An indirect, competitive enzyme-linked immunosorbent assay (ELISA) has been developed for quantitation of soy protein in meat products. The methodology allows rapid aqueous extraction of meat samples into a liquid form suitable for assay. The assay is highly specific for soy protein and is designed to measure soy protein levels between 1 and 10% of the wet weight of the sample. Standardized, stabilized reagents for carrying out the procedure are commercially available in a kit. The analysis, including sample preparation, can be completed within a workday, and the actual immunoassay in less than 60 min.  相似文献

The effects of organic matter removal, soil compaction, and vegetation control on soil microbial biomass carbon, nitrogen, C-to-N ratio, and functional diversity were examined in a 6-year loblolly pine plantation on a Coastal Plain site in eastern North Carolina, USA. This experimental plantation was established as part of the US Forest Service's Long Term Soil Productivity Study. Sampling was undertaken on eight treatments within each of three blocks. Treatments sampled included main 2×2 factorial treatments of organic matter removal (stem-only or complete tree plus forest floor) and compaction (none or severe) with split-plot treatment of vegetation control (none or total vegetation control). Two blocks were located on a somewhat poorly drained, fine-loamy, siliceous, thermic aeric Paleaquult (Lynchburg soil) and one on a moderately well drained, fine-loamy, siliceous, thermic aquic Paleudult (Goldsboro soil). Soil microbial C and N were positively related with soil C and N, respectively. Microbial C and N on the Lynchburg soil were higher than those on the Goldsboro soil. Organic matter removal decreased microbial N. Compaction reduced microbial C-to-N ratio. Vegetation control decreased microbial C and C-to-N ratio. The number of C compounds utilized by bacteria was not affected by soil type or treatment. However, soil types and treatments changed bacterial selections for a few C compounds on BIOLOG plates. Soil microbial properties varied more due to the natural soil differences (soil type) as compared with treatment-induced differences.  相似文献

The defined substrate technology (DST) method is a reagent system designed to enumerate specific target microbes(s) from a mixture of bacteria. The system simultaneously enumerates total coliforms and Escherichia coli directly from a water sample. The reagent contains o-nitrophenyl-beta-D-galactopyranoside (ONPG), which is hydrolyzed by total coliforms to produce a yellow chromogen, and 4-methylumbeilliferyl-beta-D-glucuronide (MUG), which is hydrolyzed and fluoresces when E. coli organisms grow. Noncoliform bacteria are suppressed and cannot metabolize the indicator nutrients. Nine laboratories participated in a field evaluation of the method, which covered a wide range of surface and subsurface water sources and water-processing modalities, including the examination of natural samples. The DST system was compared to multiple-tube fermentation (MTF) (quantitative) and presence-absence (P-A) (qualitative) Standard Methods formats. Comparison of water samples from natural sources by using the most probable number (MPN) procedure showed that the DST test was equivalent to the currently used MTF test. Results from the DST and the qualitative P-A procedure showed that these tests agreed with each other in 94% of the water samples analyzed. Specificity of the DST method was established by subculturing a species consistent with a total coliform or E. coli from each positive tube. Eight laboratories participated in a collaborative study of the method. Each laboratory received 3 concentrations of E. coli (organisms/100 mL); 10 (low); 60 (medium); and 120 (high). Mixtures of zeolite and phosphate rock (PR) have the potential to provide slow-release fertilization of plants in synthetic soils by dissolution and ion-exchange reactions. This study was conducted to examine solubility and cation-exchange relationships in mixtures of PR and NH4- and K-saturated clinoptilolite (Cp). Batch-equilibration experiments were designed to investigate the effect of PR source, the proportion of exchangeable K and NH4, and the Cp to PR ratio on solution N, P, K, and Ca concentrations. The dissolution and cation-exchange reactions that occurred after mixing NH4- and K-saturated Cp with PR increased the solubility of the PR and simultaneously released NH4 and K into solution. The more reactive North Carolina (NC) PR rendered higher solution concentrations of NH4 and K when mixed with Cp than did Tennessee (TN) PR. Solution P concentrations for the Cp-NC PR mixture and the Cp-TN PR mixture were similar. Solution concentrations of N, P, K, and Ca and the ratios of these nutrients in solution varied predictably with the type of PR, the Cp/PR ratio, and the proportions of exchangeable K and NH4 on the Cp. Our research indicated that slow-release fertilization using Cp/PR media may provide adequate levels of N, P, and K to support plant growth. Solution Ca concentrations were lower than optimum for plant growth.  相似文献

Summary Using two tussock grasses, Agropyron desertorum, a grazing-tolerant species, and A. spicatum, a grazing-sensitive species, we measured the responses of vesicular-arbuscular mycorhizae (VAM) to clipping and soil moisture status. The clipping was carried out over a 4-year period (1982–1985). For the soil moisture × clipping studies, a rainout shelter was used to create drought stress, and irrigation was used to create high moisture conditions over a 2-year period (1984 – 1985). No consistent patterns in VAM infection frequency and VAM fungal spore counts were observed, either between species or among treatments. Increased moisture tended to increase total root and total VAM length, but there were no consistent moisture × clipping effects. Nor did the phenology of the fungus follow consistent patterns: there were similar lengths of root containing vesicles and arbuscules in 1984, and predominantly vesicles in 1985. The greatest effect was a large difference in VAM activity between years regardless of treatment, despite our attempts to use extreme environmental conditions and a rigorous sampling design. The VAM of both plant species were highly correlated. We postulate that VAM are highly plastic and that they respond to environmental pressures that we do not yet understand.  相似文献

The cytotoxicity of cholesterol and a mixture of beta-sitosterol/campesterol (50%/40%) and their oxides was examined in a cultured-derived macrophage cell line, C57BL/6. Cell numbers, lactate dehydrogenase (LDH) leakage, protein content, lipid uptake, and mitochondria dehydrogenase activity were determined after exposure of cell mononlayers to sterols and sterol oxides at a concentration of 200 microg/mL for up to 120 h. Results indicate that the oxides of cholesterol, beta-sitosterol, and campesterol exhibited similar patterns of toxicity as indicated by LDH leakage, cell viability, and mitochondria dehydrogenase activity. Greatest cell damage was associated with treatments containing 5 alpha,6 alpha-epoxide or cholesterol oxides, followed by beta-sitosterol/campesterol oxides, cholesterol, and beta-sitosterol. The oxides of beta-sitosterol/campesterol caused less LDH leakage and less of an effect on protein content. Results of this study demonstrate that phytosterols contained in vegetable oils, when subjected to frying conditions, do oxidize and may cause cellular damage in an in vitro cell line similar to cholesterol oxides, although less severe.  相似文献

Flow-injection Donnan dialysis is demonstrated for the extraction of lead in sweeteners using flame atomic absorption spectroscopy (FAAS). For spiked concentrations in the low microgram per gram range, recoveries were greater than 90%, and the relative standard deviation was typically less than 10% for a 15-min dialysis procedure. The method detection limit is 350 ng/g. Donnan dialysis is shown to be successful for the extraction of lead in sucrose, corn syrup, and honey but limited in performance for molasses and artificial syrup. This paper also includes a comparison to other procedures for the determination of lead in sweeteners and presents options for realizing improved method performance with Donnan dialysis.  相似文献

The ability of several adsorbents to adsorb dyes from solution has been studied and the relative costs of dye removal are reported based on adsorption capacity only. The application of the results to batch scale commercial systems has been discussed. A detailed study of the adsorption of Telon Blue dye on carbon has been undertaken using fixed beds and the data correlated into a design model using the bed depth service time (BDST) method of analysis.  相似文献
