A sample of fourteen horse rugs and two saddle blankets stored in south western Sydney, Australia, an area of known high dust mite prevalence in the human environment, were analysed for the presence of house dust mites. Dust samples from the rugs, blankets and 16 control sites were collected using a vacuum cleaner with a modified attachment and filter. Dust mites were extracted using an adapted floatation technique. Eight rugs and all control samples were positive for mites, which were confirmed to be house dust mites of the genus Dermatophagoides. This study confirms that exposure to house dust mites from horse rugs can occur, indicating that house dust mite allergen reactivity on intradermal and serum allergy testing in atopic horses may represent true dust mite hypersensitivity. Nevertheless, quantification studies will be necessary to ensure that there is adequate mite exposure for development of hypersensitivity, and further evaluation of immunological responses, avoidance and provocation, and specific immunotherapy are required to confirm the clinical relevance. SummaryRadish (Raphanus sativus) is an important crucifer vegetative crop in Japan. We studied the morphological and anatomical characteristics of radish roots over a 6-week period using three radish cultivars with three different root shapes: long-type (Lt), round-type (Rt), and thin-type (Tt). We found that significant differences in the thickness of the roots first appeared 4 weeks after sowing. In Lt and Rt plants, the roots thickened rapidly, especially in the middle section that is composed of the lower part of the hypocotyl and the upper part of the taproot. The taproot increased in length in Lt plants, but failed to elongate in Rt plants. In Tt plants, the roots showed much smaller increases in mean diameter and length. Transverse sections through the taproots showed significant differences, both in cell number and in cell size 4 weeks after sowing. Lt and Rt plants contained more cells in the secondary xylem and phloem than Tt plants, and cell expansion was also greater in the storage roots of the Lt and Rt cultivars. These results will contribute to a better understanding of the development of radish roots of different shape. 相似文献

Summary Image analysis gave better precision than colorimetry in measuring the darkening caused by excess free sugar during crisping
of potatoes. The image analyser was used to measure individual crisp Average Optical Density (AOD) values in order to examine
the variability of AOD within and between tubers. It is unnecessary to measure more than one crisp per tuber when assessing
breeding lines, although two crisps per tuber are recommended for quality control. Crisps from at least ten tubers should
be measured.
A ‘Quick Crisp’ method, in which single crisps from each of four tubers are judged against standard samples, is reported.
Regression of results from this method on crisp reflectance percentage (Agtron score) gaveR
2 of 0.967. However, the observer tended to overestimate at low values and underestimate at high values. Over several trials,
the crossover point varied from 15 to 50 Agtron score units.

The physiological effects of the hydrolysates of white rice protein (WRP), brown rice protein (BRP), and soy protein (SP) hydrolyzed by the food grade enzyme, alcalase2.4 L, were compared to the original protein source. Male Syrian Golden hamsters were fed high-fat diets containing either 20% casein (control) or 20% extracted proteins or their hydrolysates as the protein source for 3 weeks. The brown rice protein hydrolysate (BRPH) diet group reduced weight gain 76% compared with the control. Animals fed the BRPH supplemented diet also had lower final body weight, liver weight, very low density lipoprotein cholesterol (VLDL-C), and liver cholesterol, and higher fecal fat and bile acid excretion than the control. Expression levels of hepatic genes for lipid oxidation, PPARα, ACOX1, and CPT1, were highest for hamsters fed the BRPH supplemented diet. Expression of CYP7A1, the gene regulating bile acid synthesis, was higher in all test groups. Expression of CYP51, a gene coding for an enzyme involved in cholesterol synthesis, was highest in the BRPH diet group. In order to confirm a microscopic diagnosis of 'eperythrozoonosis' made over 40 years ago in a captive owl monkey (Aotus trivirgatus), DNA was extracted from archived fixed and stained blood smears and subjected to generic haemotropic mycoplasma (haemoplasma) quantitative real-time PCR (qPCR) and a human glyceraldehyde-3-phosphate dehydrogenase qPCR as an amplification control. The qPCRs confirmed the extraction of host DNA from the samples and the presence of a haemoplasma species. Partial 16S rRNA and ribonuclease P ribosomal gene fragments were amplified by PCR, cloned and sequenced. Sequence data and phylogeny showed the owl monkey haemoplasma to lie in the haemominutum clade of haemoplasmas, most closely related to 'Candidatus Mycoplasma kahaneii'. This study confirms the use of generic haemoplasma qPCRs to successfully amplify haemoplasma DNA from fixed, stained and archived blood smears from the early 1970s and provides molecular confirmation of the existence of a novel haemoplasma species in an owl monkey, for which the name 'Candidatus Mycoplasma aoti' sp. nov. is proposed. 相似文献

Preliminary screening of maize (Zea mays L.) genotypes for aluminum (Al) tolerance in nutrient solutions over a 12‐day growth period showed greater plant‐induced pH changes in solutions without Al than in solutions containing Al. Such pH changes may alter the specific effect of Al on relative root length (length in Al‐containing solution/length in 0 Al solution) commonly used as an index to rank genotypes with respect to Al tolerance. The objective of this study was to examine several screening methods for identifying Al‐tolerant maize genotypes, and to identify those procedures which resulted in minimal pH fluctuations during the course of screening. The following methods of controlling pH in nutrient solutions were compared: (i) 12‐day exposure to 0 or 5 mg Al/L in nutrient solutions (a) with or (b) without daily pH adjustment or (c) with different NO3 ‐/NH4 + ratios, and (ii) 2‐day exposure to 0, 5, 10, 25 or 40 mg Al/L treatment solutions followed by a 3‐day recovery period in solutions with an initial pH at (a) 4.6 or (b) 4.0. In the 12‐day experiments, daily pH adjustment to 4.6 did not eliminate large pH fluctuations in the control (0 Al) solutions, and it substantially decreased the soluble Al concentration in the Al‐treatment solution. Varying the ratio of NO3 ‐ to NH4 + did not eliminate large pH fluctuations. Exposing the seedlings for 2 days to Al solutions at pH 4.6 resulted in large pH differences between 0 Al and Al‐containing solutions and in precipitation of large amounts of Al. In contrast, the 2‐day procedure using solutions with an initial pH at 4.0 was more satisfactory in that the pH was maintained between 4.0 and 3.7 in all solutions, and Al precipitation was minimized. When the 2‐day method at pH 4.0 was used to screen the genotypes, PDMR3 had consistently higher relative root lengths in Al‐containing solutions than did Kalimpos, IPB Varl, UPCA Varl and Trinidad Grp1&2. 相似文献
