Female mutant restricted ovulator (RO) chickens of the White Leghorn strain carry a naturally occurring single nucleotide mutation in the very low density lipoprotein receptor (VLDLR) gene. Due to this mutation, RO hens fail to express a functional VLDLR protein on the oocyte membrane, which results in an impaired uptake of circulating yolk precursor macromolecules. Mutant RO hens subsequently develop hyperlipidemia and generally fail to lay eggs due to follicular atresia. Since RO hens also reportedly have three-fold higher basal plasma estrogen concentrations, combined with four-fold lower levels of circulating progesterone as compared to wild-type (WT) hens, we hypothesized that RO hens would have an increased abundance of pituitary progesterone receptor (PR) mRNA and PR isoforms A and B as well as alterations in circulating gonadotrophin levels. The Waxy (Wx) gene in hexaploid wheat (Triticum aestivum L.) encodes granule‐bound starch synthase (GBSS1), which is involved in the synthesis of amylose, a mostly linear glucan polymer that makes up ∼25% of wheat starch. A null mutation of the Wx gene in each of the three genomes is associated with starch almost entirely consisting of the branched glucan polymer amylopectin (waxy starch), with corresponding changes in functionality. However, the rheological behavior of partially waxy starch remains unclear. The objective of this study was to characterize flour and baking quality in 16 near‐isogenic lines, null at the Wx locus on zero, one, two, or all three genomes, grown in four different environments. Across allelic groups, significant variations in amylose concentrations, flour paste viscosity, loaf structure and texture, dough stability, and proximate variables were observed. Because waxy wheat starch has greater water absorbance and resistance to retrogradation than normal starch, its inclusion in flour blends has been suggested as a means of improving the texture and appearance of bakery products and noodles. The results indicate that wheat encoding <3 functional homeologs of GBSS1 produces starch that has potential in the production of certain food items, such as Asian noodles. The interaction of antiperoxidative flavonoids artocarpin (AR), cycloartocarpin (CAR), dalspinin (DP), dalspinosin (DPO), and dalspinin-7-O-galactoside (DPG) with 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation (ABTS(.+)) and O2(-.) was studied in phosphate buffer (pH 7.4). The ability of these compounds to inhibit lipid peroxidation and DNA scission was also investigated. The radical scavenging efficiency of flavonoids is demonstrated by the reduction of nitrogen-centered radical cation (ABTS(.+)). The reduction of ABTS(.+) follows the order quercetin > morin > Trolox > AR > DPO > CAR > DP. Inhibition of lipid peroxidation was studied by following Mb(IV) reduction, induced by lipid, arachidonic acid. These results are compared with those obtained for well-known antioxidants such as quercetin, morin, and Trolox. The structure-activity relationships between chemical structures of the flavonoids and their radical scavenging activities are anlayzed. The scavenging of O2(-.), inhibition of lipid peroxidation, and DNA damage depend on the oxidation potential of the flavonoids. The possible mechanism for radical scavenging activities of flavonoids in relation to their structure is also outlined. 相似文献

A study was conducted to investigate the population dynamics of four important coccids viz.,Aonidiella orientalis(Newstead),Ceroplastes actiniformis Green,Cardiococcus bivalvata(Green) and Parasaissetia nigra(Nietner) infesting sandal in Bangalore,India.Meteorological data viz.,monthly mean maximum and minimum temperatures,morning and evening relative humidity and total rainfall were also collected during the experimental period for statistical analysis to ascertain their influence on the population of coccids.The results show that all the four coccids are infesting sandal throughout the year.Maximum temperature exhibited significantly negative correlation with the population dynamics of A.orientalis,while other weather parameters did not show much influence on its incidence.In case of C.bivalvata,minimum temperature and morning relative humidity exhibited significant positive correlation with its population dynamics while other weather parameters had less significant effect on its population dynamics.For C.actiniformis and P.nigra,none of the weather factors seem to have influence on their incidences. 相似文献

The alpha subunits of Gs and Gi link different sets of hormone receptors to stimulation and inhibition, respectively, of adenylyl cyclase. A chimeric alpha i/alpha s cDNA was constructed that encodes a polypeptide composed of the amino terminal 60% of an alpha i chain and the carboxyl terminal 40% of alpha s. The cDNA was introduced via a retroviral vector into S49 cyc- cells, which lack endogenous alpha s. Although less than half of the hybrid alpha chain is derived from alpha s, its ability to mediate beta-adrenoceptor stimulation of adenylyl cyclase matched that of the normal alpha s polypeptide expressed from the same retroviral vector in cyc- cells. This result indicates that carboxyl terminal amino acid sequences of alpha s contain the structural features that are required for specificity of interactions with the effector enzyme, adenylyl cyclase, as well as with the hormone receptor. 相似文献

To investigate whether a particular receptor subtype can be coupled to multiple effector systems, recombinant M2 muscarinic receptors were expressed in cells lacking endogenous receptor. The muscarinic agonist carbachol both inhibited adenylyl cyclase and stimulated phosphoinositide hydrolysis. The stimulation of phosphoinositide hydrolysis was significantly less efficient and more dependent on receptor levels than the inhibition of adenylyl cyclase. Both responses were mediated by guanine nucleotide binding proteins, as evidenced by their inhibition by pertussis toxin; the more efficiently coupled adenylyl cyclase response was significantly more sensitive. Thus, individual subtypes of a given receptor are capable of regulating multiple effector pathways. 相似文献

Protein microarrays provide a powerful tool for the study of protein function. However, they are not widely used, in part because of the challenges in producing proteins to spot on the arrays. We generated protein microarrays by printing complementary DNAs onto glass slides and then translating target proteins with mammalian reticulocyte lysate. Epitope tags fused to the proteins allowed them to be immobilized in situ. This obviated the need to purify proteins, avoided protein stability problems during storage, and captured sufficient protein for functional studies. We used the technology to map pairwise interactions among 29 human DNA replication initiation proteins, recapitulate the regulation of Cdt1 binding to select replication proteins, and map its geminin-binding domain. 相似文献
