The present study was undertaken to isolate buffalo preantral follicles (PFs), to test the viability and sizes of buffalo PFs and to examine the effect of various growth factors (insulin-like growth factor, fibroblast growth factor) and an antioxidant (β mercaptoethanol) on the in vitro growth, survival and antrum formation rates of buffalo PFs and growth rates of oocytes in cultured PFs. Preantral follicles from slaughtered buffalo ovaries were recovered by a combined mechanical and enzymatic method. The recovery rates of >40–100, 101–200, 201–300, 301–400 and 401–500 μm PFs were 5.1, 3.2, 3.1, 6.3 and 5.1 per ovary, respectively. The corresponding viability rates were 76.1%, 78.1%, 85.2%, 92.5% and 92.6%, respectively. There was a positive correlation ( r = 0.73) between oocyte size and the follicular size. However, there was no significant correlation between the size of oocyte and its viability at the time of its retrieval from ovary. Insulin-like growth factor and fibroblast growth factor improved the survival of buffalo PFs and regulated their growth in culture. A study was undertaken to investigate the remedial effect of some soil amendments (farmyard manure (FYM), press mud compost (PMC), cereal straw (CS) at 5 t ha?1 and fresh cow dung slurry (FCD) @ 0.5 t ha?1) on dissipation kinetics of imidacloprid and sulfosulfuron under laboratory conditions. Incorporation of CS or FCD was found to be most effective in degrading both the pesticides at faster rates. Dissipation of both the pesticides could be well accounted by two component (1 + 1) first order kinetics. The computed values of parameters revealed that use of organic amendments increased the dissipated fraction of imidacloprid and sulfosulfuron. Incorporation of CS or FCD in soil maintained relatively higher dissipation rate constants for both faster and slower dissipation processes of pesticides in comparison to control. Hence, eco-friendly practices of CS or FCD incorporation as soil amendment in soil can play a vital role in preventing soil and water pollution. 相似文献
1. An experiment was conducted to determine optimal non-phytate phosphorus (NPP) concentrations for White Leghorn (WL) layers (22–72 weeks) fed diet containing 38 g Ca/kg.
2. Eight diets with graded concentrations (1.5–3.25 g/kg in increments of 0.25 g) of NPP were prepared. Each diet was fed to eight pen replicates containing 88 birds in each. Performance data was evaluated in three different phases (phase I-22–37 weeks, phase II-38–53 weeks and phase III-54–72 weeks). Optimum levels of NPP were determined by fitting a quadratic polynomial (QP) regression model.
3. Egg production (EP) was not affected (P = 0.059) by the concentration of NPP and interaction between NPP and diet phase was non-significant, indicating that the lowest concentration (1.5 g/kg diet) of NPP used in the study was adequate across the three phases. However, EP was influenced by phase (P < 0.001).
4. Optimum concentration of NPP for feed intake (FI) was estimated to be 1.5, 1.71 and 2.40 g/kg diet during phases I, II and III, respectively. FI per egg mass (EM) or feed efficiency (FE) responded quadratically with NPP and also differed significantly between phases. Optimum concentration of NPP for FE during phases I, II and III was 1.5, 2.56 and 2.32 g/kg diet, respectively.
5. Egg weight (EW), EM, shell weight and thickness were not affected by NPP concentration although all of these variables (except shell weight) were influenced by phases.
6. Breaking strength of tibia and Ca contents in tibia ash were not affected by the concentration of NPP, but bone ash and P contents in tibia ash were influenced (P < 0.001) by NPP. Predicted optimal concentrations of NPP for responses for tibia ash at 44 or 72 weeks, tibia ash P at 44 weeks and tibia ash P at 72 weeks were 1.55, 2.63 and 1.5 g/kg diet, respectively.
7. Based on the results, it was concluded that WL layers required 1.5 g, 2.63 g and 2.4 g, respectively/kg diet during phase I, II and III with the calculated daily intake of 137.3, 278.3 and 262 mg NPP/b/d. 相似文献
The relative efficacies of three natural estrogens viz., estrone (E1), estradiol-17β (E2) and estriol (E3) to induce synthesis of vitellogenin (Vg) and choriogenin (Chg) were assessed in primary hepatocyte cultures of the Indian
freshwater spotted snakehead, Channa punctata. Hepatocytes were isolated from the spotted snakehead liver by a non-enzymatic protocol. Optimum culture conditions were
standardized for ensuring their viability and functioning. Isolated hepatocytes were cultured for 48 h for monolayer formation
and then exposed to various concentrations (0.001–10 μM) of the three estrogens. Competitive homologous ELISAs, developed
and validated for spotted snakehead Vg and Chg were employed to determine the amounts of these two proteins secreted into
the culture medium after 48 h of incubation. The results reveal that although all the three estrogens were effective in inducing
the production of Vg and Chg in a dose-dependent manner, there were differences in their relative potencies. Of three estrogens,
E1 was the least potent and could induce synthesis of Vg and Chg only at a minimum concentration of 0.5 μM; whereas significant
levels of both the proteins were quantified in culture medium by exposing the hepatocytes to E2 or E3 even at a concentration of 0.001 μM. All three estrogens were effective in inducing synthesis of Vg and Chg in vivo also.
These results suggest the possibility of employing the above in vitro experimental design to monitor the presence of estrogens/estrogen-like
chemicals in natural waters, which could interfere with the estrogen receptor system of fish. This study further points to
the possibility of using Chg, in addition to Vg, as a parameter for screening various chemicals for their estrogenic activity. 相似文献
A feeding experiment was conducted for 135 days to observe the effect of different isonitrogenous (35% crude protein) and isocaloric (385 kcal) diets on the growth and carcass composition of giant freshwater prawn, Macrobrachium rosenbergii ( De Man 1879 ). Three experimental diets (ED1, ED2 and ED3) were prepared using locally available ingredients. These diets differed mainly in terms of percent contribution of major protein sources such as fish meal, soybean meal, groundnut oil cake and mustard oil cake. Experimental diets were evaluated against a commercial diet, which served as the control (CD). Juveniles 1.87–2.44 g in size were stocked at a population density of 40 000 ha−1 and fed thrice daily at 10% in the beginning and reducing gradually to 7% and 5% of the body weight during the experimental period. No significant differences (P>0.05) in the growth performance were observed; however, a significantly (P<0.05) higher yield (721.9 kg ha−1 135 days−1) was recorded for prawn fed with control diet, followed by experimental diet 2 (676.5 kg ha−1 135 days−1, having soybean meal as a major protein source). The survival ranged between 63.8% and 77.7%, with different diets showing significantly higher survival. The apparent feed conversion ratio values of diets ranged between 3.15 and 3.49, with experimental and control diets showing non‐significantly lower AFCR values. At the end of the experiment, representative specimens from each treatment were collected and their carcass composition was measured. Analysis of variance showed that carcass protein and total carbohydrate contents were significantly (P<0.05) higher in prawns fed on a fish–soybean meal‐based diet (ED3) and a control diet. The total lipid contents of prawns, however, did not differ significantly among the various dietary treatments. The results of our study suggest that the experimental diets could be used effectively for M. rosenbergii without compromising growth and flesh quality. 相似文献
The objective of this study was to elucidate the changes in circulating levels of plasma interleukin-8 (IL-8) during peripartum
period in Murrah buffaloes (Bubalus bubalis). IL-8 was estimated in blood plasma of healthy peripartum Murrah buffaloes (n = 6) on days ±30, ±15, ±5, ±3, ±1 and 0 pre- and postpartum with respect to the day of parturition (day 0) in each of the
two different seasons (hot–humid and spring). The mean microclimate Temperature–Humidity Index (THI) during spring season
was significantly lower (p < 0.01) than the corresponding THI in hot–humid season. In both the seasons, plasma IL-8 remained lower in prepartum period
(≤46.56 ± 14.08 pg/ml during spring and ≤ 73.18 ± 18.56 pg/ml during hot–humid season) than in the postpartum period (≥51.41 ± 13.82 pg/ml
during spring and ≥ 84.13 ± 16.97 pg/ml in hot humid season). During spring, the IL-8 levels were significantly higher (P < 0.05) on days +5 and +15 postpartum in comparison to the IL-8 levels on days −30, −5, and −3 prepartum. During hot–humid
season, IL-8 level was significantly higher (P < 0.05) on day +30 as compared to the IL-8 levels on days −30 and −5 prepartum. The correlation between IL-8 and mean microclimate
THI was significant (r = 0.25, P < 0.01). From the results, it is concluded that peripartum period in buffaloes is associated with an inflammatory response

Association mapping was undertaken in common wheat to identify markers associated with pre-harvest sprouting tolerance (PHST). For this purpose, a population of 242 wheat genotypes and 250 SSR markers were used. The population used consisted of diverse germplasm, which carried sufficient phenotypic variation for PHS for conducting association mapping. The population was found to be structured and stratified into 15 sub-populations; the tolerant and moderately tolerant wheat genotypes were distributed in all the sub-populations. This feature of the population along with other information on population structure was used in association mapping using both the available models, the general linear model (GLM) and the mixed linear model (MLM); hopefully, this minimized the rate of false positives. As many as 30 markers were found to be associated with PHST, 26 markers with GLM and 17 markers with MLM; 13 markers were detected using both the approaches. Only eight SSR markers associated with QTL for PHST were such, which were located within the marker intervals that were earlier reported to carry QTLs for PHST. The remaining 22 markers that were found to be associated with PHST could not be associated with any of the genomic regions known to carry QTLs for PHST, which are known to occur on all the 42 chromosome arms of wheat genome. 相似文献

These substances include proteolytic inhibitors, phytohemagglutinins, lathyrogens, cyanogenetic compounds, compounds causing favism, factors affecting digestibility and saponins.
