The structural changes of the cell wall and crystalline cellulose of Quercus variabilis wood in a pyrolysis system at several temperatures ranging from 250 to 500°C were investigated to examine the wood carbonization characteristics. The volume of the wood sample was decreased and the weight loss was increased by increasing the carbonization temperature. Vessels collapsed severely in tangential direction during the charring process above 350°C. SEM observation indicated that the layering structure of the walls in wood fibers and parenchyma cells were retained below 300°C. However, the cell walls above 350°C changed to an amorphous-like structure without cell wall layering. X-ray diffraction confirmed that the cellulose crystalline substance was still remained at the carbonization temperature of 300°C but was not detected above 350°C. It can be concluded that the transition from Q. variabilis wood to charcoal might occur at approximately 350°C. 相似文献

Based on the model–data comparison at the eddy-covariance observation sites from CarboEastAsia datasets, we report the current status of the terrestrial carbon cycle modeling in monsoon Asia. In order to assess the modeling performance and discuss future requirements for both modeling and observation efforts in Asia, we ran eight terrestrial biosphere models at 24 sites from 1901 to 2010. By analyzing the modeled carbon fluxes against the CarboEastAsia datasets, the strengths and weaknesses of terrestrial biosphere modeling over Asia were evaluated. In terms of pattern and magnitude, the carbon fluxes (i.e., gross primary productivity, ecosystem respiration, and net ecosystem exchange) at the temperate and boreal forest sites were simulated best, whereas the simulation results from the tropical forest, cropland, and disturbed sites were poor. The multi-model ensemble mean values showed lower root mean square errors and higher correlations, suggesting that composition of multiple terrestrial biosphere models would be preferable for terrestrial carbon budget assessments in Asia. These results indicate that the current model-based estimation of terrestrial carbon budget has large uncertainties, and future research should further refine the models to permit re-evaluation of the terrestrial carbon budget. 相似文献

Fusarium wilt is one of the most widespread diseases of pea. Resistance to Fusarium wilt race 1 was reported as a single gene, Fw, located on linkage group III. The previously reported AFLP and RAPD markers linked to Fw have limited usage in marker‐assisted selection due to their map distance and linkage phase. Using 80 F8 recombinant inbred lines (RILs) derived from the cross of Green Arrow × PI 179449, we amplified 72 polymorphic markers between resistant and susceptible lines with the target region amplified polymorphism (TRAP) technique. Marker–trait association analysis revealed a significant association. Five candidate markers were identified and three were converted into user‐friendly dominant SCAR markers. Forty‐eight pea cultivars with known resistant or susceptible phenotypes to Fusarium wilt race 1 verified the marker–trait association. These three markers, Fw_Trap_480, Fw_Trap_340 and Fw_Trap_220, are tightly linked to and only 1.2 cM away from the Fw locus and are therefore ideal for marker‐assisted selection. These newly identified markers are useful to assist in the isolation of the Fusarium wilt race 1 resistance gene in pea. 相似文献

In order to facilitate inverse modeling of time-series data and/or parameter estimation for process-based models of soil organic carbon (SOC) dynamics, we developed and applied a new surrogate CENTURY SOC model (SCSOC). While SCSOC has mass balance and decomposition kinetics equations for residue and soil organic matter (SOM) pools identical to those of CENTURY, it differs by being: (1) decoupled from models of plant growth, nutrient cycling, and hydrologic processes; (2) capable of employing daily, monthly, or annual timesteps; (3) solvable using widely available non-linear regression software. Here, we describe SCSOC and demonstrate that its numerical solution to the CENTURY SOC equations is highly accurate for both daily and monthly timesteps, provided the monthly physical forcing function inputs are appropriately derived from daily-resolution hydrologic model results.As an example of the proper application of the tool to obtain site-specific parameters and to investigate hypotheses using historical data, we analyzed SOC data from a single subplot of the Morrow Plots (Urbana, Illinois, U.S.A.) that has been cropped continuously in corn since 1876 using management practices that were common for east-central Illinois. The impact of different approaches typically used during calibration were examined by objectively estimating a variety of parameter sets that both govern SOM decomposition kinetics and define the initial mass and fractionation of the SOM. The results suggest that mean SOM turnover times were circa 5-fold longer during the period 1876-1954 relative to 1955-1995, when modern management practices were employed, and significantly different from default CENTURY predictions during both.Modified versions of SCSOC were also constructed in order to (1) show that uncertainty about historical soil erosion does not confound this interpretation of the data and (2) explore alternative assumptions about the structure of the model. One such alternate model showed that unexpectedly rapid decomposition of the corn-derived SOM combined with depletion of slowly-decomposing prairie-derived SOM could explain the post-1955 acceleration in decomposition. 相似文献

The acaricidal activity of materials derived from the rhizome of Cnidium officinale against adults of Dermatophagoides farinae and Dermatophagoides pteronyssinus was examined using direct contact application and fumigation methods and compared with that of benzyl benzoate and N,N-diethyl-m-toluamide (DEET). The active constituent of the Cnidium rhizome was identified as butylidenephthalide by spectroscopic analyses. Responses varied with dose. On the basis of 24-h LD(50) values, the acaricidal activity of butylidenephthalide (6.77 microg/cm(2)) against D. farinae adults was comparable to that of benzyl benzoate (8.54 microg/cm(2)). Very low activity was observed with DEET (37.59 microg/cm(2)). Against D. pteronyssinus adults, butylidenephthalide (6.46 microg/cm(2)) and benzyl benzoate (6.68 microg/cm(2)) were equitoxic. DEET (17.98 microg/cm(2)) was relatively inactive. The typical poisoning symptom of butylidenephthalide was lethargy of treated mites, leading to death without knockdown, whereas benzyl benzoate and DEET caused death following uncoordinated behavior. In a fumigation test with both mite species, butylidenephthalide was much more effective in closed containers than open ones. Naturally occurring C. officinale rhizome-derived materials merit further study as potential house dust mite control agents or lead compounds. 相似文献

Quercetin is a plant-derived flavonoid found in fruits or vegetables that has antioxidant properties and acts as a free radical scavenger. We investigated the effects of quercetin on porcine oocyte nuclear maturation and embryonic development after parthenogenetic activation. We then evaluated the antioxidant activities of quercetin by measuring reactive oxygen species (ROS) levels in matured oocytes. Immature oocytes were untreated or treated with 1, 10, and 50 µg/mL quercetin during in vitro maturation (IVM). Quercetin treatment did not improve oocyte nuclear maturation, but significantly higher blastocyst rates (p < 0.05) of parthenogenetically activated oocytes were achieved when the IVM medium was supplemented with an adequate concentration of quercetin (1 µg/mL). However, cleavage rates and blastocyst cell numbers were not affected. Oocytes treated with 1 or 10 µg/mL quercetin had significantly lower (p < 0.05) levels of ROS than the control and group treated with the highest concentration of quercetin (50 µg/mL). Moreover, this highest concentration was detrimental to oocyte nuclear maturation and blastocyst formation. Based on our findings, we concluded that exogenous quercetin reduces ROS levels during oocyte maturation and is beneficial for subsequent embryo development. 相似文献

The genetic relatedness of 7 Korean type O field strains of foot-and-mouth disease virus (FMDV) in clinical specimens collected from 5 different geographic locations in 2000 was investigated. The sequence of 162 nucleotides (nt 478-639) at the 3' end of the 1D (VP1) genes was determined from amplified cDNA fragments, and subjected to the analysis for the sequence identity/divergence and phylogenetic relationship. The overall nucleotide sequence divergence among the 7 field strains was 0 to 3.8%, suggesting that they are closely related to each other. Phylogenetic analysis with the known Middle East-South Asia (ME-SA) topotype strains showed that the 7 Korean field strains formed two distinct clusters within the same lineage of the ME-SA topotype strains. Cluster 1 consisted of the strains of the primary foci of infection (Paju and Hongseong), and closely related to the strains prevailed in the Far East. Cluster 2 comprised those of subsequently affected regions (Boryeong, Yongin, and Chungju), and was further diverged from the Cluster 1. The result of phylogenetic analysis indicated that the Korean strains may have evolved from a common ancestor of the Pan Asia strains, and that at least 2 phylogenetically clustered variants within the same lineage were prevalent during the epidemic. The potential origin and sources of the virus introduction to Korea were discussed. 相似文献

Recombinant human epidermal growth factor (rhEGF) stimulates the proliferation and migration of epithelial cells in human cell culture systems and animal models of partial-thickness skin wounds. This study investigated the effect of a topical rhEGF ointment on the rate of wound healing and skin re-epithelialization in a rat full thickness wound model, and verified whether or not the rhEGF treatment affected both myofibroblast proliferation and collagen synthesis in the dermis. When rhEGF (10 µg/g ointment) was applied topically twice a day for 14 days, there was significantly enhanced wound closure from the 5th to the 12th day compared with the control (ointment base treatment) group. A histological examination at the postoperative 7th day revealed that the rhEGF treatment increased the number of proliferating nuclear antigen immunoreactive cells in the epidermis layer. In addition, the immunoreactive area of alpha-smooth muscle actin and the expression of prolyl 4-hydroxylase were significantly higher than those of the control group. Overall, a topical treatment of rhEGF ointment promotes wound healing by increasing the rate of epidermal proliferation and accelerating the level of wound contraction related to myofibroblast proliferation and collagen deposition. 相似文献

Studies were conducted to compare polyphenol oxidase (PPO) specific activities in various milling fractions of a variety of wheat cultivars and determine the levels of activities in a number of cultivars from different localities and harvesting seasons. Substrate specificities were also investigated. Bran was singled out as the richest source of PPO activity, which may also influence the activity in the other milling fractions that are known to have some proportion of bran content. We showed by gel electrophoresis and spectrophotometrically that the protein responsible for PPO activity apparently exists as a single isoform in bran and that the observed enzyme activity is likely to be a tyrosinase type, not a laccase or peroxidase. The specific activity was not significantly different between the reduction shorts and break shorts from the same cultivar, indicating a similar level of bran contamination in these fractions. Very low levels of PPO activity were recorded in the flour of all cultivars studied. Bran was used, therefore, to determine the varietal differences in the PPO activities in a number of cultivars from different localities and seasons of harvest. Results showed that the most significant determinant of PPO activity was the genotype, and this may be influenced by seasonality. We also determined that, apart from substrate preferences by the PPO enzyme, some phenolic acids actually inhibit PPO. Furthermore, we found that bran of some cultivars extracted with acidified methanol inhibited PPO activity substantially, whereas other extracts had less inhibitory properties. Thus, these unknown compounds in wheat may inhibit endogenous PPO activity. 相似文献
