Black spruce (Picea mariana (Mill.) BSP) and tamarack (Larix laricina (Du Roi) K. Koch) are the predominant tree species in the boreal peatlands of Alberta, Canada, where low nutrient availability, low soil temperature and a high water table limit their growth. Effects of flooding for 28 days on morphological and physiological responses were investigated in greenhouse-grown black spruce and tamarack seedlings in a growth chamber. Flooding reduced root hydraulic conductance, net assimilation rate and stomatal conductance, and increased water-use efficiency (WUE) and needle electrolyte leakage in both species. Although flooded black spruce seedlings maintained higher net assimilation rates and stomatal conductance than flooded tamarack seedlings, flooded tamarack seedlings were able to maintain higher root hydraulic conductance than flooded black spruce seedlings. Needles of flooded black spruce developed tip necrosis and electrolyte leakage after 14 days of flooding, and these symptoms were subsequently more prominent than in needles of flooded tamarack seedlings. Flooded tamarack seedlings exhibited no visible injury symptoms and developed hypertrophied lenticels at their stem base. Application of exogenous ethylene resulted in a significant reduction in net assimilation, stomatal conductance and root respiration, whereas root hydraulic conductivity increased in both species. Thus, although flooded black spruce seedlings maintained a higher stomatal conductance and net assimilation rate than tamarack seedlings, black spruce did not cope with the deleterious effects of prolonged soil flooding and exogenous ethylene as well as tamarack.  相似文献

Context

Identifying the drivers shaping biological assemblages in fragmented tropical landscapes is critical for designing effective conservation strategies. It is still unclear, however, whether tropical biodiversity is more strongly affected by forest loss, by its spatial configuration or by matrix composition across different spatial scales.

Objectives

Assessing the relative influence of forest patch and landscape attributes on dung beetle assemblages in the fragmented Lacandona rainforest, Mexico.

Methods

Using a multimodel inference approach we tested the relative impact of forest patch size and landscape forest cover (measures of forest amount at the patch and landscape scales, respectively), patch shape and isolation (forest configuration indices at the patch scale), forest fragmentation (forest configuration index at the landscape scale), and matrix composition on the diversity, abundance and biomass of dung beetles.

Results

Patch size, landscape forest cover and matrix composition were the best predictors of dung beetle assemblages. Species richness, beetle abundance, and biomass decreased in smaller patches surrounded by a lower percentage of forest cover, and in landscapes dominated by open-area matrices. Community evenness also increased under these conditions due to the loss of rare species.

Conclusions

Forest loss at the patch and landscape levels and matrix composition show a larger impact on dung beetles than forest spatial configuration. To preserve dung beetle assemblages, and their key functional roles in the ecosystem, conservation initiatives should prioritize a reduction in deforestation and an increase in the heterogeneity of the matrix surrounding forest remnants.

In a field study using soil mesocosms in an acid spruce forest soil we investigated the effects of mesofauna and macrofauna on microbial biomass, dissolved organic matter, and N cycling. Intact soil monoliths were taken from the ground, defaunated by deep-freezing, and wrapped in nets of various mesh-sizes to control re-immigration of different faunal size-classes. The monoliths were then replanted in the field. Three treatments of mesocosms were prepared: (1) with only microbiota, (2) microbiota and mesofauna, and (3) microbiota, mesofauna, and macrofauna (= complex fauna). After 8 months of exposure the mesocosms and the unmanipulated control plots (treatment 4) were destructively sampled. We estimated microbial biomass by substrate-induced respiration and the chloroform fumigation-extraction method. N cycling was measured by monitoring microbial N mineralization, the NH inf4 sup+ content, and selected amino acids and the activities of protease, urease, and deaminase. The results from the L/F layer showed that the pool of the microbial biomass was not changed by the activity of the mesofauna. However, the mesofauna and macrofauna together enhanced SIR. An increase in microbial N mineralization was only observed in treatment 3 (microbiota + complex fauna). Protease activity and NH inf4 sup+ content increased in treatments 2 (microbiota + mesofauna) and 3 (microbiota + complex fauna). The complex fauna induced a soil pH increase in treatment 3 as opposed to treatment 1 and the control. This increase was presumably due to excretory NH inf4 sup+ . Principal component analysis revealed that the complex fauna in treatment 3 caused a significantly higher N turnover per unit of microbial biomass.  相似文献

In the present study, the temporal and spatial variation of the abundance of the alkane monooxygenase gene alkB and 16S rRNA genes in different soil compartments was analysed in the presence or absence of 2-methyl-4-chlorophenoxyacetic acid (MCPA) after the addition of pea litter to soil in a microcosm study. Samples were analysed shortly after litter addition (T0) and 1?week (T1), 3?weeks (T3) and 6?weeks (T6) after the addition of litter. In addition also, the quantity and quality of litter-derived alkanes was analysed and measured. The results revealed a fast and complete degradation of MCPA in all compartments throughout the experiment. Nevertheless, significant changes in the distribution patterns of short- and middle-chained alkanes suggest an interaction of MCPA and alkane degradation. alkB gene copy numbers were highly influenced by the time point of analysis and by the investigated soil compartment. Overall, an increase in alkB gene copy numbers from T0 to T3 was visible in the upper soil compartments whereas a decrease compared to T0 was measured in the deeper soil compartments. MCPA addition resulted in an increase of alkB abundance at T6. Gene copy numbers of 16S rRNA were not influenced by sampling time and soil compartment. In contrast to the control treatments, a slight increase in 16S rRNA gene copy numbers was visible at T1 and T3 compared to T0 in all soil compartments.  相似文献

To test the direct regeneration hypothesis and support natural disturbance-based forest management we characterized the structure and composition of boreal mixedwood forests regenerating after large wildfires and examined the influence of pre-fire stand composition and post-fire competing vegetation. In stands which had been deciduous (Populus sp.)-dominated, conifer (white spruce)-dominated, or mixed pre-fire we measured regeneration stocking (presence in 10 m2 plots), density and height 10–20 years post-burn in five wildfires in Alberta, Canada. Most plots regenerated to the deciduous or mixed stocking types; plots in the older fire and in stands that were pure conifer pre-fire had higher amounts of conifer regeneration. Surprisingly, regeneration in pre-fire ‘pure’ white spruce stands was most often to pine, although these had not been recorded in the pre-fire inventory. Pre-fire deciduous stands were the most resilient in that poplar species dominated their post-fire regeneration in terms of stocking, density and height. These stands also had the highest diversity of regenerating tree species and the most unstocked plots. High grass cover negatively affected regeneration density of both deciduous and conifer trees. Our results demonstrate the natural occurrence of regeneration gaps, pre- to post-fire changes in forest composition, and high variation in post-fire regeneration composition. These should be taken into consideration when developing goals for post-harvest regeneration mimicking natural disturbance.  相似文献

Comparisons were made between the aroma volatiles of the yellow-fleshed kiwifruit, "Hort16A", at two different stages of eating ripeness: firm and soft. The firm fruit contained a small number of aroma compounds that the soft fruit did not contain. In general, however, the largest difference between the two firmness categories was in the levels of esters, with the soft fruit containing higher concentrations and a larger number of esters than the firm fruit. In vitro analysis directly after maceration using atmospheric pressure chemical ionization mass spectrometry (APCI-MS) showed the relative importance of the most intense aromas between fruit at the two different firmness stages and was used to compare the release rates of aromas. A comparison of the aroma concentrations from gas chromatography mass spectrometry (GC-MS) and APCI-MS headspace analyses showed that the APCI-MS headspace showed less bias toward enzymatically generated lipid degradation compounds. A GC-sniffing study showed that many of the most intense compounds, acetaldehyde, hexanal, ethyl butanoate, and (E)-2-hexenal but not ethanol, showed odor activity in macerated fruit. In addition, dimethyl sulfide (DMS), a volatile present at very low levels in the fruit, also appeared to be an important contributor to the odor. In vivo analyses also showed much higher levels of aroma compounds in the soft fruit compared to the firm fruit, with evidence of persistence of some compounds, including DMS. There were a number of similarities between the breath profiles of the two panelists, which confirmed the importance of DMS in "Hort16A" aroma.  相似文献

We have recently provided evidence that Texas feline immunodeficiency virus (FIV-TX) isolates are an emerging subtype sharing a common ancestry with clade B isolates. Specific, pathogen-free cats were infected, intravenously, with 500, 2000 or 8000TCID(50) of the FIV-TX53 virus to study the acute stage of infection. Infection of cats resulted in lymphadenopathy at 10 days post-infection (p.i.). By 7 weeks p.i., gag specific antibody could be detected from sera of all infected cats. Virus could be detected by culturing PBMC and by nested capsid PCR. A reduction in the absolute numbers of lymphocytes and neutrophils was observed in infected cats although there was no trend identified between this reduction and the viral dose administered. By 11 weeks p.i., the CD4(+)/CD8(+) T cell ratios from all infected cats had dropped from approximately 2 to below 1. While decrease in the ratio was dependent on the viral dose, the T cell ratios of cats receiving the highest dose had significantly dropped below 1 by 4-7 weeks p.i. This decrease in the ratio was accompanied by a sharp and temporal decline in the absolute CD4(+) T cells and a slight increase in the absolute CD8(+) T cell numbers with a dramatic expansion of cells with CD8beta(low) chain expression.  相似文献
