In Stenocereus queretaroensis (Weber) Buxbaum, an arborescent cactus cultivated in Jalisco, Mexico, for its fruits but studied here in wild populations, stem extension occurred in the autumn at the beginning of the dry season, flowering and fruiting occurred in the spring at the end of the dry season, and new roots grew in the summer during the wet season. The asynchrony of vegetative and reproductive growth reduces competitive sink effects, which may be advantageous for wild populations growing in infertile rocky soils. Seasonal patterns of sugars in the roots and especially the stems of S. queretaroensis were closely related to the main phenological stages, becoming lower in concentration during periods of major stem extension. Cessation of stem extension occurred in 100-year-old plants for which injection of GA(3) reinitiated such growth. Isolated chlorenchyma cylinders had maximum extension in a bathing solution containing 0.1 &mgr;M gibberellic acid.  相似文献

Isolated cotyledons from mature Pinus pinea L. embryos were cultured in vitro in a factorial combination of 4.4, 10 and 44.4 microM N6-benzyladenine (BA) for 2, 4, 8, 16 and 35 days to optimize shoot regeneration. Incubation of explants in 44.4 microM BA for 4 days, in place of the standard incubation in 4.4 microM BA for 35 days, reduced the entire culture period to 4 weeks. Shortening the culture period had no significant effect on the caulogenic response or the number of buds formed per cotyledon. To establish the relationship between key moments in the caulogenic process induced by 4.4 microM BA and the endogenous concentrations of the active forms of BA and other isoprenoid-type cytokinins (CKs), we examined uptake, metabolism and amount of BA, as well as the amounts of zeatin, dihydrozeatin and their ribosides in P. pinea cotyledons after 1, 2, 6, 12 and 24 h, and 2, 4, 8, 16 and 35 days of exposure to 8-[14C]BA. Uptake and release of BA were associated with water movement between explants and the medium during the first 8 days of culture. The interconvertible forms of BA were the main metabolites formed in the tissues. Inactivation of BA as a result of conjugation or oxidation was insignificant. The endogenous concentration of BA + N6-benzyladenosine was 20-fold higher than the exogenously applied BA during the competence acquisition phase (Days 0-3). The concentration of isoprenoid-type CKs also increased 16-fold and then decreased during this time. Induction of shoot buds (Days 4-8) was characterized by a second peak of BA uptake by explants that triggered the synthesis of N6-benzyladenosine-5 -monophosphate and by the maintenance of isoprenoid-type CKs. Reestablishment of CK homeostasis marked the shift from the induction phase to the shoot development phase in this organogenic process (Days 8-12).  相似文献

Protein fractions from transgenic Bt and non-transgenic maize varieties, extracted by the Osborne solvent fraction procedure, were characterized for the first time by perfusion and monolithic RP-HPLC in very short analysis times. Albumins and globulins from different transgenic Bt maizes as well as from their non-transgenic isogenic varieties were eluted in four peaks using perfusion RP-HPLC, whereas prolamins and glutelins were separated in seven peaks. Monolithic RP-HPLC enabled the separation of maize proteins in a large number of peaks showing 6 and 10 main peaks for albumins and globulins, respectively. Prolamins migrated at retention times higher than 5 min as seven peaks, whereas glutelins were separated in three main peaks appearing at retention times higher than 6.0 min. Moreover, chromatograms of the whole protein extracts showed 8 and 11 components for perfusion and monolithic RP-HPLC, respectively. A comparison of the chromatograms of the whole protein extracts relative to transgenic and non-transgenic varieties evidenced quantitative differences on the percentages of area, mainly for peaks 2 and 3 by perfusion RP-HPLC and for peaks 3 and 7 by monolithic RP-HPLC. A discriminant analysis based on these proteic profiles was carried out to classify and predict transgenic Bt maize lines, achieving 100% correct classification using perfusion RP-HPLC.  相似文献

Sustainable forest management requires knowledge of forest structure and dynamics as well as an estimation of growing stock. The forest inventory provides the data for estimating stand variables. The measurement device MU2005-01738, patented by the Center for Forest Research (INIA-CIFOR), provides stereoscopic hemispherical images which can allow the 3D restoration of the stand around the sampling point. The aim of this study is to develop a methodology for forest stand mapping as well as tree diameter and height measurement along with volume estimation from the stereoscopic hemispherical images provided by the MU2005-01738. Using the MU2005-01738, Eucalyptus globulus Labill. plantations were sampled. Distance, diameter, height and volume were derived from the stereoscopic hemispherical images of 30 trees located at distances ranging from 0 to 15 m from the device. These variables were then compared with field measurements and the estimation errors analyzed. The (root-mean-squared error) RMSE was 0.23 m (8.95 %) for tree position and 1.51 cm (10.43 %) for diameter at breast height measurement at distances of less than 8 m. In the case of stem height and individual tree volume estimation, the RMSE was 2.59 m (23.05 %) and 0.025 m3 (17.94 %), respectively. The analysis of measurement errors indicated that the measurement precision decreases beyond 8 m from the device as well as for directions close to the baseline (the line between the optical centers of the two images), whereas the precision was highest for directions near to the line which is perpendicular to the base line. Future research should focus on improving measurement accuracy and possible applications in the field of forestry of the techniques presented in this study.  相似文献

The carrageenophytic red alga Gigartina skottsbergii presents several biological constraints for its cultivation such as restricted temporal availability and high spore mortalities that affect the development of its mass cultivation. For this reason, research to develop alternative propagation methods has been undertaken. Previous laboratory studies demonstrated that manipulating temperature, light and nutrients could enhance healing and regeneration of this seaweed. In this study nursery and field experiments were undertaken to establish the possibility to regrow G. skottsbergii in conditions similar to those applied in mass cultivation practices. Frond fragments and rhizoids were tested as alternative ways to obtain new plants. Our results indicate that regeneration occurs in the field, and can be managed in nursery conditions. The addition of a nitrogen source enhances the growth from 0.6 to 1.0% d− 1 of the healed fragments and the use of a photon flux density above 50 μmol m− 2 s− 1 increases the growth rate but decrease the survival of the explants. Explants can be transferred to field conditions and grow at similar rates to those registered in the nursery (0.5% d− 1). It is also possible to propagate rhizoids of G. skottsbergii and it seems that the attachment of a frond portion to the substratum, enhanced its survival and regeneration capacity. Finally, this study demonstrated that rhizoids attached to rocks can regenerate complete plants in nature, a feature which could be a useful for developing a sustainable harvesting methodology.  相似文献

The field performance of the native Pappophorum vaginatum, the naturalized Eragrostis curvula and various cultivars of the introduced Achnatherum hymenoides and Leymus cinereus was evaluated as potential forage resources in rangelands of arid Argentina during the warm seasons of 2007/2008 and 2008/2009. Plants of these grass species, obtained from seeds, were transplanted to the field in 2006, when they were 1 year old. During the study years, there were two defoliation managements: plants of all study genotypes either remained undefoliated (controls) or were defoliated twice a year during spring at 5 cm stubble height. Despite tiller number being lower (P < 0·05) on defoliated than on undefoliated plants, and total leaf length per unit basal area being similar (P > 0·05) between defoliation managements by mid‐spring, there were no differences (P > 0·05) in dry weight production between defoliated and undefoliated plants in all genotypes at the end of the second growing season. Plants of one or more of the introduced genotypes showed a similar (P > 0·05) or greater (P < 0·05), but not lower, tiller number per plant and per square centimetre, daughter tiller production, total leaf length and dry weight production per unit basal area than the native species at the end of the first and/or second growing seasons. These morphological variables were similar (P > 0·05) or greater (P < 0·05) in the native than in the naturalized genotype. Plant survival, however, was lower (P < 0·05, overall mean = 20%) in the introduced than in the native (>70%) or naturalized (>39%) genotypes at the end of the first or second growing seasons.  相似文献

A commercially available polyester resin was reinforced with cabuya fibers. The experimental variables were the fiber loading and the length of the fiber. Tensile strength, flexural strength, and the Izod impact resistance were measured for the samples and compared to the polyester resin performance without reinforcement. Mechanical properties of the cabuya fiber reinforced material were also compared with the same resin but reinforced with glass fibers. An increase in fiber load decreases the tensile strength for the cabuya reinforced composite, where a value of 52.6 MPa corresponded to the tensile stress of the resin without reinforcement and a value of 34.5 MPa for the best reinforcement achieved with cabuya. An increase in both fiber load and length increases the Young’s modulus of the cabuya reinforced material, and a maximum value of 2885 MPa was obtained. The Young’s modulus and impact resistance values for the cabuya composite (2885 MPa and 100.87 J/m, respectively) reached higher values than those obtained for non-reinforced polyester material (2639 MPa and 5.82 J/m, respectively), and lower than the glass fiber composite (5526 MPa and 207.46 J/m, respectively); while the tensile and flexural strength obtained for the cabuya composite (34.5 MPa and 32.6 MPa, respectively) were lower than the unreinforced (52.6 MPa and 62.9 MPa, respectively) and glass fiber reinforced polyester (87.3 MPa and 155 MPa, respectively).  相似文献

Predictive ability of yet‐to‐be observed litter size (pig) grain yield (wheat) records of several reproducing kernel Hilbert spaces (RKHS) regression models combining different number of Gaussian or t kernels was evaluated. Predictive performance was assessed as the average (over 50 replicates) predictive correlation in the testing set. Predictions from these models were combined using three different types of model averaging: (i) mean of predicted phenotypes obtained in each model, (ii) weighted average using mean squared error as weight or (iii) using the marginal likelihood as weight. (ii) and (iii) were obtained in a validation set with 5% of the data. Phenotypes consisted of 2598, 1604 and 1879 average litter size records from three commercial pig lines and wheat grain yield of 599 lines evaluated in four macro‐environments. SNPs from the PorcineSNP60 BeadChip and 1447 DArT markers were used as predictors for the pig and wheat data analyses, respectively. Gaussian and univariate t kernels led to same predictive performance. Multikernel RKHS regression models overcame shortcomings of single kernel models (increasing the predictive correlation of RKHS models by 0.05 where 3 Gaussian or t kernels were fitted in the RKHS models simultaneously). None of the proposed averaging strategies improved the predictive correlations attained with single models using multiple kernel fitting.  相似文献
