Hand dissection of mature grains from two common wheats (Triticum aestivum L., cv. Caphorn and cv. Crousty) were performed to quantitatively assess their tissue composition and to obtain homogeneous samples of embryonic axis, scutellum, starchy endosperm, aleurone layer, hyaline layer, outer pericarp and a composite layer made up of testa+hyaline layer+inner pericarp. Polymeric neutral sugar and phenolic acid contents of the samples were determined and used to identify specific composition patterns in each tissue irrespective of the cultivar. The scutellum and embryonic axis showed the lowest amount of carbohydrates with similar relative amounts of arabinose and xylose (Ara+Xyl), but the scutellum differed from the embryonic axis in its high phenolic acid, in particular ferulate dehydrodimer, content. The peripheral layers of the grains were mainly composed of cell wall polysaccharides, chiefly arabinoxylans but with differing structures. The hyaline layer was mostly composed of arabinoxylan with extremely low Ara/Xyl ratio (0.1), with high amounts of ferulic acid monomers and hence very weakly crosslinked. The aleurone layer differed from the outer pericarp by its much lower Ara/Xyl ratio and lower amounts of ferulic acid dimers and trimers. High proportions of ether-linked phenolic acids (released by alkali at 170 °C) were detected specifically in the seed coat and tissues in the crease region. The possible application of biochemical markers found in the various tissues to monitor wheat grain fractionation processes is discussed.  相似文献

The annual plant Impatiens glandulifera (Himalayan balsam) is the most widespread invasive non‐native weed in the British Isles. Manual control is widely used, but is costly and laborious. Recently, biological control using the rust fungus Puccinia komarovii var. glanduliferae has been trialled. We designed an experiment to assess the impact of these control methods on invertebrate communities in relation to unmanaged and uninvaded habitats, and to determine whether mycorrhizal inoculation aided post‐control recovery of these communities. Sixty invaded and twenty uninvaded field soil blocks were transplanted to the experiment site, where a mycorrhizal inoculum was added to half of all blocks. Biological and mechanical control treatments were applied to twenty invaded blocks independently; the twenty remaining invaded blocks were left intact. Above‐ and belowground invertebrate samples were collected from the blocks at the end of the growing season. Overall, aboveground invertebrate abundance increased with the removal of I. glandulifera, and several groups showed signs of recovery within one growing season. The effect of mechanical control was more variable in belowground invertebrates. Biological control did not affect aboveground invertebrate abundance but resulted in large increases in populations of belowground Collembola. Our experiment demonstrates that mechanical removal of I. glandulifera can cause rapid increases in invertebrate abundance and that its biological control with P. komarovii var. glanduliferae also has the potential to benefit native invertebrate communities.  相似文献

Smooth pea and waxy maize starches were processed under controlled temperature (<160 °C) conditions at low moisture content (30%) using a rheometer with pre-shearing, to simulate extrusion. Specific mechanical energy was measured and remained lower than 103J/g. Starch modification was analysed at different structural levels by differential scanning calorimetry (DSC), X-ray diffractometry, light microscopy and at the granular level by transmission electron microscopy (TEM). Thermal inputs induced crystallite melting and internal disorganisation of granular structure, whereas mechanical input (SME>102J/g) induced granule fragmentation. Under shearing and heating, the concomitant destructuring mechanisms led to a higher rate of modification. Fragmentation itself induced only partial loss of crystallinity (<50%). Small fragments finally melted due to a local temperature increase contributed by interparticular friction. Susceptibility to fragmentation was related to the mechanical properties of starch granules associated with their architecture and botanical origin.  相似文献

A new methodology based on pulsed lasers has been developed in order to estimate wheat outer layer mechanical properties without sample preparation. Laser experiments were carried out with an Argon Fluoride (λ = 193 nm) excimer laser source delivering pulses of 15 ns duration. Wheat grains from two cultivars were irradiated by single laser pulses with a quasi-uniform irradiation and two fluences (2.5 and 5 J cm−2). The ablation flux was characterized by environmental scanning electron microscopy before measuring the removed material on cross-sections observed by confocal scanning laser microscopy. Specific image treatment was carried out to obtain the ablation flux (amount of removed matter per pulse). Pericarp, seed coat and aleurone layer were gradually ablated under the laser conditions used in this work. Their ablation thresholds were different and could be related to tissue cohesion. Specific behaviour of seed coat layer (8 μm) could be emphasised with this technique. Pulsed laser ablation could be a potential methodology to reveal indirectly wheat grain layer cohesion.  相似文献

Yield levels in smallholder farming systems in semi-arid sub-Saharan Africa are generally low. Water shortage in the root zone during critical crop development stages is a fundamental constraining factor. While there is ample evidence to show that conservation tillage can promote soil health, it has recently been suggested that the main benefit in semi-arid farming systems may in fact be an in situ water harvesting effect. In this paper we present the result from an on-farm conservation tillage experiment (combining ripping with mulch and manure application) that was carried out in North Eastern Tanzania from 2005 to 2008. Special attention was given to the effects of the tested treatment on the capacity of the soil to retain moisture. The tested conservation treatment only had a clear yield increasing effect during one of the six experimental seasons (maize grain yields increased by 41%, and biomass by 65%), and this was a season that received exceptional amounts of rainfall (549 mm). While the other seasons provided mixed results, there seemed to be an increasing yield gap between the conservation tillage treatment and the control towards the end of the experiment, and cumulatively the yield increased with 17%. Regarding soil system changes, small but significant effects on chemical and microbiological properties, but not on physical properties, were observed. This raises questions about the suggested water harvesting effect and its potential to contribute to stabilized yield levels under semi-arid conditions. We conclude that, at least in a shorter time perspective, the tested type of conservation tillage seems to boost productivity during already good seasons, rather than stabilize harvests during poor rainfall seasons. Highlighting the challenges involved in upgrading these farming systems, we discuss the potential contribution of conservation tillage towards improved water availability in the crop root zone in a longer term perspective.  相似文献

Infection with Brucella ovis was established by conjunctival instillation in 8 male white-tailed deer (Odocoileus virginianus). Infection was transient in young bucks, but persisted in bucks that were mature when inoculated. The deer were euthanatized and necropsied at various intervals after inoculation. Brucella ovis was recovered from a mature buck at necropsy on postinoculation day 429. Four deer had gross lesions and histopathologic changes of the epididymides. A mature noninfected buck confined for 7 months with an infected buck acquired infection and developed epididymal lesions.  相似文献

The backyard chicken (BYC) movement in the USA has increased human contact with poultry and subsequently, human contact with the pathogen Salmonella. However, to date, there have been few studies assessing prevalence of Salmonella in backyard flocks, despite the known public health risk this zoonotic bacterium poses. The objective of this study was to characterize human‐BYC interactions and assess the prevalence of Salmonella among BYC flocks. We interviewed 50 BYC owners using a structured questionnaire to determine flock and household characteristics that facilitate contact with BYC and that may be associated with Salmonella in the BYC environment. Composite faecal material, cloacal swabs and dust samples from 53 flocks housed on 50 residential properties in the Greater Boston, Massachusetts area were tested for Salmonella using standard culture techniques and confirmed using Matrix‐Assisted Laser Desorption/Ionization‐Time of Flight Mass Spectrometer. Microbroth dilution and whole genome sequencing were used to determine phenotypic and genotypic resistance profiles, respectively, and sequence results were used to determine multilocus sequencing type. No owners self‐reported a diagnosis of salmonellosis in the household. Over 75% of a subset of owners reported that they and their children consider BYC pets. This perception is evident in how owners reported interacting with their birds. Salmonella enterica subspecies enterica serotype Kentucky ST152 (serogroup C)—a strain not commonly associated with human infection—was confirmed in one flock, or 2% of tested flocks, and demonstrated resistance to tetracycline and streptomycin. We detected Salmonella at low prevalence in BYC. Further study of the health effects of exposure to zoonotic gastrointestinal pathogens such as Salmonella among families with BYC is warranted.  相似文献
