In previous studies, it has not been reported that protein isolated from chia interferes favorably with antibacterial activity, and reduces cholesterol synthesis. The objective of this study was to determine whether commonly used commercial microbial proteases can be utilized to generate chia protein-based antibacterial and hypocholesterolemic hydrolysates/peptides, considering the effects of protein extraction method. Alcalase, Flavourzyme and sequential Alcalase-Flavourzyme were used to produce hydrolysates from chia protein (CF), protein-rich fraction (PRF) and chia protein concentrates (CPC1 and CPC2). These hydrolysates were evaluated for their antimicrobial activity against Gram-positive (G+) and Gram-negative (G?) microorganisms. The protein hydrolysates were purified by ultrafiltration through a membrane with 3 kDa nominal molecular weight, for evaluation of hypocholesterolemic activity. An inhibition zone was observed when the hydrolysate was tested against S. aureus, and minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) values were obtained. Peptides from chia protein with molecular mass lower than 3 kDa reduced up to 80.7% of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG-CoA reductase) enzymatic reaction velocity. It was also observed that, independent of the method used to obtain chia proteins, the fractions showed relevant bioactivity. Moreover, the intensity of the bioactivity varied with the method for obtaining the protein and with the enzyme used in the hydrolysis process. This is the first report to demonstrate that chia peptides are able to inhibit cholesterol homeostasis.  相似文献

During calving time on an experimental farm, 32 newborn calves were selected at random and monitored for infection with Cryptosporidium parvum for the first 30 days of their lives. The animals were fed pooled colostrum for 2-3 days after birth and housed in individual pens, which were washed daily using a pressure hose. Fecal smears were examined by microscopy after staining with carbol fuschin for visualization of oocysts. Oocyst shedding was scored semiquantitatively according to the average number of oocysts in 20 randomly selected fields at 1000x magnification. All the animals acquired the infection before 18 days of age. The period of maximum risk was between 9 and 12 days; 50% of the animals were infected by 9.4 days of age. It was found that the earlier the animals acquired the infection, the longer the patent period. Oocyst shedding, which did not always begin with the onset of diarrhea, lasted between 8 and 23 days (mean 12.4+/-3.3 days). Furthermore, fecal samples from 32 periparturient cows (within +/-7 days of giving birth) were filtered, concentrated and examined for oocysts using a fluorescent monoclonal antibody test, which revealed that six of the cows, although asymptomatic, were excreting C. parvum oocysts.  相似文献

The effects of sublethal heat treatments on heat resistance were studied forFusarium oxysporum f.sp.dianthi (Fod) andVerticillium dahliae (Vd), one isolate of each pathogen. Treatments of propagule suspensions of Fod at 55°C and of Vd at 45°C for 30 min were survived by less than 0.001% and 0.01% of the propagules, respectively. Pretreatment of suspensions of Fod at 45°C increased survival of the 55°C treatment up to 0.73% of the propagules and pretreatment of suspensions of Vd at 40°C increased survival of the 45°C treatment up to 0.40%. Induction of heat resistance was dependent on duration of the exposure to the sublethal temperature. With Fod, this duration was shorter for propagules from old cultures than for those from young cultures. Pretreatment at 45°C of a suspension of an 1-week-old culture of Fod induced resistance when lasting 30 min or longer, but not when 20 min or shorter. With Vd, the duration of the pretreatment inducing heat resistance depended on type of culture — white or black — due to differences in microsclerotia formation. Implications of induced heat resistance for control of plant diseases by thermotherapy are discussed.  相似文献

Source water used by woody perennials in a Brazilian savanna (Cerrado) was determined by comparing the stable hydrogen isotope composition (deltaD) of xylem sap and soil water at different depths during two consecutive dry seasons (1995 and 1996). Plant water status and rates of water use were also determined and compared with xylem water deltaD values. Overall, soil water deltaD decreased with increasing depth in the soil profile. Mean deltaD values were -35 per thousand for the upper 170 cm of soil and -55 per thousand between 230 and 400 cm depth at the end of the 1995 dry season. Soil water content increased with depth, from 18% near the surface to about 28% at 400 cm. A similar pattern of decreasing soil water deltaD with increasing depth was observed at the end of the 1996 dry season. Patterns consistent with hydraulic lift were observed in soil profiles sampled in 1995 and 1997. Concurrent analyses of xylem and soil water deltaD values indicated a distinct partitioning of water resources among 10 representative woody species (five deciduous and five evergreen). Among these species, four evergreen and one deciduous species acquired water primarily in the upper soil layers (above 200 cm), whereas three deciduous and one evergreen species tapped deep sources of soil water (below 200 cm). One deciduous species exhibited intermediate behavior. Total daily sap flow was negatively correlated with xylem sap deltaD values indicating that species with higher rates of water use during the dry season tended to rely on deeper soil water sources. Among evergreen species, minimum leaf water potentials were also negatively correlated with xylem water deltaD values, suggesting that access to more readily available water at greater depth permitted maintenance of a more favorable plant water status. No significant relationship between xylem water deltaD and plant size was observed in two evergreen species, suggesting a strong selective pressure for small plants to rapidly develop a deep root system. The degree of variation in soil water partitioning, leaf phenology and leaf longevity was consistent with the high diversity of woody species in the Cerrado.  相似文献

A panel of 377 commercially available monoclonal antibodies (mAbs) specific for a total of 144 CD antigens was submitted to the animal homologue section of the Eighth International Workshop on Human Leukocyte Differentiation Antigens (HLDA8, Adelaide, Australia) for cross-reactivity studies in a range of vertebrate species. Each of the mAbs in this study was screened for positive reactivity with guinea pig splenocytes by flow cytometry. In the first phase of this study 36 of the total 367 mAbs (9.81%) cross-reacted with splenocyte surface molecules. The majority (26 of 36) of these cross-reactive mAbs were analysed further to confirm appropriate cell subset expression by two-color immunofluorescence. Our results indicate that 15 anti-human CD9, CD10, CD14, CD20 (two clones), CD22, CD25, CD29 (two clones), CD32, CD47 (two clones), CD49d, CD49e, and CD86 mAbs exhibit clear cross-reactivity with guinea pig splenocytes. These mAb can potentially be added to the limited repertoire of reagents available for studies in this model system. This data clearly indicates that mouse anti-human CD mAb guinea pig cross-reactions have been defined and that an aim of this HLDA8 section has been fulfilled, i.e., to identify mAbs which recognize conserved, species-independent CD epitopes. These results will contribute to the availability of mAbs and tools in veterinary medicine and immunology.  相似文献

Abstract This study shows that disturbance caused by cover-crop removal (CCR) in an organic olive orchard affects ground-spider populations. The effect of CCR on various organic olive-orchard plots was assessed by monitoring the abundance and diversity of ground-dwelling spiders. Covered plots in the organic olive orchard were compared with uncovered plots where the covers had been removed mechanically. CCR positively affected the most abundant spider species Zodarion styliferum (Simon) (Araneae: Zodariidae) as well as other species of running spiders belonging to the families Gnaphosidae and Lycosidae. Over time, the two types of plots did not significantly differ in diversity or dominance. Similarly, no differences were detected between the study plots in terms of the distribution of individuals when a cluster-similarity analysis was performed. This lack of difference in diversity might be due to the spatial scale used in the study or climatology. Because of their general effects, CCR profoundly changed the abundance of spiders in the olive orchard, but with no clear impact on spider diversity.  相似文献

This paper focuses on strategic management planning of Eucalyptus globulus Labill. plantations in Galicia (northwestern Spain). Several problems associated with the management of these plantations were identified first. Current management plans for plantations of this species consist of the systematic and somewhat arbitrary application of the area control method. Thus, these plans do not take into consideration the optimum length for one full plantation cycle that maximizes the land expectation value, neither do they formulate the area control method through any mathematical programming model for scheduling regeneration harvests. In this paper, we present a modelling approach based on linear and goal programming. The models consider area and volume control regulation strategies, and take into account variations in land productivity among site classes and successive rotation intervals. To illustrate this approach, we applied the models to a neighbourhood community-owned forest currently managed by the Regional Forest Service with a recent management plan. The results showed that the models provided more flexible harvest schedules, and the profitability of eucalypt stands was 64% higher than that under the current management plan. Finally, extensions to this study were identified.  相似文献
