A multiresidue method analyzing 209 pesticides in 24 agricultural commodities has been developed and validated using the original Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS) procedure and high performance liquid chromatography-positive electrospray ionization-tandem mass spectrometry (LC-MS/MS) analysis. Using solvent-only calibration standards (SOCSs) and matrix-matched calibration standards (MMCSs), it was demonstrated that a minimal concentration of 5-10 μg/kg (part per billion, ppb) of analytes in matrix is required for the consistent identification of targeted pesticides with two MRM transitions. Method performance was validated by the precision and accuracy results obtained from fortification studies at 10, 25, 100, and 500 ppb and MMCSs. The method was demonstrated to achieve an average recovery of 100 ± 20% (n = 4) for >75% of evaluated pesticides at the low fortification level (10 ppb) and improved to >84% at the higher fortification concentrations in all 24 matrices. Matrix effects in LC-MS/MS analysis were studied by evaluating the slope ratios of calibration curves (1.0-100 ng/mL) obtained from the SOCSs and MMCSs. Principal component analysis (PCA) of LC-MS/MS and method validation data confirmed that each matrix exerts its specific effect during the sample preparation and LC-MS/MS analysis. The matrix effect is primarily dependent on the matrix type, pesticide type and concentration. Some caution is warranted when using matrix matched calibration curves for the quantitation of pesticides to alleviate concerns on matrix effects. The QuEChERS method with LC-MS/MS was used to identify and quantitate pesticides residues, with concentrations ranging from 2.5 to >1000 ppb in a variety of agricultural samples, demonstrating fitness for screening and surveillance applications.  相似文献

This paper is an extensive review of the research that has been undertaken on near-infrared (NIR) and mid-infrared (MIR) spectroscopy applied to soil particularly for determining carbon (C) content. The objective is to determine which acquisition method (NIR, MIR, in the field or in the laboratory) might be recommended for the purpose of C stock measurement with a particular interest in carbon credit trading. For this purpose, an optimal method has to satisfy the dual constraints of low-cost and high throughput analysis. The various methods proposed in the literature are compared. In order to make comparisons as reliable as possible, special attention has been paid to the conditions of data acquisition (sample preparation), and to calibration and validation procedures. In particular, whether the validation has been carried out on fully independent samples or on samples similar to the ones of the calibration set greatly influences the results. Also, for C stock measurement, it is absolutely necessary to measure the bias of the prediction in order to be conclusive about the feasibility of the method. However, only few researchers provide this parameter and we recommend including it as a matter of course in future reports. Finally, although MIR on dried and ground samples is the most accurate method, on-the-go and in-field sensors provide predictions accurate enough to show promise in being a valuable component of technologies that would be used for C-credit purposes. But in order to meet the cost/accuracy trade-off, the main issue using such field sensors is to be able to simultaneously measure the bulk density or, better, to directly measure the volumetric concentration of C in soil. This circumvents the costs of field extraction and laboratory analysis. This is the next great challenge to be met by soil scientists.  相似文献

By manipulating colloidal microspheres within customized channels, we have created micrometer-scale fluid pumps and particulate valves. We describe two positive-displacement designs, a gear and a peristaltic pump, both of which are about the size of a human red blood cell. Two colloidal valve designs are also demonstrated, one actuated and one passive, for the direction of cells or small particles. The use of colloids as both valves and pumps will allow device integration at a density far beyond what is currently achievable by other approaches and may provide a link between fluid manipulation at the macro- and nanoscale.  相似文献

The production of spin-polarized hydrogen atoms from the photodissociation of hydrogen chloride with circularly polarized 193-nanometer light is inferred from the measurement of the complete angular momentum distributions of ground state Cl(2P3/2)and excited state Cl(2P1/2)cofragments by slice imaging. The experimentally measured and ab initio predicted a q(k) (p)parameters, which describe the single-surface and multiple-surface-interference contributions to the angular momentum distributions, are in excellent agreement. For laser pulses longer than about 0.7 ns, the polarization of the electron and the proton are both 36%.  相似文献

The molecular mechanisms involved in the development of type 2 diabetes are poorly understood. Starting from genome-wide genotype data for 1924 diabetic cases and 2938 population controls generated by the Wellcome Trust Case Control Consortium, we set out to detect replicated diabetes association signals through analysis of 3757 additional cases and 5346 controls and by integration of our findings with equivalent data from other international consortia. We detected diabetes susceptibility loci in and around the genes CDKAL1, CDKN2A/CDKN2B, and IGF2BP2 and confirmed the recently described associations at HHEX/IDE and SLC30A8. Our findings provide insight into the genetic architecture of type 2 diabetes, emphasizing the contribution of multiple variants of modest effect. The regions identified underscore the importance of pathways influencing pancreatic beta cell development and function in the etiology of type 2 diabetes.  相似文献

Obesity is a serious international health problem that increases the risk of several common diseases. The genetic factors predisposing to obesity are poorly understood. A genome-wide search for type 2 diabetes-susceptibility genes identified a common variant in the FTO (fat mass and obesity associated) gene that predisposes to diabetes through an effect on body mass index (BMI). An additive association of the variant with BMI was replicated in 13 cohorts with 38,759 participants. The 16% of adults who are homozygous for the risk allele weighed about 3 kilograms more and had 1.67-fold increased odds of obesity when compared with those not inheriting a risk allele. This association was observed from age 7 years upward and reflects a specific increase in fat mass.  相似文献

Abstract

Most seed potatoes in Brazil are produced in beds, pots or boxes containing substrate, but the yields are low. Aeroponic systems are one strategy for increasing the yield. However, the concentration of nutrient solutions, managed by varying the electrical conductivity (EC), is of upmost importance and can alter plant responses. Plant density is another important factor for obtaining high yields. The objective of this study was thus to determine the effect of nutrient-solution EC and plant density on plant and seed potato production in aeroponic system. The experimental design was a randomized block with a split-plot design and four replicates, with plots for EC and subplots for plant density. Two potato cultivars were tested. The best ECs for “Agata” and “Asterix” were 2.1 and 1.7?dS?m?1, respectively. The highest plant density (100 plants m?2) for both cultivars produced the highest minituber yield and economic feasibility.  相似文献
