Previous studies characterizing the Theileria parva p67 gene in East Africa revealed two alleles. Cattle-derived isolates associated with East Coast fever (ECF) have a 129 bp deletion in the central region of the p67 gene (allele 1), compared to buffalo-derived isolates with no deletion (allele 2). In South Africa, Corridor disease outbreaks occur if there is contact between infected buffalo and susceptible cattle in the presence of vector ticks. Although ECF was introduced into South Africa in the early 20th century, it has been eradicated and it is thought that there has been no cattle to cattle transmission of T. parva since. The variable region of the p67 gene was amplified and the gene sequences analyzed to characterize South African T. parva parasites that occur in buffalo, in cattle from farms where Corridor disease outbreaks were diagnosed and in experimentally infected cattle. Four p67 alleles were identified, including alleles 1 and 2 previously detected in East African cattle and buffalo, respectively, as well as two novel alleles, one with a different 174 bp deletion (allele 3), the other with a similar sequence to allele 3 but with no deletion (allele 4). Sequence variants of allele 1 were obtained from field samples originating from both cattle and buffalo. Allele 1 was also obtained from a bovine that tested T. parva positive from a farm near Ladysmith in the KwaZulu-Natal Province. East Coast fever was not diagnosed on this farm, but the p67 sequence was identical to that of T. parva Muguga, an isolate that causes ECF in Kenya. Variants of allele 2 were obtained from all T. parva samples from both buffalo and cattle, except Lad 10 and Zam 5. Phylogenetic analysis revealed that alleles 3 and 4 are monophyletic and diverged early from the other alleles. These novel alleles were not identified from South African field samples collected from cattle; however allele 3, with a p67 sequence identical to those obtained in South African field samples from buffalo, was obtained from a Zambian field isolate of a naturally infected bovine diagnosed with ECF. The p67 genetic profiles appear to be more complex than previously thought and cannot be used to distinguish between cattle- and buffalo-derived T. parva isolates in South Africa. The significance of the different p67 alleles, particularly the novel variants, in the epidemiology of theileriosis in South Africa still needs to be determined.  相似文献

Confocal laser scanning microscopy (CLSM) is the method of choice to study interfacial biofilms and acquires time-resolved three-dimensional data of the biofilm structure. CLSM can be used in a multi-channel modus where the different channels map individual biofilm components. This communication presents a novel image quantification tool, PHLIP, for the quantitative analysis of large amounts of multichannel CLSM data in an automated way. DNA replication in eukaryotes requires nucleosome disruption ahead of the replication fork and reassembly behind. An unresolved issue concerns how histone dynamics are coordinated with fork progression to maintain chromosomal stability. Here, we characterize a complex in which the human histone chaperone Asf1 and MCM2-7, the putative replicative helicase, are connected through a histone H3-H4 bridge. Depletion of Asf1 by RNA interference impedes DNA unwinding at replication sites, and similar defects arise from overproduction of new histone H3-H4 that compromises Asf1 function. These data link Asf1 chaperone function, histone supply, and replicative unwinding of DNA in chromatin. We propose that Asf1, as a histone acceptor and donor, handles parental and new histones at the replication fork via an Asf1-(H3-H4)-MCM2-7 intermediate and thus provides a means to fine-tune replication fork progression and histone supply and demand.  相似文献

Salts were applied as aqueous solution to the outer surfaces of astomatous isolated cuticles, and the water was allowed to evaporate. Effects of salt residues on the surfaces of cuticles on water permeability of cuticles were measured at 25°C. A surface dose of 0.2 mol m–2 Cs2CO3 and K2CO3 increased the water permeability of pear leaf cuticles by factors of 9.9 and 3.9, respectively. Na2CO3 was barely effective, and Li2CO3 as well as (NH4)2CO3 had no influence on water permeability. Potassium applied as sulfate, nitrate, or chloride had no effect on the water permeability of pear leaf cuticles, while K+ salts of weak acids (0.2 mol m–2) were effective, as they increased water permeability by factors of 5.4 (K+‐acetate), 3.9 (K2CO3), and 2.0 (K2HPO4), respectively. Sensitivity of Idesia polycarpa leaf cuticles to treatment with K2CO3 at 0.2 mol m–2 was greatest as water permeability was increased 35‐fold, while the water permeability of cuticles from other species tested (astomatous leaf cuticles from Hedera helix, Citrus aurantium, Prunus laurocerasus, Pyrus communis, and Populus canescens; fruit cuticles from Capsicum annuum and Lycopersicon esculentum) increased only by factors ranging from 1.7 to 3.9. Data are discussed in relation to swelling and ion‐exchange properties of cuticles.  相似文献

Purpose

Occlusion of carbon (C) within phytoliths, biogenic silica deposited in plant tissues and returned to the soil, is an important mechanism for long-term terrestrial biogeochemical C sequestration and might play a significant role in mitigating climate change.

Materials and methods

Subtropical and tropical soil profiles (to 100 cm depth) developed on granite and basalt were sampled using a mass-balance approach to explore the influence of climate and lithology on soil phytolith-occluded carbon (PhytOC) accumulation.

Results and discussion

Soil PhytOC storage in the subtropics was significantly greater than in the tropics, with the soil profiles developed on granite storing greater PhytOC than soils derived on basalt. Phytolith and PhytOC content decreased with depth in all soil profiles. Phytolith content showed a positive correlation with the soil bio-available silicon in the soil profiles developed on basalt, while a negative correlation was observed in soil profiles developed on granite.

Conclusions

Climate and lithology have a significant impact on soil PhytOC sequestration. The management of forests (e.g., afforestation and reforestation) and external silicon amendments (e.g., basalt powder amendment) in soils, especially those developed on granite, have the potential to enhance PhytOC accumulation in forest ecosystems.

A combination of direct analysis in real time (DART) ionization coupled to time-of-flight mass spectrometry (TOFMS) and chemometrics was used for animal fat (lard and beef tallow) authentication. This novel instrumentation was employed for rapid profiling of triacylglycerols (TAGs) and polar compounds present in fat samples and their mixtures. Additionally, fat isolated from pork, beef, and pork/beef admixtures was analyzed. Mass spectral records were processed by principal component analysis (PCA) and stepwise linear discriminant analysis (LDA). DART-TOFMS profiles of TAGs were found to be more suitable for the purpose of discrimination among the examined fat types as compared to profiles of polar compounds. The LDA model developed using TAG data enabled not only reliable classification of samples representing neat fats but also detection of admixed lard and tallow at adulteration levels of 5 and 10% (w/w), respectively. The presented approach was also successfully applied to minced meat prepared from pork and beef with comparable fat content. Using the DART-TOFMS TAG profiles of fat isolated from meat mixtures, detection of 10% pork added to beef and vice versa was possible.  相似文献

Indonesian small-scale forest holders are facing a dilemma due to the implementation of a mandatory national timber-legality verification scheme and an internationally popular forest certification scheme. The problems arise from limited financial, technical, and administrative information concerning the most preferred scheme and the “imperfection of such a scheme” for long-term business needs. Using the Forest Certification Assessment Guide (FCAG) this paper identifies the characteristics of four third-party forest certification schemes currently working in Indonesia, namely FSC, PEFC, LEI, and SVLK. An online questionnaire was used to survey a wide range of respondents about the future development of contested schemes and those preferred by small-scale forest holders. Our findings show that although FSC scheme obtained the highest score and is considered the best scheme according to the FCAG, small-scale forest holders prefer SVLK scheme, which had the lowest FCAG score because of its mandatory nature and available subsidies. Statements by the four schemes’ proponents, which delegitimize other schemes, reveal they are in competition especially in winning market acceptance and local adaptability. Finally, we suggest proponents enhance aspects where their schemes are lacking and contrive a comparable certification scheme in order to induce willingness to be certified.  相似文献

Single-molecule fluorescence spectroscopy was used to determine the electronic properties of individual single-walled carbon nanotubes. Carbon nanotube structure was determined simultaneously from Raman spectroscopy. Fluorescence spectra from individual nanotubes with identical structures have different emission energies and linewidths that likely arise from defects or the local environment. Unlike most other molecules studied to date, the fluorescence intensity or spectrum from a single nanotube unexpectedly did not fluctuate.  相似文献

As part of a larger survey to map the geographical distribution of Babesia and Theileria parasites in the southern African rhinoceros population, white rhinoceroses were sampled during routine immobilisations in the Kruger National Park. Polymerase chain reaction (PCR) and reverse line blot (RLB) hybridisation assays were used to screen for the presence of piroplasms and complete blood counts were used to assess associated changes in clinical parameters. Of the 195 rhinoceroses sampled, 71 (36.4 %) tested positive for the presence of Theileria bicornis, with no significant change in the haematological parameters measured, while 18 (9.2 %) tested positive for Theileria equi. None of the rhinoceroses sampled tested positive for Babesia bicornis, a parasite associated with mortalities in black rhinoceroses.  相似文献
