Ethiopia has a large and diverse population of small ruminants, which contribute substantially to the livelihood and income of the rural poor and the country at large. However, the sector faces a number of challenges. Productivity per animal and flock offtake are both very low. Reasons attributed for the apparent low productivity are as follows: absence of appropriate breeding programmes, lack of technical capacity, inadequate and poor‐quality feeds, diseases leading to high lamb mortality, and underdeveloped markets in terms of infrastructure and information. Historically, sheep and goats have received little policy or investment attention. Genetic improvement of small ruminants could contribute to bridging the productivity gap. In the past, the government of Ethiopia has placed much emphasis on importing exotic genetics and cross‐breeding with local stock as a strategy for genetic improvement. However, this has not led to a significant productivity improvement and the programmes have generally been unsustainable. Currently, there is a change in approach and a recognition of the need to focus genetic improvement efforts on the local genetic resources that are well adapted to the diverse agro‐ecologies and production environments in the country. Community‐based breeding programmes (CBBPs), which focus on indigenous stock and consider farmers’ needs, views, decisions and active participation, from inception through to implementation, have been identified as programmes of choice. The Ethiopian government and the private sector need to invest in strategic areas around CBBPs to make the programme work for the poor and be sustainable in low‐input systems.  相似文献

Doxycycline is an antibiotic that, in addition to the classic antibacterial use, is also prescribed to fight parasitic diseases, like heartworm disease in dogs. Despite the concern that the overuse of this antibiotic may decrease susceptibility of clinically important bacteria, the consequences of the prolonged doxycycline therapy in heartworm-infected dogs have never been studied before. We have analyzed the impact of this therapy on Staphylococcus aureus and Enterococcus antimicrobial resistance. In this study, 17 heartworm-infected dogs (10 that had completed the doxycycline treatment and 7 dogs that had not yet begun) were included. Twenty-four isolates of Staphylococcus aureus were obtained from two locations of each dog. After treatment, 73.3% of isolates were resistant to at least one antibiotic but only 22.2% of isolates before treatment. Most of doxycycline resistant isolates were obtained from dogs that have received treatment. Erythromycin resistance or intermediate susceptibility was detected in 45.6% of isolates, most of them from dogs after treatment. For Enterococci, 48 isolates were obtained from fecal samples (25 before treatment and 23 after treatment). Before treatment, 32% of isolates were resistant at least to one antibiotic while after, this data increase up to 65%. Comparing isolates before and after treatment, a clear increase in resistance to doxycycline (12% against 21.74%) and erythromycin (20% against 39.13%) was observed. Although the present work is a preliminary research, the results encourages the development of further studies to determinate the effect of prolonged doxycycline therapy on antimicrobial resistance.  相似文献

The dynamics of antibody production against Neospora caninum during the gestation period was examined in chronically infected dairy cows. Data were obtained from 86 pregnant parous dairy cows, 21 of which had suffered abortion. The cows belonged to two herds in which a diagnosis of N. caninum infection had been previously confirmed in aborted foetuses. Pregnancy diagnosis and blood collection were performed on post-insemination Days 40, 90, 120, 150, 180, 210, and at parturition or until the time of abortion detection. Blood plasma was tested for antibodies against N. caninum using ELISA. The non-aborting cows were divided into two groups according to whether their antibody values in the second half of gestation had increased or not, while aborting cows were classified as those showing an antibody peak before abortion or those not showing a pre-abortion peak. Differences in antibody values throughout pregnancy in each group of non-aborting and aborting cows were analysed by GLM repeated measures of analysis of variance. While 32 non-aborting cows (49%) showed a significant and consistent increase in anti-Neospora antibody values during the second half of gestation, antibody values in the remaining 33 non-aborting cows were practically constant throughout gestation. An antibody peak around abortion was observed in 11 aborting cows (52%), while antibody values in the remaining 10 aborting cows were similar before and at abortion. Seroprevalence fluctuations, defined as seronegative blood samples at some point during the gestation period, were, furthermore, observed in 2 aborting and 11 non-aborting cows. Our results indicate two clearly distinguishable types of humoral immune dynamics throughout gestation: an increased or flat production of antibodies during the second half of gestation in non-aborting animals and before abortion in aborting cows. The observation that some Neospora-infected dams can exhibit negative antibody values at any time during gestation, particularly at parturition or abortion, prompts future studies designed to explore the use of new ELISA strategies at the farm level.  相似文献

Five mutants (obtained by UV mutagenesis) and the parent strain were selected to produce sparkling wines following the traditional or champenoise method. The wines were aged with the yeast for 9 months, with samples being taken each month for analytical and sensory determinations. The wines elaborated with mutant strain IFI473I demonstrated an accelerated release of protein, amino acids, and polysaccharides. An analysis of the secreted polysaccharides revealed that mannose was the major sugar present. The effects of the products released by yeasts on the foaming properties of the wines were determined by both sensory and instrumental analysis. In all cases, the wines elaborated with mutant strain IFI473I showed improved foaming properties as compared to wines fermented without this strain. Similar results were obtained at a decreased aging time of 6 months, thereby confirming the capacity of IFI473I strain to carry out an accelerated autolysis. These results demonstrate that mutant strain IFI473I can significantly reduce production times of high-quality sparkling wines.  相似文献

Plant pathogens have developed different strategies during their evolution to infect and colonize their hosts. In the same way, plants have evolved different mechanisms acting against potential pathogens trying to infect and colonize their tissues. Regulation of a wide variety of proteins is required in order to perceive the pathogen and to activate the plant defence mechanisms. The apoplast is the first compartment where these recognition phenomena occur in most plant–pathogen interactions, allowing the exchange of different molecules and facilitating inter‐ and intracellular communication in plant cells. Proteomic analysis of the apoplast in recent years has found the initial biochemical responses involved in pathogen recognition and early defence responses. However, this proteomic approach requires some specific experimental conditions to obtain an extract free of cytoplasmic proteins and nonprotein contaminants that affect the subsequent stages of separation and quantification. Obtaining the highest proportion of proteins from the apoplastic space in infected tissues requires different steps such as extraction of apoplastic washing fluids and preparation of total secreted proteins (protein precipitation, solubilization, separation and digestion). Protein identification using mass spectrometry techniques and bioinformatics tools identifying peptides for the extracellular exportation is required to confirm the apoplastic location. This review compiles the most commonly used techniques for proteomic studies, focusing on the early biochemical changes occurring in the apoplast of plants infected by a wide range of pathogens. The scope of this approach to discover the molecular mechanisms involved in the plant–pathogen interaction is discussed.  相似文献

In Mexico, corn production, part of which is sweet corn, is mainly destined for human consumption. In the present work, the morphological quality of sweet corn ears was assessed in response to four levels of soil moisture tension indicating irrigation start (−5, −30, −55, and −80 kPa) and three levels of phosphate fertilization (60, 80 and 100 kg ha−1) in carstic soils in the south-east of Mexico. A factorial experimental design with three replicates was used. The following variables were determined: fresh weight (SCFWh), dry weight (SCDWh), diameter (SCDh), and length (SCLh) of sweet corn ears, all without husk, as well as number of kernels (NKxE), number of unfilled kernels (NUK), number of rows (NRxE), and dry kernel weight per ear (DKW). Yield of fresh (YFSCh) and dry (YDSCh) sweet corn ears, both without husk, and the harvest index (HI) were also determined. HI did not show significant statistical differences among irrigation or fertilization treatments. Regarding the other variables, the effect of the more humid treatments (−5 and −30 kPa) and the effect of the higher phosphorus doses (80 and 100 kg ha−1) were statistically equal (P ≤ 0.01) with the lowest NUK and the highest values of all other variables; therefore, irrigation start at soil moisture tension of −30 kPa and phosphate fertilization application of 80 kg ha−1 are recommended. At this level of soil moisture, the mean values over the three fertilization levels and all the replicates, obtained for SCFWh, SCDh, SCLh and NKxE were 198.5 g, 4.39 cm, 26.72 cm and 467 grains, respectively. According to the regression models, moisture tensions from −11.8 to −24.0 kPa, and phosphate fertilization doses from 87.7 to 102.2 kg ha−1 minimize NUK and maximize the values of the rest of the variables. The highest irrigation water use efficiency was found in the moisture tension treatment of −30 kPa with an increase of 27 kg ha−1 ears for each millimeter of applied irrigation water.  相似文献

The concern that climate change may increase fire frequency and intensity has recently heightened the interest in the effects of wildfires on ecosystem functioning. Although short-term fire effects on forest soils are well known, less information can be found on the long-term effects of wildfires on soil fertility. Our objective was to study the 17-year effect of wildfires on forest net mineralization rates and extractable inorganic nitrogen (N) and phosphorus (P) concentrations. We hypothesize that (1) burned forest stands should exhibit lower net mineralization rates than unburned ones; (2) these differences would be greatest during the growing season; (3) differences between soil variables might also be observed among plots from different years since the last fire; and (4) due to fire-resistant geochemical processes controlling P availability, this nutrient should recover faster than N. We used a wildfire chronosequence of natural and unmanaged Pinus canariensis forests in La Palma Island (Canary Islands). Soil samples were collected during winter and spring at 22 burned and unburned plots. We found significantly higher values for net N mineralization and extractable N pools in unburned plots. These differences were higher for the winter sampling date than for the spring sampling date. Unlike extractable N and N mineralization rates, extractable P levels of burned plots exhibited a gradual recovery over time after an initial decrease. These results demonstrate that P. canariensis forest soils showed low resilience after wildfires, especially for N, and that this disturbance might induce long-term changes in ecosystem functioning.  相似文献

Plasma concentrations of PAG‐1 are used for pregnancy diagnosis and as a marker of placental/foetal well‐being, while those of PAG‐2 may be an indicator of abortion risk in Neospora caninum‐infected cows. Studies have shown that N. caninum infection modifies PAG‐1 and PAG‐2 patterns in maternal blood plasma. However, no prior work has examined the effects of N. caninum infection on concentrations of PAGs in foetal fluids. In this study, PAG‐1, PAG‐2 and pH levels were determined in the amniotic and allantoic fluids of foetuses collected at 152 days of gestation from control uninfected dams and from dams experimentally infected with N. caninum on Day 110 of gestation. Foetal fluids from infected foetuses had significantly higher PAG‐2 concentrations (p = 0.026) and pH values (p = 0.02) than fluids from non‐infected foetuses. In infected foetuses, significantly higher concentrations of PAG‐1 (p < 0.001) and PAG‐2 (p < 0.001) were detected in fluid samples showing antibodies against N. caninum than those without antibodies. Moreover, pH values were significantly higher (p = 0.011) in foetal fluid samples with antibodies than in samples from non‐infected foetuses. In conclusion, this is the first report on the effect of N. caninum infection on PAG levels in foetal fluids. Our results indicate that following the experimental infection of dams with N. caninum on Day 110 of gestation, foetal fluids collected from the infected foetuses of these dams featured higher PAG‐1 and PAG‐2 levels and pH values than fluids from non‐infected controls, provided that the samples tested showed the presence of antibodies. The clinical implications of these findings are that following infection with N. caninum, most cows will experience some level of placental damage and that this injury correlates with foetal fluid PAG levels and pH.  相似文献
