The Glycine max (L.) Merr. cultivar Waseshiroge is highly resistant to several races of Phytophthora sojae in Japan. In order to determine which Rps gene might be present in Waseshiroge, 15 differential cultivars were challenged with 12 P. sojae isolates. None had a reaction pattern identical to that of Waseshiroge, indicating that Waseshiroge may contain a novel Rps gene. In order to characterize the inheritance of Waseshiroge resistance to P. sojae isolates, 98 F2 progeny and 94 F7:8 lines were produced from crosses between the susceptible cultivar Tanbakuro and Waseshiroge. Chi-square tests indicated that segregation fit a 3:1 ratio for resistance and susceptibility in two F2 sub-populations of 42 and 56 seedlings. This and a 46.27:1.46:46.27 (or 63:2:63) ratio for resistance: segregation: susceptibility among the 94 F7:8 lines indicated that resistance was controlled by a single dominant gene. DNA analyses were carried out on Tanbakuro, Waseshiroge and the 94 F7:8 lines, and a linkage map was constructed with 17 SSR markers and nine new primer pairs that amplify marker loci linked to Rps1 on soybean chromosome 3 (linkage group N). The closest markers, Satt009 and T000304487l, map to locations 0.9 and 1.6 cM on each side of the estimated position of the Rps gene, respectively. The results showed that the Rps gene in Waseshiroge is either allelic to Rps1, or resides at a tightly linked locus in a gene cluster. A three-way-contingency table analysis indicated that marker-assisted selection with the two flanking markers could be used in the development of new resistant cultivars.  相似文献

This study examined the effects of residual trees on the height of 18- and 19-year-old Cryptomeria japonica planted in group selection openings of about 0.1 ha at Yufuin in Oita Prefecture in southern Japan. We first developed a general model expressing variation in the height for all of the planted trees within the openings from indices of both between-cohort competition (the effects of residual trees) and within-cohort competition (among the even-aged planted trees). The between-cohort competition index was calculated both with and without taking into account the direction that residual trees were positioned from subject trees. The two models suggested that residual trees located near the northern edge of the opening were not effective competitors for light for the planted trees within the opening. In addition, our results indicated that both between-cohort competition and within-cohort competition are important for explaining tree height in closed stands within group selection openings. Using the general model, we then estimated the potential loss of height growth for locally dominant trees within the openings that could be attributed only to the influence of residual trees. These results indicated that in addition to the effect of opening size, both the degree of crowding and the direction of residual trees from the planted trees are important factors affecting the height of individual trees planted in group selection openings.  相似文献

Phytophthora stem and root rot, caused by Phytophthora sojae, is one of the most destructive diseases of soybean [Glycine max (L.) Merr.], and the incidence of this disease has been increasing in several soybean-producing areas around the world. This presents serious limitations for soybean production, with yield losses from 4 to 100%. The most effective method to reduce damage would be to grow Phytophthora-resistant soybean cultivars, and two types of host resistance have been described. Race-specific resistance conditioned by single dominant Rps (“resistance to Phytophthora sojae”) genes and quantitatively inherited partial resistance conferred by multiple genes could both provide protection from the pathogen. Molecular markers linked to Rps genes or quantitative trait loci (QTLs) underlying partial resistance have been identified on several molecular linkage groups corresponding to chromosomes. These markers can be used to screen for Phytophthora-resistant plants rapidly and efficiently, and to combine multiple resistance genes in the same background. This paper reviews what is currently known about pathogenic races of P. sojae in the USA and Japan, selection of sources of Rps genes or minor genes providing partial resistance, and the current state and future scope of breeding Phytophthora-resistant soybean cultivars.  相似文献

Pacific bluefin tuna (PBF), Thunnus orientalis, is commercially one of the most important species of tuna. In this study, amplified fragment length polymorphism (AFLP) screening was conducted to find the growth‐related polymorphic DNA in cultured PBF. Fish hatched in 2007 were harvested at an age of 818–1994 days. They were categorized into superior, average and inferior growth groups, depending on their growth score at the time of harvest. On AFLP screening of 24 fish, with eight fish from each group, 215 polymorphic DNA fragments were observed. A second amplification, with EcoRI + ACC and MseI + CCC primers, generated a polymorphic fragment of 630 bp at a rate of 80.0% (n = 15) in the superior, 56.3% (n = 16) in the average and 20.0% (n = 15) in the inferior growth groups. Polymerase chain reaction (PCR) primers, which could amplify both AFLP‐positive and AFLP‐negative loci, were developed using the consensus sequence outside the AFLP target fragment. Eleven haplotypes were obtained by sequence analysis of the PCR product at the AFLP target loci. Among those, haplotype 1 was statistically significant in the superior and average growth groups and could be used as a molecular marker for distinguishing the individuals with superior and average growth from those with inferior growth.  相似文献

To apply otolith microstructure to examination of age and growth of juvenile chum salmon Oncorhynchus keta inhabiting coastal waters, formation of otolith increments was investigated for juveniles reared in a seawater aquarium and in net pens. In all otoliths examined, a distinctive check was formed at the time of sea entry of the fish. The deposition of otolith increments after the check was daily for rearing both in the aquarium (57 days) and in the net pens (26 days). Check formation associated with sea entry was also observed in otoliths of juvenile salmon collected 1 km off the coast of Shari, Hokkaido, Japan. Transmitted light observation of otoliths of those fish revealed a transition in otolith increment appearance from dark to light. Otolith Sr: Ca ratio remarkably changed from a low to a high level, coinciding with the transition in otolith appearance. It is suggested that the transition was associated with individual sea entry. This study demonstrated that the check and/or transition associated with sea entry are applicable to a benchmark for otolith increment counts of juvenile chum salmon inhabiting coastal waters.  相似文献

A 303-nucleotide viroid was isolated from an apple tree (Malus × domestica, ‘Fuji’) cultivated in Japan. The viroid had 84.9% overall nucleotide sequence homology to Apple dimple fruit viroid (ADFVd), a member of Pospiviroidae, reported from Italy. This viroid differed from the Italian variant by 47 mutations (38 substitutions, six deletions and three insertions), and most of these mutations occurred on either side of the central conserved region. The leaves and branches of the infected trees did not have any disease symptoms, but the fruits were dimpled and yellow. The infected scions were top-grafted onto a healthy ‘Fuji’ apple tree, which tested positive for this viroid in a northern hybridization analysis, and yellow dimple fruits were produced in the second growing season. We propose that this viroid is a new variant of ADFVd and causes yellow dimple fruit formation in ‘Fuji’ apple trees.  相似文献

 利用 12对AFLP引物 ,以饭豆标准品系M 0 0作对照 ,对来自中国、日本、韩国、尼泊尔、印度、不丹的 14 6份小豆栽培种 (Vignaangularisvar.angularis)和野生种 (Vignaangularisvar.nipponensis)种质的基因组DNA进行扩增 ,得到 313条多态性带。据AFLP多态性数据绘制的聚类图 ,可区分其中的 14 3份种质 ,表明小豆物种 (Vi gnaangularis)存在足够的遗传多样性 ,可用于资源材料的准确鉴别与分类。鉴于此 ,采用新开发的利用AFLP数据揭示核苷多样性的Innan’s进化树分析方法 ,进一步将 14 6份小豆资源分成 7个明显不同的地理演化群 ,即中国栽培种、日本栽培种、日本综合群 韩国栽培种、中国台湾野生种、中国野生种、尼泊尔 不丹栽培种和喜马拉雅野生种演化群。就上述地理演化群的遗传多样性、地理分布以及野生种与栽培种之间可能的演化关系进行了分析 ,初步认为栽培小豆至少应当有 4个不同类型的野生祖先和 3个不同的地理起源。  相似文献

To examine the efficacy of juvenile salmon research as a tool for forecasting adult returns, the results from a study on the early marine life stage of juvenile chum salmon, conducted in the Nemuro Strait during 1999–2002 (i.e., 1998–2001 brood years), were compared with the return rates of adult salmon. Among the four brood years, the 2000 brood year (i.e., salmon migrating to the sea in 2001) was previously reported as showing higher abundance, higher growth rate and better somatic condition during the coastal residency period. Consequently, we expected it to have the highest return rate, under a hypothesis that juvenile survival in coastal residency regulates brood-year strength. Contrary to this expectation, the 2000 brood year had almost the lowest return rate. Alternatively, a statistical model in which sea surface temperature during the first year of marine life and size at release were utilized as explanatory variables reconstructed the actual variability in return rates more accurately than that based on the early marine life stage. Possible reasons for the discrepancy between the results of the juvenile salmon research and adult returns are discussed, and we suggest improvements for future research on juvenile salmon.  相似文献

A canopy height‐to‐row spacing ratio (CHRSR) of 1.0 is expected to be a simple and practical onsite index for soybean growers to determine when further weed control is no longer necessary. This is decided when relative photosynthetic photon flux density (PPFD) reaches 50% under the soybean canopy. To evaluate the validity of this index, a 2‐year field experiment was conducted to compare days after sowing (DAS) to reach a relative PPFD of 50% (defined as PPFD50) and DAS to reach a CHRSR of 1.0 (defined as CHRSR1.0) in 190 accessions with different canopy architectures from a soybean core collection. A total of 104 accessions, for which final relative PPFD reached <10% in 2014, were examined for the differences between CHRSR1.0 and PPFD50. The mean and median values of the difference in 2014 were 0.02 and 2.2, respectively. Similarly, the mean and median values in 2015 were 2.8 and 2.6, respectively. Thus, CHRSR1.0 tended to be achieved slightly later than PPFD50, so a consideration was made that there would be a negligible risk of failure if growers used CHRSR1.0 to determine the termination time for weed control in the most of accessions. Although there were some accessions with a standing type of morphological characteristics or with a smaller or narrower leaf, showing negative differences between CHRSR1.0 and PPFD50 larger than 1 week, a CHRSR of 1.0 could be used to estimate PPFD50 in most of accessions without risk of failure to control weeds.  相似文献
