The purpose of this study was to verify the ability of a probiotic in the feed to maintain the stability of the gut microbiota in chickens after antibiotic therapy and its association with growth performance. One thousand six hundred twenty 1‐day‐old Cobb male were housed in floor pens (36 pens, 45 birds/pen) and were fed corn‐/soya bean meal‐based diets supplemented with or without probiotic (Bacillus subtilis) during the entire rearing phase. From 21 to 24 days of age (three consecutive days), the chickens were submitted to antibiotic therapy via drinking water (bacitracin and neomycin) in order to mimic a field treatment and induce dysbiosis. Growth performance was monitored until 42 days of age. At 2, 4 and 6 days after antibiotic therapy, three chickens from each pen were euthanized and the contents of the small intestine and caeca were collected and pooled. The trial was conducted with four treatments and nine replicates in a 2 × 2 factorial arrangement for performance characteristics (with and without probiotic × with and without antibiotic therapy); for the intestinal microbiota, it was in a 2 × 2 × 3 factorial arrangement (with and without probiotic × with and without antibiotic therapy × 2, 4 and 6 days after the antibiotic therapy) with three replicates per treatment. Terminal restriction length polymorphism (T‐RFLP) analysis showed that the structure of gut bacterial community was shaped by the intestinal segment and by the time after the antibiotic therapy. The number of 16S rDNAs copies in caecum contents decreased with time after the therapeutic treatment. The antibiotic therapy and dietary probiotic supplementation decreased richness and diversity indexes in the caecal contents. The improved performance observed in birds supplemented with probiotic may be related to changes promoted by the feed additive in the structure of the intestinal bacterial communities and phylogenetic groups. Antibiotic therapy modified the bacterial structure, but did not cause loss of broiler performance.  相似文献

We investigated the effectiveness of rotational and permanent grazing exclosure periods for improving topsoil quality in three commercial farms devoted to cattle breeding in sodic grassland (halophytic steppe) soils of the Flooding Pampa of Argentina. We compared two plots under continuous grazing (C1-C2) with two plots under more than 8 yr of rotational grazing management (R1-R2) and two adjacent plots under permanent grazing exclosure for more than 8 (E1) and 4 (E2) yr. Periodic and permanent grazing exclosure periods caused significant (P < 0.05) and progressive increases in topsoil organic carbon content and organic carbon stock (0 ? 20 cm; from 24 to 61 Mg ha? 1) as follows: (C1 = C2) < (R1 = R2 = E2) < E1 plots. Topsoil physical properties (bulk density, structural instability, and bearing capacity) and salinity were higher (P < 0.05) in C1 and C2 than in the other plots, while infiltration rate was higher in the oldest exclosure (E1) than in the other plots. Topsoil pH decreased from C1-C2 plots (9.5 ? 9.9) to R1-R2 plots (7.3 ? 8.2) to E1-E2 plots (6.5 ? 7.5), while SAR was highest in C1-C2 and lowest in E1 plots. We propose a conceptual model leading to soil recovery in this halophytic steppe community, triggered by organic carbon accumulation induced by grazing management. Short-time grazing exclusion periods (i.e., rotational grazing) are a plausible and low-cost management option to be recommended to the farmers in this highly restrictive environment.  相似文献

Florfenicol (FF) is employed in fish farms to contest or prevent bacterial infections. However, this pharmaceutical may produce reactive oxygen species that may cause biochemical changes in antibiotic-treated fish. The aim of this study was to evaluate the effect of FF on Rainbow Trout Oncorhynchus mykiss treated for 10 d with 7.5 and 15 mg/kg FF followed by a withdrawal period of 5 d. Superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, glutathione S-transferase, glyoxalase I and glyoxalase II, total glutathione, lactic dehydrogenase, and alkaline phosphatase were investigated in the livers of treated and untreated fish. A general impairment of antioxidant enzymes and metabolic indicators was measured in FF-treated Rainbow Trout. Onset of oxidative damage may have occurred during the antibiotic treatment as a consequence of the effect of FF toxicity at mainly the highest dose. Nevertheless, the rise in levels of total glutathione and glutathione S-transferase even after the withdrawal period may shield the antibiotic-mediated oxidative processes.

Microbial processes have shown promise for the remediation of uranium and nitrate in groundwater impacted by uranium mine tailings. This study investigated the inhibitory impact of uranium(VI) towards different microbial populations in anaerobic biofilms, including methanogenic, denitrifying, and uranium-reducing microorganisms, which are commonly found at uranium bioremediation sites. Results of batch activity bioassays indicated a very distinct level of toxicity depending on the targeted microbial community. U(VI) caused severe inhibition of acetoclastic methanogenesis as indicated by a 50?% inhibiting concentration (IC50) of only 0.16?mM. Denitrifying populations were also impacted by uranium, but their sensitivity depended on the electron donor utilized. Sulfur-oxidizing denitrifiers were the least affected (IC50 for denitrification activity?=?0.32?mM), followed by H2- and acetate-utilizing denitrifiers (IC50 of 0.20 and 0.15?mM, respectively). In contrast, exposure to U(VI) concentrations up to 1.0?mM did not inhibit the rate of U(VI) bioreduction with H2 as electron donor in the presence or absence of nitrate. On the contrary, a considerable increase in the uranium-reducing activity of the denitrifying and methanogenic mixed cultures was observed with increasing uranium concentrations. The results suggest that microorganisms responsible for U(V) reduction could tolerate much higher uranium concentrations compared to the other microbial populations assayed.  相似文献

Leguminous trees have a potential for phytoremediation of oil-contaminated areas for its symbiotic association with nitrogen-fixing bacteria and arbuscular mycorrhizal fungi (AMF). This study selects leguminous tree associated with symbiotic microorganisms that have the potential to remediate petroleum-contaminated soil. Seven species of trees were tested: Acacia angustissima, Acacia auriculiformis, Acacia holosericea, Acacia mangium, Mimosa artemisiana, Mimosa caesalpiniifolia, and Samanea saman. They were inoculated with AMF mix and nitrogen-fixing bacteria mix and cultivated over five oil levels in soils, with five replicates. The decreasing of total petroleum hydrocarbons (TPH) values occurred especially with S. saman and its symbiotic microorganisms on highest oil soil contamination. Despite the large growth of A. angustissima and M. caesalpiniifolia on the highest level of oil, these species and its inoculated microorganisms did not reduce the soil TPH. Both plants were hydrocarbon tolerant but not able to remediate the polluted soil. In contrast were significative hydrocarbon decrease with M. artemisiana under high oil concentrations, but plant growth was severely affected. Results suggest that the ability of the plants to decrease the soil concentration of TPH is not directly related to its growth and adaptation to conditions of contamination, but the success of the association between plants and its symbionts that seem to play a critical role on remediation efficiency.  相似文献

Understanding forest regeneration at sites previously used for agriculture underlies the establishment of science-based woodlands management policies. This is especially relevant in semi-arid areas, where the tree cover is critical in ameliorating the effects of aridity and in preventing desertification and land degradation. Natural regeneration in semi-arid areas occurs very slowly, which in part explains why it has hardly been studied. In the present work, we sought to devise a method to predict the natural regeneration potential of woodlands in semi-arid areas, to be used in guiding restoration efforts. Specifically, we evaluated holm oak coverage at a long-term ecological research site and then designed and validated a model to predict the natural regeneration of holm oak based on a few environmental variables. Unlike available studies, we obtained long-term information on tree regeneration (making use of >60 years of aerial photography) and climate (using long-term climate and microclimate data). We found that microclimate, measured using the potential solar radiation as a proxy, was a key driver of natural regeneration: after 60 years of agricultural abandonment, less sun-exposed areas attained a tree cover >90 %, whereas in more sun-exposed areas it remained below 20 %. We then used the model to map the natural regeneration potential, first in the study area and then in an area where holm oak plantations had been unsuccessfully introduced. In the latter case, the model successfully predicted the failure of this reforestation effort. Our results support the use of this model by decision makers to optimize management practices, as it will encourage the concentration of efforts in areas more prone to successful reforestation and allow the identification of areas more likely to benefit from natural regeneration processes.  相似文献

A wide spectrum of welfare issues is encountered on working equids. The objectives of this study were: (i) to determine the prevalence of cutaneous lesions in working equids in tropical regions of the North-Central of Veracruz State, Mexico, (ii) to identify the most prevalent cutaneous pathological conditions in this population of working equids, and (iii) to identify risk factors associated to the presence of work-related skin wounds. A number of 467 working equids presented for treatment at Donkey Sanctuary Mobile Clinic Program - National Autonomous University of Mexico from seven different villages in the North-Central region of Veracruz State were evaluated. Animals with signs of cutaneous pathology underwent a physical examination and samples were obtained of exudates, hair, and tissue for cytology examination, fungal cultures, and dermo-histopathology examination. The overall prevalence of cutaneous pathological conditions in working equids was 20.6 %. The prevalence per species was 22.6 % for horses, 18.2 % for donkeys, and 14.3 % for mules. The most common cause of skin lesions was trauma (abrasions, lacerations, and abscesses) followed in frequency by parasitic dermatitis and sarcoids. No strong associations among species were observed (P?>?0.20). Multivariate analysis showed that there was greater association between BCS and age with the presence of skin wounds (P?<?0.05). Equids who's BCS was rated as “moderate” as well as adult animals showed significantly more skin wounds compared with the young and the elderly (P?<?0.05). These findings can help to develop a better management practices to improve the welfare of equids in tropical regions of Veracruz, Mexico.  相似文献

The objective of this study was to determine the effect of dietary inclusion of benzoic acid on utilization of the macrominerals Ca, P, Mg, K, Na, and Cl in growing pigs. Eighteen barrows, initial BW of 28 ± 1.7 kg, were assigned to 3 diets: a basal diet based on barley, wheat, soybean meal, corn, and field pea and formulated to contain 9.31 MJ NE kg− 1 and 8.84 g− 1 kg standardized ileal digestible lysine, or the basal diet containing 10 or 20 g kg− 1 benzoic acid by replacing tapioca starch. The pigs were fed the experimental diets a rate of 2.7 times the maintenance requirement for ME for 21 days. Faeces and urine were collected quantitatively from days 11 to 21, and blood and plasma was collected on days 1, 10, and 21. On day 21, the pigs were killed and the left femur was removed. Benzoic acid linearly decreased (P = 0.001) the urine pH from 7.32 to 5.32, and quadratically increased (P < 0.05) blood pH on day 21. Benzoic acid linearly increased (P < 0.05) the apparent total tract digestibility (ATTD) of Ca, P, and Na from 65 to 72%, 46 to 55%, and 78 to 90%, respectively, linearly decreased (P < 0.05) the ATTD of Cl from 94 to 93%, and did not affect the ATTD of Mg and K. Benzoic acid linearly increased (P < 0.05) the retention of Ca, P, and K from 58 to 67%, 46 to 54%, and 31 to 38%, respectively, linearly decreased (P < 0.05) the retention of Na and Cl from 57 to 48% and 75 to 44%, respectively, and did not affect retention of Mg. On day 21, benzoic acid linearly increased (P = 0.001) plasma P and quadratically increased (P < 0.05) plasma K or tended to increase (P = 0.05) plasma Na. Benzoic acid linearly reduced (P < 0.05) the concentration of ash in femur but not the amount of ash, reduced (P < 0.05) the concentration of Ca and Cl in femur ash, and linearly increased (P < 0.05) the concentration of P in femur ash. In summary, benzoic acid increased the utilization of dietary Ca, P, and K, did not affect the utilization of dietary Mg, and reduced the utilization of dietary Na and Cl. During swine feed formulation, effects of benzoic acid on macromineral utilization should be considered to ensure that macromineral requirements are met and not exceeded following benzoic acid supplementation.  相似文献
