Late blight, caused by the oomycete Phytophthora infestans, is considered the most important and destructive disease of potato in Turkey. In this study, characterization of 367 isolates of P. infestans obtained from the potato-growing areas of the country was carried out to evaluate the pathogen population structure over the 2017–2019 production seasons. The isolates were characterized by numerous features including mating type, in vitro mefenoxam sensitivity, simple-sequence repeat (SSR) markers, and virulence against a set of potato differential lines. Most isolates were A2 mating type (353 isolates). Also, 68% of isolates were resistant to mefenoxam; the remainder were intermediate in their sensitivity and there were no sensitive isolates. SSR-based genotypic analysis of P. infestans populations showed a low genetic diversity. The 13_A2 clonal lineage predominated with a frequency of 92.1%, followed by 34_A1 (3.3%) and 37_A2 (2.7%). Genotypes 34_A1 and 37_A2 were detected only in 2019. This is the first report of 34_A1 and 37_A2 clonal lineages causing late blight disease of potato in Turkey. The most abundant virulence type was one overcoming resistance genes R1, R2, R3, R4, R6, R7, R10, and R11. These results emphasized that the migration of individuals and the asexual generation of subclonal differences were the main factors driving the population structure of P. infestans in Turkey.  相似文献

Sixty Nicotiana species were examined for tolerance against various osmotica for seed germination and seedling growth in vitro. The species showed a wide variety of tolerance, and based on the results of the in vitro tests, 31 species were selected and further evaluated for salt and drought tolerance in a glasshouse. The degrees of tolerance of germination among the 57 species toward NaCl were approximately related to those toward mannitol, indicating that the osmolarity plays a majorrole in seed germination. However, the responses during the seedling growth differed in NaCl and mannitol or drought, and there was no correlation between salt and drought tolerance. Based on the responses in vitro and in the glasshouse, N. paniculata and N. excelsior were selected as the salt tolerant species, and N. arentsii as the salt sensitive species. The degrees of accumulation of dry matter and of Na+ in the leaves were different in the two tolerant species; during NaCl treatment, N. paniculata and N. arentsii accumulated less dry matter relative to the control plants than N. excelsior, and N. paniculata accumulated more Na+ in its leaves than N. excelsior and N. arentsii. It is assumed that the two salt tolerant species have different mechanisms for tolerance to the salt. Three groups of beef cow and calf pairs were studied to determine plasma vitamin E and blood selenium (Se) concentrations of calves at 1 month old. Group 1 was managed on irrigated pasture and calves received no Se/vitamin E injections at birth. Group 2 was managed on irrigated pasture, and the calves were injected with Se/vitamin E at birth. Group 3 was managed on dry foothill grasslands, and these cows were supplemented with 56.3 mg vitamin E and 3 mg Se daily, and the calves received a Se/vitamin E injection at birth. The plasma concentration of vitamin E in group 1 and 2 cows (9.5 +/- 1.24 and 8.43 +/- 1.0 microg/ml, respectively) was significantly higher than that of the group 3 cows (2.28 +/- 0.42 microg/ml; P < 0.05). The blood Se concentrations in group 3 cows (169 +/- 37 ng/ml) were significantly higher than those in group 1 and 2 cows (36.4 +/- 15.9 and 31.1 +/- 12.5 ng/ml, respectively; P < 0.05). Calf Se was highly correlated to cow Se (r = 0.965), and calf vitamin E was moderately correlated to cow vitamin E (r = 0.605). Calf vitamin E concentrations were consistently lower than cow vitamin E concentrations, and many values would be considered deficient.  相似文献

We investigated wheat (Triticum aestivum) grain quality under Free Air CO2 Enrichment (FACE) of 550 ± 10% CO2 μmol mol−1. In each of two full growing seasons (2008 and 2009), two times of sowing were compared, with late sowing designed to mimic high temperature during grain filling. Grain samples were subjected to a range of physical, nutritional and rheological quality assessments. Elevated CO2 increased thousand grain weight (8%) and grain diameter (5%). Flour protein concentration was reduced by 11% at e[CO2], with the highest reduction being observed at the late time of sowing in 2009, (15%). Most of the grain mineral concentrations decreased under e[CO2] - Ca (11%), Mg (7%), P (11%) and S (7%), Fe (10%), Zn (17%), Na (19%), while total uptake of these nutrients per unit ground area increased. Rheological properties of the flour were altered by e[CO2] and bread volume reduced by 7%. Phytate concentration in grains tended to decrease (17%) at e[CO2] while grain fructan concentration remained unchanged. The data suggest that rising atmospheric [CO2] will reduce the nutritional and rheological quality of wheat grain, but at high temperature, e[CO2] effects may be moderated. Reduced phytate concentrations at e[CO2] may improve bioavailability of Fe and Zn in wheat grain.  相似文献

Salmonella Kentucky is among the most frequently isolated S. enterica serovars from food animals in the United States. Recent research on isolates recovered from these animals suggests there may be geographic and host specificity signatures associated with S. Kentucky strains. However, the sources and genomic features of human clinical S. Kentucky isolated in the United States remain poorly described. To investigate the characteristics of clinical S. Kentucky and the possible sources of these infections, the genomes of all S. Kentucky isolates recovered from human clinical cases in the State of Maryland between 2011 and 2015 (n = 12) were sequenced and compared to a database of 525 previously sequenced S. Kentucky genomes representing 12 sequence types (ST) collected from multiple sources on several continents. Of the 12 human clinical S. Kentucky isolates from Maryland, nine were ST198, two were ST152, and one was ST314. Forty‐one per cent of isolates were recovered from patients reporting recent international travel and 58% of isolates encoded genomic characteristics similar to those originating outside of the United States. Of the five isolates not associated with international travel, three encoded antibiotic resistance genes conferring resistance to tetracycline or aminoglycosides, while two others only encoded the cryptic aac(6′)‐Iaa gene. Five isolates recovered from individuals with international travel histories (ST198) and two for which travel was not recorded (ST198) encoded genes conferring resistance to between 4 and 7 classes of antibiotics. Seven ST198 genomes encoded the Salmonella Genomic Island 1 and substitutions in the gyrA and parC genes known to confer resistance to ciprofloxacin. Case report data on food consumption and travel were, for the most part, consistent with the inferred S. Kentucky phylogeny. Results of this study indicate that the majority of S. Kentucky infections in Maryland are caused by ST198 which may originate outside of North America.  相似文献

Fasted Brazilian squirrel monkeys (BrSMs) exhibited slightly higher serum bilirubin levels (0.30±0.05 mg/dl) than others in the fed state (0.13±0.01). The mean liver weight was 50% lower following a 22 h fast. The rate of bile flow was unaffected by fasting and averaged 13.8 l/min/kg and 47.5 l/min/100g liver in six BrSMs. No significant difference in mean bilirubin excretion/min was observed on a body weight basis following fasting. When the mean rate of bilirubin excretion was calculated as a function of liver weight, a two-fold higher rate was present in fasted monkeys, but only at the p=0.06 level of statistical significance. From data collected in this and earlier studies, it would appear that BrSMs represent the best animals studied to date to serve as experimental controls in comparative studies with Bovilian squirrel monkeys which exhibit a Gilbert-like syndrome.  相似文献

Three Portuguese water dog siblings, all females aged 5 to 7 months, were killed following a brief period of neurologic disease. Tissues were processed for light and electron microscopy and for biochemical analyses. All pups had membranous cytoplasmic inclusions in neurons throughout the brain and spinal cord. Cytoplasmic vacuoles were present in cells of many organs outside the nervous system. GM1 ganglioside in brain was markedly elevated in all three dogs, and beta-galactosidase activity was less than 10% of control values. These findings are similar to those in GM1 gangliosidosis of man and animals although the number of organs and tissues containing vacuolated cells is greater.  相似文献

The brainstem auditory evoked response (BAER) was recorded from 7 unanesthetized and 27 methoxyflurane anesthetized dogs. A 0.1 msec, 70 dB stimulus delivered at 10 Hz evoked the expected seven wave BAER. Mean peak wave latencies and standard deviations were calculated. Differences were not found between neither right and left ears, nor male and female dogs. The anesthetized dogs had a significantly longer latency for all waves, except wave I, than the unanesthetized dogs. Use of the BAER as a diagnostic technique for brainstem lesions is recommended.  相似文献
