A DNA vaccine against contagious agalactia was developed for the first time, encoding the P48 of Mycoplasma agalactiae. Specific immune responses elicited in BALB/c mice were evaluated. Both total IgG and IgG1 were detected in mice vaccinated with pVAX1/P48. Proliferation of mononuclear cells of the spleen, levels of gamma interferon, interleukin-12, and interleukin-2 mRNAs were enhanced in immunized animals. Results indicate that pVAX1/P48 vaccination induced both Th1 and Th2 immune responses. Nucleic acid immunization could be a new strategy against M. agalactiae infections and may be potentially used to develop vaccines for other Mycoplasma diseases.  相似文献

We report the evaluation of the floral origin of honey by analysis of its volatile organic compounds (VOCs) profile, joined with the use of combined pattern recognition techniques. Honey samples, from five floral origins, were analyzed by headspace solid-phase microextraction-gas chromatography-mass spectrometry, selecting 35 VOCs out of the entire profiles, which were analyzed by hierarchical cluster analysis (HCA), stepwise discriminant analysis (SDA), and K-nearest-neighbor (KNN). Both HCA and SDA were used as exploratory tools to select a group of VOCs representing similitude and differences among studied origins. Thus, six out of 35 VOCs were selected, verifying their discriminating power by KNN, which afforded 93% correct classification. Therefore, we drastically reduced the amount of compounds under consideration but kept a good differentiation between floral origins. Selected compounds were identified as octanal, benzeneacetaldehyde, 1-octanol, 2-methoxyphenol, nonanal, and 2-H-1-benzopyran-2-one. The analysis of VOC profiles, coupled to HCA, SDA, and KNN, provides a feasible alternative to evaluate the botanical source of honey.  相似文献

The activity of 8-prenylapigenin (8-PA) and its 3'-methoxylated analogue isocannflavin B (IsoB) was investigated in estrogen-dependent T47-D and estrogen-independent MDA-MB-231 human breast cancer cell lines. 8-PA showed a biphasic effect on T47-D cell proliferation, while no significant effect was observed on MDA-MB-231 cells. Conversely, IsoB exhibited only an inhibitory effect on T47-D cell proliferation, accompanied by the appearance of an intense intracytoplasmic vacuolization of autophagic origin. Moreover, biochemical analysis showed that IsoB reduced Akt phosphorylation and p21Cip1 expression in T47-D cells. These data show that the prenylflavone moiety is a versatile platform for the induction and modulation of bioactivity.  相似文献

Lippia alba (Mill.) N.E. Brown, the ginger grass, is a medicinal and aromatic plant widely used in Latin America. In this work the effect of the geographical origin and different drying methodologies on L. alba leaves production, essential oil content and composition were evaluated. Field experiments were conducted in three different municipalities (Amargosa, Cruz das Almas and Santo Antônio de Jesus) from Bahia region (Brazil) and two drying methodologies (traditional and artificial with controlled conditions) were assessed. Fresh and dried leaves essential oils were extracted by hydro-distillation and their volatiles were determined by gas chromatography.The geographical origin affected the mass production, essential oil content and both qualitative and quantitative fractions of their composition. Leaves from Cruz das Almas had higher yield in essential oil content. Overall, 26 oil constituents were identified. Carvone, germacrene D and limonene were the main oil constituents. The drying methodologies combined with the geographic origin influenced essential oil composition showing that L. alba constituents were influenced by post-harvest treatments. Some constituents were sensitive to both methods applied, while some enhanced their content like germacrene D.  相似文献

A trial was conducted to evaluate the growth performance and survival of Chelidonichthys lucerna. A total of 13 352 180‐days old juveniles (5.5 ± 2 g; 5 ± 1 cm) were reared at two different densities (A‐EXP = 68 fish m?3; B‐FFA = 15 fish m?3) in eight tanks (four tanks per group) for 360 days. The welfare status and meat quality of fish were evaluated for the A‐EXP and B‐FFA groups in comparison with wild‐caught fishery gurnard (C‐WID). The survival rate was high for both A‐EXP (79%) and B‐FFA (93.5%). B‐FFA fish had the highest specific growth rate (1.16 vs. 1.07; P < 0.05), and were heavier than A‐EXP fish (321 ± 40 g vs .239 ± 44 g; P < 0.01). Rearing conditions did not affect blood metabolites, except for glucose concentrations, which were higher in C‐WID (P < 0.05). The meat quality traits showed that reared groups were fatter (2.8–3.2%) than C‐WID (0.94%); total n‐3 fatty acids (19.02–19.26%) were lower in reared groups than C‐WID (29.99%); and EPA + DHA were similar in all groups (15.1–16.61% vs. 27.99%). Despite the good growth and survival, the final mean weight was below that requested by the market (400–500 g). Future research efforts should focus on reducing the feed conversion rate (3:1).  相似文献

BACKGROUND: Three groups of natural enemies are fundamental in citrus IPM in Spain: coccinellid and phytoseiid predators and hymenopteran parasitoids. Tetranychus urticae Koch is an important pest affecting citrus, for which biological control has not yet been achieved; therefore, acaricides are commonly used to control it. The goal of this study was to measure the efficacy of different acaricides on this mite and their side effects on three natural enemies relevant for citrus (Cryptolaemus montrouzieri Mulsant, Neoseiulus californicus McGregor and Aphidius colemani Viereck).RESULTS: Some products proved highly effective against T. urticae and harmless to A. colemani (mineral oil, tebufenpyrad, clofentezine and fenazaquin). However, almost all products tested were slightly harmful for both the predators considered. Fenazaquin was even moderately harmful for N. californicus. CONCLUSION: Further studies, like that presented here, are necessary to gain a better understanding of integrating biological and chemical controls. When considering both efficacy and side effects on beneficial arthropods, the best options would seem to be mineral oil, tebufenpyrad and clofentezine. However, it is urgent to complete testing of the side effects of the acaricides used in citrus. This question is crucial if the fact that two recently introduced Tetranychidae are being controlled in citrus by chemical means exclusively is considered.  相似文献

The interactions taking place in composite dough containing rice flour and soybean proteins (5% w/w) in the presence of transglutaminase, an enzyme with cross‐linking activity, were studied using different electrophoretic analyses. The interaction between rice proteins and soybean proteins was intensified by the formation of new intermolecular covalent bonds catalyzed by transglutaminase and the indirect formation of disulfide bonds among proteins. The main protein fractions involved in those interactions were both β‐conglycinin and glycinin of soybean and the glutelins of the rice flour, although albumins and globulins were also cross‐linked. The addition of soybean proteins to rice flour improves the amino acid balance and they also might play an important role on the rice dough properties because soybean proteins interact with rice proteins, yielding protein aggregates of high molecular weight.  相似文献

Numerous plants ofSilene nicaeensis having symptoms resembling those associated with the presence of phytoplasmas were observed in an extensive coastal area in the south of Italy. Microscopic observation showed histological abnormalities in the organization of tissues in symptomatic plants, and molecular tests, including PCR/RFLP analyses and nucleic acid sequencing, revealed the presence of phytoplasmas belonging to the aster yellows group (‘Candidatus phytoplasma asteris’). The African citrus psyllid (AfCP), Trioza erytreae (Del Guercio) (Hemiptera, Triozidae), recently has been found in northwestern Iberian Peninsula (Spain and Portugal). The insect is an important citrus pest because it transmits the phloem-limited bacteria Candidatus liberibacter spp., the causal agents of huanglongbing (HLB), the most devastating disease affecting citrus. The bacteria can be acquired by both AfCP nymphs and adults, but only adults can spread the pathogen. HLB has been detected neither in the Iberian Peninsula nor in Madeira and the Canary Islands, where the psyllid was recorded more than a decade ago. In the latter European islands, the eradication programmes of AfCP failed. Chemical-based control strategies are costly, could trigger increases of other pests and may have negative impact on the environment. The adoption of rigorous quarantine measures is extremely important for areas free of the psyllid. These measures likely represent the most effective prevention strategies for psyllid containment, because the geographic expansion of psyllids in citrus is mainly due to human activities, especially by the transport of plants and freshly harvested fruits from infested to uninfested areas. Many natural enemies may contribute to the reduction of its populations and consequent spread; hence, conservation biological control should be emphasised, especially in areas where the psyllid is not abundant. Classical biological control programmes should also be implemented in HLB-free areas by introducing effective AfCP parasitoids. In areas where HLB is detected, biological control is difficult to apply, and a rigorous chemical control program targeting the psyllid could complement this strategy. This report is an updated review of AfCP and strategies for its control in anticipation of its possible further spread in Europe and the Mediterranean Basin.  相似文献
