetic analysis identified eight polymorphic loci that generated 66 alleles ranging from two alleles (PSCA1B3) to nine (PSCA2H1). The polymorphic information content ranged from 0.031 to 0.827. High genetic differentiation was observed between the wild population and the stocks, and moderate differentiation between the three stocks. Deviation in the Hardy-Weinberg equilibrium was observed in one locus in the wild population, in five loci in the common morphological variant, in two in the marble, and in two in the clown morphological variant. Bayesian analysis of genetic structure revealed the existence of two clusters, one represented by the natural population and the other by the stocks. The developed microsatellite markers serve as a tool for the analysis of diversity and genetic structure and conservation studies of P. scalare.
Microalgal biotechnology shows considerable promise as a sustainable contributor to a broad range of industrial avenues. The field is however limited by processing methods that have commonly hindered the progress of high throughput screening, and consequently development of improved microalgal strains. We tested various microplate reader and flow cytometer methods for monitoring the commercially relevant pigment fucoxanthin in the marine diatom Phaeodactylum tricornutum. Based on accuracy and flexibility, we chose one described previously to adapt to live culture samples using a microplate reader and achieved a high correlation to HPLC (R2 = 0.849), effectively removing the need for solvent extraction. This was achieved by using new absorbance spectra inputs, reducing the detectable pigment library and changing pathlength values for the spectral deconvolution method in microplate reader format. Adaptation to 384-well microplates and removal of the need to equalize cultures by density further increased the screening rate. This work is of primary interest to projects requiring detection of biological pigments, and could theoretically be extended to other organisms and pigments of interest, improving the viability of microalgae biotechnology as a contributor to sustainable industry. 相似文献
