The present research reproduces the chemical and microbiological reactions that occur naturally when a metal sulfide is discharged onto a natural soil, with special emphasis on iron cycle. The role of indigenous microbiota from an extremely acidic site on both weathering and attenuation processes related to the iron mobilization has been studied and the iron cycle has been reproduced at laboratory scale. In the first stage, the weathering phase, a residual sulfide mineral was bioleached using a mixed culture of iron-oxidizing bacteria isolated from the own substrate. The acid liquor obtained (pH 2), with a high metal concentration (160 mM in total iron), was filtered and neutralized. Solids obtained from the two sources (from the weathering process and after the neutralization stage) were characterized by X-ray and scanning electron microscope/energy dispersive X-ray spectroscopy, resulting ferric iron precipitates such as jarosites, goethites, and ferrihydrites with different crystalline properties. The contribution of ferric iron-reducing bacteria on the attenuation of high-content iron effluents was also studied. Mixed cultures of ferric iron-reducing bacteria, isolated from those acidic substrates, were active in reducing soluble ferric iron (60 mM in concentration), and a 66% of bioreduction was reached after 15 days. ABSTRACT Among potential components to construct Technosols for urban greening purposes, the commercially available geogenic coarse porous materials (CPMs) are mainly used in practice because of their high porosity. However, the knowledge of the hydraulic behavior of CPMs as well as of their mixtures with other substrates is limited, provoking their suboptimal usage. Therefore, we determined the water retention characteristics, including the available water capacity (AWC) of six geogenic CPMs: porlith, expanded shale, expanded clay, tuff, pumice, and lava. In order to obtain the water retention characteristics of the CPMs as well as of their mixture with sand (1:4 per volume), the following methods adapted from soil physics were applied over a wide range of pressure heads: Equi-pF apparatus, ceramic tension plates, pressure plate extractors, WP4C apparatus, and water vapor adsorption. The results were used to parametrize the modified Kosugi model (using SHYPFIT 2.0). Porlith and tuff have the highest AWC (0.37 m3 m?3 and 0.17 m3 m?3, respectively) and are the only ones which can be recommended as effective water-retaining materials. Further materials exhibit an AWC less than 0.10 m3 m?3. The CPMs exhibit a bimodal pore size distribution, which can be well described by the applied model, except for pumice and expanded shale. The mixtures present overall low AWCs up to 0.07 m3 m?3, with the pure sand having less than 0.03 m3 m?3. For practical application a quite high ratio of CPM is needed, and the mixing material must be adapted to the hydraulic properties of the CPMs. The water inside the CPMs may be easily available for plant roots able to penetrate in the CPMs’ coarse pores. 相似文献

The use of spent coffee grounds (SCG) as organic amendment is a triple solution: a reuse of this bio-residue (6 millions of tons per year), an increase in soil organic carbon (SCG contain 82% of carbohydrates and 13% of proteins) and a decrease in CO2 emissions into the atmosphere. Thus, we investigate the effect of SCG on soil and plants in an in vitro assay. The variables considered were SCG dose (2.5 and 10%), two agricultural soils (Calcisol and Luvisol), and four cultivation times (15, 30, 45 and 60 days). The chemical and physicochemical soil properties, SEM images and growth parameters were analyzed. The highest dose of SCG increased organic carbon, total N and available K and P by 286%, 188%, 45%, and 9%, respectively, while decreasing lettuce growth by 233% compared to control. The SEM study revealed that SCG particles are incorporated into soil aggregates and microorganisms grow over them, which is also corroborated by a 10-times increase of soil respiration rate. The behavior of both soils differs for some SCG effects. In conclusion, SCG have a great potential to increase soil fertility and further investigations are needed in order to improve the use of SCG as amendment. 相似文献

Little is known regarding the effect of fragmentation and human agricultural management on the genetic variation and gene flow of Cucurbita pepo L., 1753 in moderate fragmented areas in central Guatemala. We hypothesize that the genetic variation of C. pepo is affected by forest fragmentation and by traditional agricultural management. Therefore, we aim to determine: (1) the genetic diversity and genetic structure of C. pepo in the Cloud Forest Corridor (CFC) (2) the extent of genetic admixture between commercial variety (CV) and traditional landraces (TL) of C. pepo, (3) the effect of habitat fragmentation in the population genetics of C. pepo with a landscape approach, and (4) the potential relationship between traditional management practices and genetic diversity of C. pepo in the CFC. We detected the existence of high level of genetic diversity (AR = 3.43; He = 0.50), inbreeding (Fis = 0.25) and moderate population structure of C. pepo in the CFC (Fst = 0.16). No correlation between landscape and genetics was found. Also, we found high genetic admixture between CV and TL. It seems that human practices, mainly related with seed exchange patterns, could affect genetic diversity of C. pepo in the CFC. C. pepo populations in the CFC are structured, with inbreeding, and show admixture with the CV, an aspect that could affect its genetic diversity. The agricultural management influenced the population genetics of C. pepo in the CFC, but the landscape did not. We suggest that special efforts should be made to preserve the diversity of this important indigenous food source for Guatemalan people as well as their management practices. 相似文献

This paper presents the main results obtained using the heterogeneous photocatalysis with TiO2 technology in the removal of the emerging contaminant benzophenone-3 at pilot plant scale in a cylinder parabolic solar collector. In this sense, the effects of the operational parameters, pH and catalyst concentration, were analyzed, and after the use of the response surface methodology, the conditions that, under the experimental range, conduct to a higher removal of the analyte were selected. Additionally, analysis of the total organic carbon content and the biodegradability index (relationship between BDO5 and COD) of the treated samples were carried out. In general, results allowed us to infer that this kind of treatment is effective in removing a considerable percentage of the substrate, and that it could become an effective alternative to water treatment and decontamination. 相似文献

The process for extracting sugarcane juice (Saccharum officinarum) represents the point of greatest contamination in sugarcane mills. Sodium dithiocarbamate also known as metam-sodium or MS is added to inhibit the growth of microorganisms especially Leuconostoc mesenteroides which is responsible for forming polysaccharides. Metam-sodium, upon decomposition, produces highly toxic byproducts. According to literature, under acidic conditions, MS is hydrolyzed resulting in methylamine (MA), CH3NH2, and carbon disulfide (CS2), and in dilute alkaline solutions, MS produces an oxidation reaction characterized by the formation of elemental sulfur (S) and methyl isothiocyanate (MITC). In this paper, it was studied how MS decomposes to MITC and/or MA considering the effects of the matrix (methanol and water); of temperature (4 and 25 °C); of processing time (0, 1, 2, 3, 4 days); and of pH (4.0, 4.5, 7.0). A second experimental design considering the effects of the matrix (water and sugarcane juice); of temperature (4, 25, 35, 45 °C); of processing time (30, 300 min); and of pH (4.0, 4.5, 7.0) was derived from the results obtained considering MITC and/or MA formation. According to the statistical analysis of these results (p < 0.05), the order of the influential factors was as follows: time > matrix > pH > temperature. Results also indicated that the water matrix at pH = 4.5 and 45 °C had the lowest degradation rate (k), with a value of 8.82 day?1, while for the sugarcane juice matrix at the same pH and temperature conditions was larger, with a k value of 30.07 day?1. This study evaluates quantitatively the suitability of the use of site-specific socio-economic and environmental data as indicators
to rapidly assess patterns of diversity and genetic erosion risk in cassava. Socio-economic data as well as farmers’ estimation
of genetic erosion were collected in the study area, the Ucayali region of the Peruvian Amazon, through interviews with 285
cassava farmers in 50 communities, while diversity was assessed based on agromorphological characterization of 295 cassava
accessions. Using multivariate regression analyses, 50 and 45% of the variation in respectively diversity and genetic erosion
estimation could be explained by a selected set of socio-economic and environmental indicators. In both regression models
four out of the total of 38 variables proved to contribute significantly (at p < 0.10 level). Additionally, the study revealed that farmers are a good direct source of information on the diversity present
at community level, which can contribute to the development of methodologies to assess diversity more rapidly. The results

Thirty Portuguese and eight foreign olive (Olea europaea L.) cultivars were screened using Random Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeat (ISSR) markers.
Twenty RAPD primers amplified 301 reproducible bands of which 262 were polymorphic; and 17 ISSR primers amplified 204 bands
of which 180 were polymorphic. The percentage of polymorphic bands detected by ISSR and RAPD was similar (88 and 87%, respectively).
The genetic variability observed was similar in the Portuguese and foreign olive cultivars. Seven ISSR and 12 RAPD primers
were able to distinguish individually all 38 olive cultivars. Twenty specific molecular markers are now available to be converted
into Sequence Characterised Amplified Region (SCAR) markers. Relationships among Portuguese and foreign cultivars is discussed. 相似文献

Free and total fluorescent compounds, browning index, and color formation were measured in milk-based powdered infant formulas (IF) during 2 years of storage at 20 and 37 degrees C. The excitation spectra from 415 nm emission show three peaks (ex lambda1 = 270 nm, lambda2 = 325/315 nm, lambda3 = 350 nm) and from 347 nm excitation two emission peaks (415 and 520 nm), and no wavelength shifts were observed. Temperature and time of storage exert in general no significant effect on the development of fluorescence emission intensity and browning index. However, an important increase in pentodilysine was recorded-probably because of the iron and ascorbic acid contents of the samples-as well as in browning index in adapted IF. In both IF a color increase (deltaE) throughout storage was observed, this increase being greater in samples stored at 37 degrees C than in those stored at 20 degrees C. The increase in color with time fitted a linear regression model. Color appeared to be an indicator of sufficient sensitivity to measure the effect of temperature or storage time. 相似文献
