The Far Eastern Leopard (Panthera pardus orientalis; Schlegel, 1857) is perhaps the world’s most endangered large felid subspecies occurring in a single population of ?30 adults, and faces immediate risk of extinction unless additional populations can be established within its historical range in the Russian Far East. We used locations of leopard tracks (and their ungulate prey) collected from snow track surveys from 1997 to 2007 to develop resource selection functions (RSF) to identify potential habitat for reintroduction. We compared models that include prey versus those based on landscape covariates, and also included covariates related to human-induced mortality. To estimate potential population size, we used a habitat-based population estimate based on the ratio of population size and RSF value of occupied range. Far Eastern leopards selected for areas with high ungulate density, lower-elevation Korean pine forests on southwest facing slopes, and in areas far from human activity. Using this RSF model, we identified a total of 10,648 km2 in eight patches >500 km2 of potential Far Eastern leopard habitat that could harbor a potential population of 105.3 (57.9–147.2) adults. In combination with the existing population, successful reintroductions could result in a total of 139.2 (76.5–194.6) adult leopards, a 3–4-fold increase in population size. Our habitat models assist the reintroduction planning process by identifying factors that predict presence and potential suitable habitat. Identifying the highest quality, most connected patches, in combination with appropriate selection and training of released animals, is recommended for successfully reintroducing Far Eastern leopards, and potentially other endangered carnivores into the wild.  相似文献

OBJECTIVES: To assess the feasibility of optical coherence tomography (OCT) for examining the cat ocular fundus, to provide normative data on retinal thickness in different fundus regions, and to demonstrate selected surgically induced vitreoretinal pathologies in the cat. ANIMAL STUDIED: Forty-five eyes of 28 healthy domestic cats and two eyes of domestic cats that had undergone subretinal implantation surgery for a visual prosthesis were examined. PROCEDURES: An optical coherence tomograph (Zeiss-Humphrey) was used to examine the anesthetized animals. At least five vertical and five horizontal scans in regular distribution were recorded for each cat including (1) the peripapillary region, (2) the area centralis, and (3) the peripheral retina. Thickness was measured manually at five locations in each scan. Retinal thickness was compared in the three above-mentioned fundus regions, between eyes and between vertical and horizontal scans. OCT was additionally performed in animals with retinal detachment and a subretinal visual prosthesis. RESULTS: OCT measurements required only minimal adjustments of human settings and yielded high quality images. In comparison to humans intraretinal layers were more difficult to differentiate. Retinal thickness was highest in the peripapillary region (245 +/- 21 microm), followed by the peripheral retina (204 +/- 11 microm) and the area centralis (182 +/- 11 microm; all P < 0.0001). There was no statistically significant difference between right and left eye or between vertical and horizontal scans. OCT demonstrated retinal detachment, an iatrogenic break and a subretinal prosthetic device in high detail. CONCLUSIONS: Retinal thickness was measurable with high precision; values compare well to older histologic studies. OCT bears significant advantages over histology in enabling one to repeat measurements in living animals and thus allowing longitudinal studies. Various vitreoretinal pathologies common in feline eyes are detectable and quantifiable by OCT.  相似文献

OBJECTIVE: To compare cardiac output (CO) measured by use of the partial carbon dioxide rebreathing method (NICO) or lithium dilution method (LiDCO) in anesthetized foals. SAMPLE POPULATION: Data reported in 2 other studies for 18 neonatal foals that weighed 32 to 61 kg. PROCEDURES: Foals were anesthetized and instrumented to measure direct blood pressure, heart rate, arterial blood gases, end-tidal isoflurane and carbon dioxide concentrations, and CO. Various COs were achieved by administration of dobutamine, norepinephrine, vasopressin, phenylephrine, and isoflurane to allow comparisons between LiDCO and NICO methods. Measurements were obtained in duplicate or triplicate. We allowed 2 minutes between measurements for LiDCO and 3 minutes for NICO after achieving a stable hemodynamic plane for at least 10 to 15 minutes at each CO. RESULTS: 217 comparisons were made. Correlation (r = 0.77) was good between the 2 methods for all determinations. Mean +/- SD measurements of cardiac index for all comparisons with the LiDCO and NICO methods were 138 +/- 62 mL/kg/min (range, 40 to 381 mL/kg/min) and 154 +/- 55 mL/kg/min (range, 54 to 358 mL/kg/min), respectively. Mean difference (bias) between LiDCO and NICO measurements was -17.3 mL/kg/min with a precision (1.96 x SD) of 114 mL/kg/min (range, -131.3 to 96.7). Mean of the differences of LiDCO and NICO measurements was 4.37 + (0.87 x NICO value). CONCLUSIONS AND CLINICAL RELEVANCE: The NICO method is a viable, noninvasive method for determination of CO in neonatal foals with normal respiratory function. It compares well with the more invasive LiDCO method.  相似文献

DNA photolyases use light energy to repair DNA that comprises ultraviolet-induced lesions such as the cis-syn cyclobutane pyrimidine dimers (CPDs). Here we report the crystal structure of a DNA photolyase bound to duplex DNA that is bent by 50 degrees and comprises a synthetic CPD lesion. This CPD lesion is flipped into the active site and split there into two thymines by synchrotron radiation at 100 K. Although photolyases catalyze blue light-driven CPD cleavage only above 200 K, this structure apparently mimics a structural substate during light-driven DNA repair in which back-flipping of the thymines into duplex DNA has not yet taken place.  相似文献

The HIV envelope (Env) protein gp120 is protected from antibody recognition by a dense glycan shield. However, several of the recently identified PGT broadly neutralizing antibodies appear to interact directly with the HIV glycan coat. Crystal structures of antigen-binding fragments (Fabs) PGT 127 and 128 with Man(9) at 1.65 and 1.29 angstrom resolution, respectively, and glycan binding data delineate a specific high mannose-binding site. Fab PGT 128 complexed with a fully glycosylated gp120 outer domain at 3.25 angstroms reveals that the antibody penetrates the glycan shield and recognizes two conserved glycans as well as a short β-strand segment of the gp120 V3 loop, accounting for its high binding affinity and broad specificity. Furthermore, our data suggest that the high neutralization potency of PGT 127 and 128 immunoglobulin Gs may be mediated by cross-linking Env trimers on the viral surface.  相似文献

Molecular noise restricts the ability of an individual cell to resolve input signals of different strengths and gather information about the external environment. Transmitting information through complex signaling networks with redundancies can overcome this limitation. We developed an integrative theoretical and experimental framework, based on the formalism of information theory, to quantitatively predict and measure the amount of information transduced by molecular and cellular networks. Analyzing tumor necrosis factor (TNF) signaling revealed that individual TNF signaling pathways transduce information sufficient for accurate binary decisions, and an upstream bottleneck limits the information gained via multiple integrated pathways. Negative feedback to this bottleneck could both alleviate and enhance its limiting effect, despite decreasing noise. Bottlenecks likewise constrain information attained by networks signaling through multiple genes or cells.  相似文献

Background

In many species of birds, pair bonded males and females precisely co-ordinate their vocalisations to form duets. Duetting behaviour, although still somewhat of an enigma, is thought to function primarily in territorial defence and mate guarding. We identify an additional function of duetting in an afrotropical bird, the tropical boubou (Laniarius aethiopicus), that uses one duet type as a postconflict display probably to advertise victory to other boubous.

Results

We simulated intrusions into boubou territories in the field in Ivory Coast, West Africa using playbacks of four different types of boubou duets to test the use of the presumptive acoustic victory display before, during and after playbacks. These staged encounters resulted in either retreat of the focal birds during playback or continued presence accompanied by vocal displays after playback had ceased. Losers of encounters never sung after retreating whereas 11 out of 18 pairs sung the presumptive victory duet after the encounter. Analysis revealed that the presumptive victory display was sung significantly more often after than before or during the playback treatment.

Conclusion

We conclude that, most likely, the investigated duet type is a postconflict victory display – a novel function of duets. Furthermore the duet is a rare example among birds of a context-specific song. The conspicuousness of the display suggests that it is directed not only to losers of an agonistic encounter but also to other pairs of birds in neighbouring territories.  相似文献

Abstract

The ability of 7 day old wheat seedlings to take up nitrate or ammonium from hydroponic solution was measured. Seedlings were grown under fully aerated hydroponic conditions. The growth solution consisted of either 0.5 mM CaSO4 alone or in combination with high nitrate (5 mM NO3 ), high ammonium (2 mM NH4 +) or modified 1/10 Hoaglands solution with nitrate N only (14 mM) or ammonium N only (2 mM). After washing the roots for one hour in CaSO4, nitrate or ammonium uptake was measured with an ion selective electrode. Plants grown in high nitrate were unable to take up nitrate from a 0.1 mM external solution. Those grown in CaSO4 were able to take up nitrate at the same external concentration (flux = 10.2 +/‐ 3.0 μmol nitrate/g dry wtlbh). The same result was seen for plants grown in high ammonium vs those grown in CaSO4 (flux = 21.0 +/‐ 10.0 μmol/g dry wtlbh). Similar results were obtained when modified Hoagland's solution was substituted for the high N solutions. These data indicate that wheat roots possess both high and low affinity nitrate and ammonium uptake systems. The data further indicate that, for a given ion, the high and low affinity systems do not operate simultaneously under high N conditions. The high affinity system is switched off in the range of 1 mM for both ionic forms of N. Developmental studies show that the expression of the high affinity trait is reversible and may be induced (repressed) by conditioning for 24 h in low (high) N media. Plants grown in high N solutions showed efflux of the ion under assay conditions. Neither ion interfered with the induction/repression of the high affinity trait for the other under the conditions used in this study.  相似文献

Objective To determine if systemic administration of lidocaine during intraocular surgery reduces post-operative ocular pain. Study design Randomized, masked, controlled experimental trial. Animals Twelve dogs weighing 15.5 ± 1.7 kg (mean ± SD) and aged 2.5 ± 0.6 years. Methods All dogs underwent a baseline ophthalmic examination and subjective pain score. Anesthesia consisted of acepromazine (0.1 mg kg−1, IM), propofol (4–6 mg kg−1, IV), and isoflurane in oxygen. There were three groups each receiving a bolus followed by an infusion (n = 4): saline (0.3 mL kg−1 IV + 0.2 mL kg−1hour−1 IV); morphine (0.15 mg kg−1 IV + 0.1 mg kg−1hour−1 IV); and lidocaine (1.0 mg kg−1 IV + 0.025 mg kg−1minute−1 IV). All treatments began 15 minutes prior to starting of phacoemulsification and lens removal from the right eye. Pain scores were recorded at 0.5, 1, 2, 3, 4, 6, 8, 16, and 24 hours after t = 0 (extubation). Rescue morphine was administered (1.0 mg kg−1 IM) if the subjective pain score ≥9 (maximum = 24), and the dog was excluded from further data analysis. Differences in pain scores and time-to-treatment failure (TTF) were analyzed using the Wilcoxon's rank sum test. Differences in incidence of treatment failure were analyzed using Fisher's exact test. Physiologic data were analyzed using repeated measures anova . Significance was defined as P < 0.05. Results Incidence of treatment failure was 100% in saline-treated dogs and 50% in morphine- or lidocaine-treated dogs. There was no difference in intraocular pressure, aqueous flare, cell count (or protein) between groups in the operated eye at any time following extubation. Conclusion and clinical relevance This pilot study suggests that intraoperative lidocaine may provide analgesic benefits similar to morphine for intraocular surgery in dogs, but more definitive research is needed. This model appears to be appropriate for pain assessment studies as the negative control group demonstrated 100% failure rate.  相似文献
