Tick-borne encephalitis (TBE) in animals is not well understood yet. TBE virus (TBEV) serology in several host species could be valuable for epidemiological analyses in the field as well as for the detection of clinical cases. However, performance and suitability of the available test systems are not well assessed. Therefore, we evaluated two commercial TBEV-ELISA kits in a pilot study and compared them for their suitability in veterinary applications. For this purpose, we tested 163 field collected goat sera and evaluated the results by serum neutralization test (SNT) as "gold standard". Twenty-eight SNT positive sera (17.2%) were detected. The best suited ELISA kit was used for determination of a species-specific cutoff for horses, cattle, sheep, goats, pigs, mice, dogs, rabbits and monkeys with defined sera from animals without known or with improbable contact to TBEV. The level of non-specific ELISA results does not only differ between animal species but may also be influenced by the age of the tested animals. The number of sera which tested false positive by ELISA was higher in older than in young sheep. In order to obtain defined polyclonal sera as references, two dogs, cattle, goats, sheep, rabbits and pigs each, as well as one horse and 90 mice were immunized four times with a commercially available TBEV vaccine. In conclusion, our results demonstrated that commercial TBEV-ELISA kits are suitable for application in veterinary medicine for both, verification of clinical TBE cases and epidemiological screening. However, positive ELISA results should be verified by SNT. Only a very low number of false negative ELISA-results were found. 相似文献

The physical properties of lipids can have a major influence on lipid oxidation reactions. Edible oils contain surface-active compounds and water that can form physical structures such as reverse micelles. This study used the fluorescence probe, 5-dodecanoylaminofluorescein (DAF), to study both the physical and the chemical properties of stripped corn oil containing oleic acid and phosphatidylcholine. The fluorescence intensity of DAF increased with increasing water concentration in the edible oil. The addition of oleic acid decreased DAF fluorescence due to the ability of the free fatty acid to decrease the pH of the aqueous phase of the bulk oil. Phosphatidylcholine increased DAF fluorescence due to its ability to increase DAF exposure to the aqueous phase. Oleic acid had no impact on interactions between DAF and water-soluble peroxyl radicals, while phosphatidylcholine decreased peroxyl radical degradation of DAF. These results suggest that DAF could be a useful analytical tool to study the impact of the aqueous environment of bulk oil on lipid oxidation. 相似文献

The effect of sonication during chitin extraction from freshwater prawn shells on yield, purity, and crystallinity of chitin was investigated. Dry prawn shells were suspended for 4 h in 0.25 M HCl at 40 degrees C while they were sonicated for 0, 1, and 4 h. Demineralized shells were lyophilized, resuspended in 0.25 M NaOH, and sonicated again for 0, 1, and 4 h for protein removal. The yield of chitin decreased from 8.28 to 5.02% for nonsonicated and sonicated samples, respectively, which was attributed to losses of depolymerized materials in the wash water. The application of ultrasound enhanced the removal of proteins. In nontreated shells, the amount of protein was 44.01% and was reduced to 12.55, 10.59, and 7.45% after 0, 1, and 4 h of sonication treatments. The glucosamine content slightly decreased with sonication probably because of losses due to depolymerization. The crystallinity indices of chitins decreased as the time of sonication increased. The degree of acetylation of chitins was unaffected by sonication, but the degree of acetylation of chitosans produced from sonicated chitin decreased from 70.0 to 68.7 and 61.4% for 1 and 4 h sonicated samples, respectively. 相似文献

Headspace solid phase microextraction in combination with comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (HS-SPME-GC × GC-TOF-MS) was used for the detailed investigation of the impact of malolactic fermentation (MLF) using three commercial Oenococcus oeni strains on the volatile composition of Vitis vinifera cv. Pinotage wines. GC × GC allowed the identification of 115 volatile compounds, including both major constituents and trace-level compounds, in a single analysis. A number of compounds differing in mean concentration levels between the control wines and those fermented with different starter cultures were shown for the first time to be influenced by MLF and/or the bacterial strain. Principal component analysis (PCA) provided excellent separation between the wines fermented with different MLF starter cultures and the control wine. Significantly different levels for some volatile compounds in wines fermented with one of the LAB starter cultures could be indicative of metabolic differences of this strain. 相似文献

The charge distribution in atoms with closed electron shells is spherically symmetric, whereas atoms with partially filled shells can form covalent bonds with pointed lobes of increased charge density. Covalent bonding in the bulk can also affect surface atoms, leading to four tiny humps spaced by less than 100 picometers in the charge density of adatoms on a (001) tungsten surface. We imaged these charge distributions by means of atomic force microscopy with the use of a light-atom probe (a graphite atom), which directly measured high-order force derivatives of its interaction with a tungsten tip. Avian rotaviruses (AvRVs) represent a diverse group of intestinal viruses, which are suspected as the cause of several diseases in poultry with symptoms of diarrhoea, growth retardation or runting and stunting syndrome (RSS). To assess the distribution of AvRVs in chickens and turkeys, we have developed specific PCR protocols. These protocols were applied in two field studies investigating faecal samples or intestinal contents of diseased birds derived from several European countries and Bangladesh. In the first study, samples of 166 chickens and 33 turkeys collected between 2005 and 2008 were tested by PAGE and conventional RT-PCR and AvRVs were detected in 46.2%. In detail, 16.1% and 39.2% were positive for AvRVs of groups A or D, respectively. 11.1% of the samples contained both of them and only four samples (2.0%) contained rotaviruses showing a PAGE pattern typical for groups F and G. In the second study, samples from 375 chickens and 18 turkeys collected between 2009 and 2010 were analyzed using a more sensitive group A-specific and a new group D-specific real-time RT-PCR. In this survey, 85.0% were AvRV-positive, 58.8% for group A AvRVs, 65.9% for group D AvRVs and 38.9% for both of them. Although geographical differences exist, the results generally indicate a very high prevalence of group A and D rotaviruses in chicken and turkey flocks with cases of diarrhoea, growth retardation or RSS. The newly developed diagnostic tools will help to investigate the epidemiology and clinical significance of AvRV infections in poultry. 相似文献

In a 2-year field experiment (2002/2003) on a loess soil near Göttingen/Germany, pea (Pisum sativum L.) and oat (Avena sativa L.) were grown alone and intercropped at a range of densities. Shoot biomass, grain yields and amount of N in grain were evaluated and optimized using two different replacement series and a hyperbolic yield–density equation describing a response surface to address the following questions: (i) what is the optimal composition of the pea–oat intercrop with regard to maximum yields, (ii) which intercropping design is most suitable to describe competition effects in pea–oat intercrops and the optimal intercrop compositions and (iii) which intercropping design is best suited for the evaluation of field data. For (i), the optimal intercrop compositions varied depending on the growth conditions for the crops. Furthermore, optimal intercrop compositions were found above the recommended sole crop densities. The density of oat had to be reduced more than that of pea, especially when optimal grain-N yields were desired and soil-N content was high. For maximum grain-N yields, pea could be sown at high densities in combination with 5–50% of the recommended density of oat. Thus, density can be used as a yield regulator for specific purposes such as a high N yield. The effects of competition at final harvest were described equally by both designs (ii). Oat was the clearly stronger and pea the inferior competitor. In contrast to the replacement series design, the hyperbolic yield–density equation was capable of adding valuable information about the extent of intra and interspecific competition. As intraspecific competition was consistently more important than interspecific competition, resource complementarity could be hold responsible for intercrop advantages. The highest intercrop advantage was found when total intraspecific competition was low, as shown by the relative yield total (RYT) and niche differentiation index (NDI) values >1. However, due to the RYT dependence on sole crops and total densities, the replacement series design led to misleading interpretations of the yield advantages. Both experimental designs were able to describe the field-data reliably (iii), but the response surface design had the advantage of being unaffected by insufficient field emergences, as it is not based on total densities. Numbers of plants m−2 instead of seeds m−2 can be used for the evaluation. Data from sole crops are not needed for the response surface design and thus the feared high experimental effort of this design can be reduced. However, when using the replacement series design, experimental effort should be greater than normal, as different sole crop densities and more intercrop compositions within a replacement series can lead to a more precise interpretation of the competition effects. 相似文献

Synchrotron x-radiography and a fast x-ray detector were used to record the time evolution of the transient fuel sprays from a high-pressure injector. A succession of 5.1-microsecond radiographs captured the propagation of the spray-induced shock waves in a gaseous medium and revealed the complex nature of the spray hydrodynamics. The monochromatic x-radiographs also allow quantitative analysis of the shock waves that has been difficult if not impossible with optical imaging. Antibiosis is assumed to be an essential mechanism exerted by potential biocontrol agents (BCAs) of Trichoderma spp. Therefore, in the present study, we report for the first time on the elucidation and production of viridiofungin A (VFA)
from T. harzianum isolate T23 cultures and investigate the antifungal potential of VFA and some other secondary metabolites purified from T. harzianum cultures against Fusarium moniliforme. The bioautography assay revealed that T. harzianum isolates T16 and T23 excreted several secondary metabolites with antifungal activity. Following isolation and purification
of the antifungal zones, three fractions (F223, F323 and F423) from extracts of isolate T23 and two fractions (F416 and F516)
from extracts of isolate T16 exhibited pronounced fungitoxic activity in the bioautography and antibiotic disk assays against
Cladosporium spp. and F. moniliforme, respectively. The structure of the antifungal metabolite in fraction F323 was identified as viridiofungin A (VFA), the first
report of production of VFA by isolate T23 of T. harzianum. Following cultivation of isolate T23 in PDB medium for 9 days, 94.6 mg l−1 of VFA were determined. VFA and fraction F516 retarded the mycelial growth of F. moniliforme in the non-volatile phase assay by >90% for each 250 μg ml−1 7 days post-inoculation (dpi). While VFA and fraction F416 showed both volatile and non-volatile effects, fraction F516 seemed
to exhibit mainly non-volatile activity. Microscopic examination revealed that hyphae of F. moniliforme grown on VFA-amended medium were less branched and appeared thicker than untreated hyphae. Furthermore, in the presence of
VFA, formation of chlamydospores by F. moniliforme was increased. Finally, the antifungal spectrum of VFA towards various important plant pathogens was evaluated. Germination
of propagules of a variety of fungal pathogens in vitro was differentially inhibited by VFA. While in the presence of 100 μg ml−1 VFA conidial germination of V. dahliae was completely inhibited, a slightly higher concentration (150 μg ml−1) of the inhibitor was required to suppress germination of Phytophthora infestans sporangia or sclerotia of Sclerotinia sclerotiorum. Contrary to several reports in the literature, VFA proved to be fungistatic rather than fungicidal. However, neither VFA

Predicting single-cross performance is of special interest in hybrid breeding of triticale. We used molecular and phenotypic data of factorial triticale crosses and compared several approaches to predict their single-cross performance. Twenty-three inbred lines and their 76 incomplete factorial crosses were field evaluated for grain yield, plant height, and heading time at five locations in Central Europe. In addition, the parental lines were genotyped with 52 SSR markers. Plant height and heading time were predicted with high accuracy based on mid-parent performance. In contrast, prediction of hybrid performance based on mid-parent value was not accurate for grain yield. Using general combining ability effects led to an enhanced prediction accuracy of hybrid grain yield performance. This accuracy could be slightly improved using best linear unbiased prediction approaches. The prediction accuracy was considerably high even if the number of tested hybrids was small. Consequently, best linear unbiased prediction of hybrid performance is a promising tool for hybrid triticale breeding programs. 相似文献
