In the present study, attempt was made to compare agar with gum karaya as gelling agent in micropropagation of rough lemon (Citrus jambhiri Lush.). Initially nodal segments were cultured on agar-gel MS medium containing benzyladenine (BA), kinetin (KN), zeatin (ZN) (1.0–2.5 mg L?1) and malt extract (200–1,200 mg L?1) to standardize the medium. Maximum shoot regeneration (66.66%) was observed with KN 2 mg L?1 with an average shoot length of 0.73 cm. Gum karaya and agar was then evaluated at different concentration and combinations in same medium. The shoot regeneration response on media gelled with 30 g L?1 gum karaya was 62.49% with an average shoot length of 0.80 cm. Regenerated shoots were rooted on MS medium gelled with agar and supplemented with different concentrations (0.5–2.5 mg L?1) of indole-3-acetic acid (IAA), naphthalene acetic acid (NAA), and indole-3-butyric acid (IBA). Maximum response (52.77 %) was observed with IBA 2.0 mg L?1 with an average number of 2.58 roots/shoot. A maximum of 53.47% cultures showed root regeneration with an average number of 2.91 roots/shoot in 30 g L?1 gum karaya-gel medium. Texture measurements revealed that firmness of gum karaya-gel medium was nowhere near to that of agar. However, in their capability of supporting growth and differentiation of explants they are equal to agar medium. Gum karaya forms less adhesive and gummy medium as compared to agar. This study indicates that gum karaya can be used as gelling agent in place of agar.  相似文献

Patchouli oil is one of the most important essential oils used in modern perfumery and cosmetic industries. There is hardly any preparation of oriental nature where patchouli oil is not used. It is used mainly because of fixative property as it gives tenacity to other perfumes. Field experiments were conducted at Bangalore, India, to study the influence of sources and doses of N and K on herbage, oil yield, nutrient uptake, nitrogen utilization efficiency and oil content of patchouli [Pogostemon cablin (Blanco) Benth.]. The results revealed that application of 200 kg N/ha and 41.5 kg K/ha produced significant higher patchouli herbage and oil yields compared with controls. Similarly, N and K uptake were also higher at 200 kg N and 41.5 kg K/(ha year) compared with controls. DCD-coated urea produced higher herbage, oil yield and N uptake and utilization efficiency compared with prilled urea. There was no effect of sources of K on the yield of patchouli. The oil content was not influenced by N, K doses or sources applied. N and K depletion were noticed in the soil.  相似文献

Vanilla is a large genus of about 110 species in the orchid family (Orchidaceae), including the species Vanilla planifolia from which commercial vanilla flavoring is derived. Since most species of vanilla are considered rare and endangered there is an urgent need to conserve them through genetic analysis and propagation/conservation studies on this crop.The present study investigated the genetic diversity among nine leafy- and leaf-less Vanilla species employing 30 decamer RAPD primers and 10 ISSR primers. The species under study were diverse and displayed a range of variability (0–66% and 0–81% for RAPD and ISSR, respectively). A total of 154 RAPD polymorphic markers (83.24%, h = 0.378) and 93 ISSR polymorphic markers (86.11%, h = 0.363) were used to generate a genetic similarity matrix followed by the cluster analysis. Specific groupings were revealed by each cluster analysis with slight variation between two different markers. Among the nine species studied, V. planifolia, Vanilla aphylla and Vanilla tahitensis revealed very low level of variation within their collections, thus indicating a narrow genetic base. The large genetic distance of Vanilla andamanica from other species suggests its different origin. A close genetic affinity was observed between the pairs V. planifolia, V. tahitensis and Vanilla albida, V. aphylla. These are the first comparative results for RAPD and ISSR reporting inter-relationship among nine cultivated, wild and hybrid Vanilla species.  相似文献

The feasibility of studying avian primordial germ cells in blood smears has been demonstrated. Blood smears prepared from chick embryos of stages 13 to 15 (48 to 55 hours) contained primordial germ cells, which were revealed by the periodic acidSchiff reaction. The presence of glycogen in the cytoplasm of the primordial germ cells facilitated their selective identification.  相似文献

Different forms of manganese (Mn) were investigated, including total, diethylenetriamine penta-acetic acid (DTPA) extractable, soil solution plus exchangeable (Mn), Mn adsorbed onto inorganic sites, Mn bound by organic sites, and Mn adsorbed onto oxide surfaces, from four soil taxonomic orders in northwestern India. The total Mn content was 200–950 mg kg?1, DTPA-extractable Mn content was 0.60–5.80 mg kg?1, soil solution plus exchangeable Mn content was 0.02–0.80 mg kg?1, Mn adsorbed onto inorganic sites was 2.46–90 mg kg?1, and Mc adsorbed onto oxide surfaces was 6.0–225.0 mg kg?1. Irrespective of the different fractions of Mn their content was generally greater in the fine-textured Alfisols and Inceptisols than in coarse-textured Entisols and Aridisols. The proportion of the Mn fractions extracted from the soil was in the order as follows: Adsorbed onto oxide surfaces > adsorbed onto inorganic site > organically bound > DTPA > soil solution + exchangeable. Based on coefficient of correlation, the soil solution plus exchangeable Mn, held onto organic site and oxide surface (amorphous) and DTPA-extractable Mn, increased with increase in organic carbon of the soil. The two forms, adsorbed onto inorganic site (crystalline) and DTPA extractable, along with organic carbon, increased with increase in clay content of the soil. DTPA-Mn and Mn adsorbed onto oxide surfaces and held on organic site decreased with increased with an increase in calcium carbonate and pH. Total Mn was strongly correlated with organic carbon and clay content of soil. Among the forms, Mn held on the organic site, water soluble + exchangeable and adsorbed onto oxide surface were positively correlated with DTPA-extractable Mn. DTPA-extractable Mn seems to be a good index of Mn availability in soils and this form is helpful for correction of Mn deficiency in the soils of the region. The uptake of Mn was greater in fine-textured Inceptisols and Alfisols than in coarse-textured Entisols and Aridisols. Among the different forms only DTPA-extractable Mn was positively correlated with total uptake of Mn. Among soil properties Mn uptake was only significantly affected by pH of the soil.  相似文献

Milled wood lignin samples from Loblolly pine stem wood, forest residue, and bark were isolated and characterized by quantitative (13)C and (31)P nuclear magnetic resonance (NMR), Fourier transform infrared spectroscopy (FT-IR), and gel permeation chromatography (GPC) for molecular weight determination. Results from (13)C NMR show the stem wood and forest residue samples have similar functional group contents. However, the bark has fewer methoxyl groups, β-O-4 structures, dibenzodioxocin, and side chains than the other two lignins. The bark lignin has the highest amounts of p-hydroxyphenyl (h) and C-5 condensed lignin, stem wood has the lowest, and the residue lies between. (31)P NMR analysis indicates that bark lignin contains more C-5 substituted phenolics and fewer aliphatic hydroxyl groups than the lignin isolated from stem wood or residue. The molecular weight distribution analysis indicates the bark lignin has higher weight-average molecular weight (M(w)) and polydispersity index than the lignin recovered from stem wood or residue.  相似文献

It is globally accepted that soil carbon (C) dynamics are at the core of interlinked environmental problems,deteriorating soil quality and changing climate.Its management remains a complex enigma for the scientific community due to its intricate relationship with soil nitrogen (N) availability and moisture-temperature interactions.This article reviews the management aspects of soil C dynamics in light of recent advances,particularly in relation to the availability of inorganic N pools and associated microbial processes under changing climate.Globally,drastic alterations in soil C dynamics under changing land use and management practices have been primarily attributed to the variation in soil N availability,resulting in a higher decomposition rate and a considerable decline in soil organic C (SOC) levels due to increased soil CO2 emissions,degraded soil quality,and increased atmospheric CO2 concentrations,leading to climate warming.Predicted climate warming is proposed to enhance SOC decomposition,which may further increase soil N availability,leading to higher soil CO2 efflux.However,a literature survey revealed that soil may also act as a potential C sink,if we could manage soil inorganic N pools and link microbial processes properly.Studies also indicated that the relative,rather than the absolute,availability of inorganic N pools might be of key importance under changing climate,as these N pools are variably affected by moisture-temperature interactions,and they have variable impacts on SOC turnover.Therefore,multi-factorial studies are required to understand how the relative availability of inorganic N pools and associated microbial processes may determine SOC dynamics for improved soil C management.  相似文献

The aim of the study was to evaluate the degradation and persistence of 2-amino 4-chloro 6-methyl pyrimidine (AM), nitrification inhibitor at 1 and 2 µg g?1 application rates in soil. The extraction of AM was done by QuEChER’s (Quick, Easy. Cheap. Rugged and Safe) method and the quantitative analysis by high-performance liquid chromatography (HPLC). AM decreased with time at both the levels of application with the decline being faster in the beginning up to 7 d. Dissipation of AM occurred in a single phase with the persistence data fitting well to the first-order kinetics. Half-lives of AM were determined to be 14.33 and 16.7 d at 1 and 2 µg g–1 levels application rates. Since AM remains effective for an adequate period of time, it can be used for increasing efficiency of nitrogenous fertilizers in rice–wheat cropping systems as well as a safeguard for controlling environmental pollution in subtropical soils.  相似文献
