Theileria equi (T. equi) is an obligate intra- and extra-erythrocytic parasite that causes equine theileriosis (ET) in equids. Equine theileriosis is considered a notifiable disease of global significance, a major constraint to the international movement of horses, and endemic in many countries. This disease may be difficult to diagnose, as it can produce variable and nonspecific clinical signs. A cross-sectional study was designed for the molecular characterisation of T. equi and to investigate the associated risk factors of ET accompanied by its consequences on haematological and sero-biochemical parameters. A convenience sampling of 500 blood samples were collected from ET suspect horses from January to December 2017. PCR was performed on all blood samples targeting the 18S rRNA gene of T. equi followed by sequencing; 9% animals tested positive with confirmed sequences. The isolates of this study showed high homology with Cuban, Russian and Brazilian isolates of T. equi (accession numbers KY111762.2 , MG551915.1 and KY952237.1 , respectively). Based on multivariate analysis, the principal risk factors consisted of absence of dogs on the premises and presence of tick infestation. The haemato-biochemical parameters showed a decrease in granulocytes and erythrocytes, and an increase in lymphocytes, monocytes, mean corpuscular volume, mean corpuscular haemoglobin, mean platelet volume, glucose, phosphorus and aspartate aminotransferase in positive horses. This is the first study which identified ET in Punjab (Pakistan) using molecular techniques and risk factors together with the haemato-biochemical variations in horses.  相似文献

The spike 1 (S1) surface glycoprotein of infectious bronchitis virus (IBV) is the major inducer of the generation of virus neutralizing antibodies, and the administration of purified S1 has been shown to elicit a protective immune response against virulent virus challenge. On the basis of these observations, recombinant fowl poxvirus (rFPV) containing a cDNA copy of the S1 gene of IBV Mass 41 (rFPV-S1) was constructed and its immunogenicity and vaccine potential were evaluated. Initially, rFPV-S1 was shown to express the S1 in vito by indirect immunofluorescence staining and western blot analyses. Later, in vivo expression was demonstrated by the detection of IBV-specific serum immunoglobulin G and neutralization antibodies in the sera of chickens immunized with rFPV-S1. That the recombinant virus elicited anti-IBV protective immunity was indicated by the manifested, relatively mild clinical signs of disease, decreased titers of recovered challenge virus, and less severe histologic changes of the tracheas in virulent IBV Mass 41-challenged chickens previously receiving rFPV-S1 as compared with parental fowl poxvirus (FPV)-vaccinated control birds. In contrast, chickens immunized with either recombinant or parental FPV were resistant to a subsequent virulent FPV challenge. As to a preferred method of immunization, wing web administration appeared to be superior to the subcutaneous route because a greater percentage of birds vaccinated by the former protocol exhibited an anti-IBV humoral immune response. Thus, rFPV-S1 has potential as a poultry vaccine against both fowl pox and infectious bronchitis.  相似文献

BACKGROUND: Application of insecticides in modern agriculture in order to enhance legume production has led to their accumulation in soils to levels that adversely affect soil microflora such as rhizobia and exert a negative impact on the physiological activities associated with them. This study was therefore designed to identify rhizobial strains expressing higher tolerance to insecticides fipronil and pyriproxyfen and synthesising plant growth regulators even amid insecticide stress. RESULTS: The fipronil‐ and pyriproxyfen‐tolerant Rhizobium sp. strain MRL3 produced plant‐growth‐promoting substances in substantial amounts, both in the presence and in the absence of the insecticides. In general, both insecticides at recommended and higher rates reduced plant dry biomass, symbiotic properties, nutrient uptake and seed yield of lentil plants. Interestingly, when applied with any concentration of the two insecticides, Rhizobium sp. strain MRL3 significantly increased the measured parameters compared with plants grown in soils treated solely with the same concentration of each insecticide but without inoculant. CONCLUSION: This study suggests that Rhizobium strain MRL3 may be exploited as a bioinoculant to augment the efficiency of lentil exposed to insecticide‐stressed soils. Endocannabinoids are fatty acid amides (FAE; oleoylethanolamide and anandamide) which have orexigenic, anorexigenic or anti‐inflammatory properties. We examined mRNA expression via qPCR of endocannabinoid receptors (CNR1 and CNR2), enzymes that synthesize FAE (HRASLS5 and N‐acyl phosphatidylethanolamine phospholipase D), enzymes that degrade FAE [fatty acid amide hydrolase (FAAH), N‐acylethanolamine acid amidase (NAAA) and monoglyceride lipase (MGLL)], and the hormone precursor proopiomelanocortin (POMC) in liver at ?14, 7, 14 and 30 days around parturition from cows fed with a control (CON; NEL = 1.34 Mcal/kg) or moderate‐energy (OVER; NEL = 1.62 Mcal/kg) diet during the dry period. Expression of CNR2 and POMC was greater at 7 days in cows fed with OVER because of a decrease in expression between ?14 and 7 days in cows fed with CON. Cows fed with CON had an increase in expression of FAAH, HRASLS5, NAA, MGLL and POMC between 7 and 14 days; for FAAH and HRASLS5, such response led to greater expression at 14 days vs. cows fed with OVER. Cows fed with OVER vs. CON had a approximately twofold increase in expression of MGLL between ?14 and 7 days followed by a gradual decrease through 30 days at which point expression was still greater in OVER vs. CON. FAAH, MGLL and HRASLS5 were the most abundant genes measured. Expression of the hepatic endocannabinoid system and POMC was altered by plane of dietary energy prepartum particularly during the first 2‐week postpartum. Such responses may play a role in the physiological adaptations to the onset of lactation, including energy balance and feed intake.  相似文献

中国-巴基斯坦喀喇昆仑公路(以下简称"中巴公路")是中国政府于20世纪60年代中期援建的一条连接中国西部城市喀什和巴基斯坦北部城市塔科特的国际公路。2009年4月、7月、9月和2010年6~7月对中巴公路沿线分布的动物进行了考察,经全面系统分类、整理和统计,该地区共有陆栖野生动物22目51科105属147种,其中爬行类1目3科5属8种,鸟类14目34科72属103种,哺乳类/兽类7目14科28属36种。从野生动物分布的种类数量上看,分布最少的是爬行类(8种),而较多的是鸟类(103种)和哺乳类/兽类(36种),分别占该地区分布野生脊椎动物种数的5.44%、70.07%和24.49%。野生动物区系以北方广布种、高地型、古北型和中亚型为主,全北型、东洋型和喜马拉雅-横断山型渗透混杂,表明古北界温带陆栖动物地理属性是该地区动物区系的基本特征之所在。  相似文献

Induced resistance was studied in three sorghum genotypes (IS2205, ICSV1 and ICSV700) against Chilo partellus (Swinhoe) (Lepidoptera; Pyralidae) infestation and jasmonic acid (JA) and salicylic acid (SA) application. The activity of plant defensive enzymes [peroxidase (POD), polyphenol oxidase (PPO), superoxide dismutase (SOD), and catalase (CAT)], and the amounts of total phenols, hydrogen peroxide (H2O2), malondialdehyde (MDA), and proteins were recorded at 6 days after infestation. The induction of enzyme activities and the amounts of secondary metabolites varied among the genotypes and treatments. The genotype IS2205 showed a stronger effect than that of ICSV1 or ICSV 700. Treatment with JA followed by insect infestation induced greater levels of enzymes and secondary metabolites. The results suggest that JA induces greater levels of resistance components in sorghum plants against insect pests. Thus, pretreatment of plants with elicitors including JA and SA could provide a greater opportunity for plant defense against herbivores.  相似文献

Field experiments were conducted to study soil properties, soil enzymes activities, water use efficiency (WUE) and crop productivity after six years of soya bean straw mulching in the semi‐arid conditions of China. The experiment included four treatments: CK (Control), N (240 kg N ha‐1), H (soya bean straw mulching at half rate 700 kg ha‐1 with 240 kg N ha‐1) and F (soya bean straw mulching at full rate 1,400 kg ha‐1 with 240 kg N ha‐1). Soil organic carbon (SOC), soil labile organic carbon (LOC), soil available N (AN), available P (AP) and enzyme activities were analysed after wheat harvesting in 2016 and 2017. Results show that straw amounts had positive effects on the soil fertility indices being higher for treatment F. The SOC, LOC, AN, AP and enzyme activities (i.e. saccharase, urease and alkaline phosphatase) were in the order of F > H > N > CK. High wheat grain yield and WUE were observed for F treatment. A total of six years mulching along with 240 kg ha‐1 nitrogen fertilizer application is sufficient for wheat yield stability and improving soil properties except urease activities in the semi‐arid condition of China. However, the straw mulching amount should be further studied with minimum nitrogen fertilizer for an environment‐friendly and effective approach for improving the soil biological properties with adequate crop production on a sustainable basis in the semi‐arid region of China.  相似文献

Long‐whiskered catfish, Sperata aor (Hamilton 1822), is commercially important in food, ornamental and sport fisheries. The fish is mainly caught from the wild populations because its aquaculture practices are not commercialised. Inland fishery in the Ganga basin is mostly unorganised; hence, no published report is available on the trend of S. aor production from the selected habitat. In India, S. aor has been categorised vulnerable mainly due to natural and anthropogenic threats. Otolith chemistry shows variation with changing physico‐chemical conditions of the fish habitat. Therefore, the present study was conducted with the objective to analyse spatio‐temporal variations in water chemistry in relation to environmental factors; relationship between water and otolith chemistry; and spatio‐temporal variations in otolith chemistry to discriminate the stocks of S. aor inhabiting the River Ganga. Most of the element: Ca ratios in water samples did not show significant correlations with environmental factors, viz. temperature and conductivity. Only few element: Ca concentrations in otoliths were positively correlated to their corresponding ratios in the ambient water. In the selected study area, the S. aor populations were discriminated into four stocks possibly because of heterogeneous water chemistry at the sampling sites, and physical barriers. In the present study, otolith chemistry showed relatively low temporal variability as compared to spatial variability; thus, the classification accuracy of individuals to their original populations remained consistent over the selected time period. The findings could be useful in devising scientifically sound management strategies and/or any conservation plans for the vulnerable S. aor populations inhabiting the River Ganga.  相似文献

Global temperature is rising because of increasing concentrations of CO2 and other greenhouse gases (e.g. methane, nitrous oxide, etc.) in the atmosphere because of the excessive use of fossil fuels. High temperature causes heat stress which reduces crop productivity. The development of heat-tolerant varieties is feasible and will help mitigate the effects of climate change. Fifty-one cotton accessions were screened in heat-stressed and non-stressed conditions in the glasshouse and field. Relative cell injury percentage (RCI %), a measure of cell membrane thermostability (CMT), was used to assess heat tolerance. Heat-tolerant accessions had more stable yield and yielded more seed cotton with better quality fibre than the heat-intolerant accessions across four environments. The responses of the 51 accessions to all four environments for the measured traits were strongly associated. RCI % of the accessions and hybrids was strongly and negatively associated with yield and fibre traits. CMT was concluded to be a useful technique for identification of heat-tolerant cotton.  相似文献

The methanol extracts of Picrasma javanica, leaves, seeds, stem and root barks were partitioned (petrol, dichloromethane, ethyl acetate, butanol). All obtained extracts and fractions showed a broad spectrum of antibacterial activity, while none was active against the tested moulds.  相似文献
