The monophenolase activity of quince pulp polyphenol oxidase was characterized by extracting samples using a combination of a two-phase partition step in Triton X-114, followed by a PEG 8000/phosphate partition step, and a final ammonium sulfate fractionation between 30 and 75%. The purification method avoids the loss of cresolase activity described in another quince pulp polyphenol oxidase. The activity was characterized by a lag period, whose duration depended on the substrate concentration, the pH, and the presence of catalytic amounts of o-diphenol. By increasing the concentration of o-diphenols, it was possible to evaluate the enzyme activation constant, K(act), which showed a value of 4.5 microM for 4-methylcatechol. A general kinetic mechanism for this enzyme is used to explain the loss of activity that normally occurs during quince pulp polyphenol oxidase purification. 相似文献

Thirteen metal elements were determined in 40 honey samples from Galicia with different environmental origins: rural, urban, and industrial areas. The data set of the honey metallic profiles was studied with a double purpose: first, to make a preliminary evaluation of honey as an environmental indicator in Galicia with the aim of monitoring pollution and, second, to compare the different capabilities of diverse pattern recognition prediction procedures for modeling the environmental surrounding of the hive. A certain level of similarity for urban and industrial samples was obtained using principal component analysis and cluster analysis, whereas significant differences for urban and industrial honeys were found in relation to rural honey samples. Different classification rules to associate metal content of honeys with their environmental surrounding were obtained by chemometric pattern recognition procedures. In general, the classification methods developed by neural networks provided better results than the traditional pattern recognition procedures. The metal profiles of honey seem to provide sufficient information to enable categorization criteria for classifying samples according to their environmental surrounding. Thus, honey could be a potential pollution indicator for the Galician area. 相似文献

The feasibility of novel controlled release systems for the delivery of active substances from films intended for food packaging was investigated. Because polyolefins are used highly for food-packaging applications, the reported high retention degree of antioxidants has limited their use for active packaging. Thus, in this study, PP films modified with different chain extenders have been developed to favor and control the release rates of the low molecular weight antioxidant tocopherol. The use of different chain extenders as polymer modifiers (PE-PEG M(w), 575; and PPG-PEG-PPG M(w), 2000) has caused significant changes in tocopherol-specific release properties. High-performance liquid chromatography coupled to PDA-FL and PDA-MS was used to test tocopherol and chain extender migration, respectively. The release of tocopherol from the prepared films with two chain extenders into two food simulants was studied. Different temperatures and storage times were also tested. Varying the structural features of the films with the incorporation of different levels of PPG-PEG-PPG, the release of tocopherol (food-packaging additive) into different ethanolic simulants could be clearly controlled. The effect of the temperature and storage time on the release of the antioxidant has been outstanding as their values increased. The migration of the chain extender, also tested, was well below the limits set by European legislation. 相似文献

Morphological and cultural features and restriction fragment length polymorphism analysis of ITS regions, including 5.8S rDNA, from 26 isolates of Colletotrichum species revealed that isolates from olive fruits, previously identified as C. gloeosporioides, belong to two taxa: C. acutatum and C. gloeosporioides. The objective of this research was to study the levels of some organic solutes, such as total protein, total soluble sugars, starch and proline in leaves, as well as abscisic acid concentration in xylem of pepper plants inoculated with Verticillium dahliae Kleb. Healthy and inoculated plants were always kept well watered. Measurements were made at time intervals after inoculation. Leaf water potential rapidly decreased as a consequence of fungal infection. However, relative water content in leaves only changed significantly from day 20 after inoculation, and such decreases coincided with a sharp build up of proline and total soluble sugars in leaves. Starch and protein levels, as well as abscisic acid concentration in xylem, declined in healthy and inoculated peppers as they became older. However, such decreases were more pronounced in infected plants, especially soon after inoculation. Results suggest that proline and total soluble sugars accumulation could be sensors of the damage caused by the fungal infection. 相似文献

Polymerase chain reaction (PCR) amplification of the nuclear 5S rDNA gene, has been used for the identification of sole (Solea solea) and Greenland halibut (Reinhardtius hippoglossoides). Two species-specific primers were designed to amplify specific fragments of the 5S rDNA gene in each species. The remarkably different size of the amplicons obtained gives, by simple agarose gel electrophoresis, two distinguishable band patterns for both flatfish species. This genetic marker can be very useful for the accurate identification of S. solea and Greenland halibut, to enforce labeling regulations. 相似文献

The Covadonga National Park was the first to be established (1918) in Spain and is one of the oldest in Europe. Its fauna and flora include many species of interest and the park area is representative of the prevailing ecosystem throughout the wet zone of Spain neighbouring the Cantabrican mountain range. Its history is complex, having been influenced by different political ideologies dominant within the Government. The present study compares a control diet to four experimental diets, in which two red beet (14% and 28%) and two betaine levels (0.9% and 1.63%) were incorporated in rainbow trout diets according to a factorial design. The effects of the inclusion of different red beet and betaine concentrations on fatty acid (FA) profile, lipid peroxidation, and antioxidant activity on rainbow trout fillets were investigated. Although no significant differences were observed with the control group, results indicated that red beet and betaine improved fish fillet FA profile, producing an increase in polyunsaturated FAs, mainly docosahexaenoic acid. Higher red beet and betaine concentrations increased flavonoid and phenolic content in the diets; however, no effects were observed for the antioxidant properties of rainbow trout fillets. 相似文献

The blue shark (Prionace glauca) is the most frequently captured shark in pelagic oceanic fisheries, especially pelagic longlines targeting swordfish and/or tunas. As part of cooperative scientific efforts for fisheries and biological data collection, information from fishery observers, scientific projects and surveys, and from recreational fisheries from several nations in the Atlantic and Indian Oceans was compiled. Data sets included information on location, size and sex, in a total of 478,220 blue shark records collected between 1966 and 2014. Sizes ranged from 36 to 394 cm fork length. Considerable variability was observed in the size distribution by region and season in both oceans. Larger blue sharks tend to occur in equatorial and tropical regions, and smaller specimens in higher latitudes in temperate waters. Differences in sex ratios were also detected spatially and seasonally. Nursery areas in the Atlantic seem to occur in the temperate south‐east off South Africa and Namibia, in the south‐west off southern Brazil and Uruguay, and in the north‐east off the Iberian Peninsula and the Azores. Parturition may occur in the tropical north‐east off West Africa. In the Indian Ocean, nursery areas also seem to occur in temperate waters, especially in the south‐west Indian Ocean off South Africa, and in the south‐east off south‐western Australia. The distributional patterns presented in this study provide a better understanding of how blue sharks segregate by size and sex, spatially and temporally, and improve the scientific advice to help adopt more informed and efficient management and conservation measures for this cosmopolitan species. 相似文献

Lipid composition of the mantle and digestive gland of Octopus vulgaris that were not fed for 27 days were determined. Every 3 days, three octopuses were killed and samples of the mantle and the
digestive gland (DG) were taken, in order to determine total lipids as well as lipid classes and fatty acids. Composition
in total lipids (TL) for the mantle was similar until day 21, then decreased and remained similar until the end of the experiment.
Composition in total lipids for the DG decreased significantly after 3 days, then remained similar until day 21, and then
decreased until the end of the experiment. As for the lipid classes, in the DG the main components were triglycerides and
sterol esters. Sterol esters suffered strong reductions after 10 days of starvation, while triglycerides remained similar
until day 21 and then decreased until the end of the experiment. Cholesterol decreased gradually throughout the experimental
period. For polar lipids, phosphatidylcholine and phosphatidylethanolamine increased during the first 3 days and then decreased
throughout the experiment. In the mantle, the only neutral classes that decrease were triacylglycerols and sterol esters,
while no polar lipid classes decreased in this organ. It was noticeable the decrease in almost all fatty acids in the DG after
