[目的]测定水体、土壤沉积物以及鱼体内有机氯及含氮杀虫剂残留物,为科学评估杀虫剂残留对巴基斯坦地区公共卫生、农业和环境的影响提供依据。[方法]以食用鱼南亚野鲮为材料,设计2组体重,采用高效液相色谱法分别测定水体、土壤沉积物以及南亚野鲮体内α-硫丹、DDE、甲基对硫磷、异丙隆、呋喃丹、阿特拉津等含量。[结果]土壤沉积物中DDE的含量达(2.340±0.025)μg/g,在250~750g的南亚野鲮体内的含量分别为(0.270±0.0006)μg/g,但在水体中未发现DDE残留;不同饲料中农药残留物90%为有机磷、呋喃丹以及有机氯杀虫剂,6%为杀菌剂,仅4%为除草剂,在250~750g以及800~1300g的南亚野鲮体内硫丹的含量分别达到(0.491±0.0006)μg/g和(3.050±0.0608)μg/g,异丙隆的含量分别达到(0.010±0.0003)μg/g和(0.014±0.0006)μg/g,且随体重上升,其脂肪含量增加,积累的农药残留物则越多;硫丹、甲基对硫磷、阿特拉津和呋喃丹的含量在水体达到最大残留限量水平0.001μg/g。[结论]通过生物积累和在自然界中的运输以及再沉积作用,有机氯及其他杀虫剂的使用给全球的环境造成了严重污染,因此,在巴基斯坦已经禁止使用DDT等有机氯农药。  相似文献

常见稻麦秸秆在实际作业过程中与刀具作用发生2种变形:挤压秸秆发生弯曲变形和剪切秸秆发生剪切变形,根据2种变形设置弯曲试验和剪切试验,分别测试15~30 mm/min加载速率下弯曲强度和剪切强度。试验结果表明,小麦秸秆抗弯强度随着加载速率增加而减小;水稻秸秆抗弯强度随着转速增加,表现为先增加后减小。小麦秸秆剪切强度随着加载速率增加,表现为先增加后减小;水稻秸秆剪切强度随着加载速率增加而增加。其次在田间试验中,根据2种变形设置横纵向秸秆以及180~280 r/min的刀轴转速,并对其反旋作业。采用同位素示踪法,即根据标记秸秆前后坐标变化值来代替机具在幅宽范围内纵横向秸秆的位移变化,将得到的标记点坐标在三维坐标系中用Matlab绘制成曲线,该曲线形状与旋耕刀片在刀轴上排列相似。耕作后位移和坐标结果表明:水稻秸秆位移大于小麦秸秆位移,水稻秸秆标记点位移变化较小麦秸秆标记点位移变化均匀;2种秸秆排列方式对应2种分布情况:纵向标记秸秆坐标在X轴上以零点对称分布,横向标记秸秆坐标在X、Z轴上两侧对称分布。基于以上因素考虑,实际作业中,选择刀轴转速230 r/min以及改变秸秆在田间排列方式,以使秸秆还田效果达到更佳。  相似文献

Airborne dust particles were sampled in a broiler house using the quartz crystal microbalance cascade impactor designed for the measurement of mass concentration and size distribution from 0.05 to 25 gm. No significant difference was noticed in the dust concentration from morning to evening or at different sampling sites in the broiler house. The concentration of airborne dust particles decreased as the age of the chickens increased. Ratio of dust particles smaller than 0.8 pm to the total dust ranged from 10 to 50%. The highest concentration of dust was in the particle size range of 3.2 μm. The fogging system in use reduced the concentration of larger particles but not that of the smaller particles.  相似文献

Monounsaturated fatty acids (MUFA)-rich and n-6 polyunsaturated fatty acid (n-6 PUFA)-rich vegetable oils are increasingly used as fish oil replacers for aquafeed formulation. The present study investigated the fatty acid metabolism in juvenile European sea bass (Dicentrarchus labrax, 38.4 g) fed diets containing fish oil (FO, as the control treatment) or two different vegetable oils (the MUFA-rich canola/rapeseed oil, CO; and the n-6 PUFA-rich cottonseed oil, CSO) tested individually or as a 50/50 blend (CO/CSO). The whole-body fatty acid balance method was used to deduce the apparent in vivo fatty acid metabolism. No effect on growth performance and feed utilization was recorded. However, it should be noted that the fish meal content of the experimental diets was relatively high, and thus the requirement for n-3 long-chain polyunsaturated fatty acid (n-3 LC-PUFA) may have likely been fulfilled even if dietary fish oil was fully replaced by vegetable oils. Overall, relatively little apparent in vivo fatty acid bioconversion was recorded, whilst the apparent in vivo β-oxidation of dietary fatty acid was largely affected by the dietary lipid source, with higher rate of β-oxidation for those fatty acids which were provided in dietary surplus. The deposition of 20:5n-3 and 22:6n-3, as % of the dietary intake, was greatest for the fish fed on the CSO diet. It has been shown that European sea bass seems to be able to efficiently use n-6 PUFA for energy substrate, and this may help in minimizing the β-oxidation of the health benefiting n-3 LC-PUFA and thus increase their deposition into fish tissues.  相似文献

Chemical mass balance (CMB) and principal component analysis (PCA) are used together for source identification and source apportionment in this air pollution modeling study. Source profile sets, each of which contains five source profiles based on ten pollutant species, were generated using a computer program. Another algorithm was implemented to produce ten random data sets, which was composed of 100 simulated measurement results for all of ten pollutant species. Ten source profile sets were selected. Five of them contained sources of dissimilar characteristics, whereas the other five were chosen from those of similar emission profiles. Ten simulated data sets for each source profile set were used in the analyses. PCA was applied to all simulated data sets; a number of principal factors were extracted and interpreted. The identified sources for each data set were used in fitting with CMB analyses, and source contributions were estimated. The performance of PCA–CMB combination was evaluated in the aspect of percent variance explained, percent apportionment, R 2, and χ 2. PCA was able to explain 89.6% to 100% of the variance within the data sets used. Two to five sources were extracted depending on the characteristics of source profile sets used. CMB was found to be successful in the aspect of percent apportionment since 95.4% to 100% of mass concentrations were apportioned. The values of R 2 and χ 2 were found out to range from 0.981 to 1.000 and from 0.000 to 29.947, respectively. Evaluating overall results from the analyses, PCA–CMB combination produced satisfactory results in the aspect of source identification and source apportionment.  相似文献

Effect of nickel sulphate at the sublethal dose of 64 ppm (0.8 of LC50 96 hr) on the blood glucose levels of the freshwater fish, Colisa fasciatus, has been estimated from 3 to 96 hr. The blood glucose level exhibits a steady increase due to Ni toxicity. A maximum increase of 85.08% is observed at 96 hr (P < 0.001). It is suggested that the hyperglycemia in C. fasciatus, caused by exposure to nickel sulphate, is possibly a reflection of stress-induced hormone mediated response. It appears that the blood glucose level is a reliable indicator of Ni toxicity to fish.  相似文献

In this study, the polyphenol oxidase (PPO) of artichoke (Cynara scolymus L.) was first purified by a combination of (NH(4))(2)SO(4) precipitation, dialysis, and a Sepharose 4B-L-tyrosine-p-aminobenzoic acid affinity column. At the end of purification, 43-fold purification was achieved. The purified enzyme migrated as a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Polyacrylamide gel electrophoresis indicated that PPO had a 57 kDa molecular mass. Second, the contents of total phenolic and protein of artichoke head extracts were determined. The total phenolic content of artichoke head was determined spectrophotometrically according to the Folin-Ciocalteu procedure and was found to be 425 mg 100 g(-1) on a fresh weight basis. Protein content was determined according to Bradford method. Third, the effects of substrate specificity, pH, temperature, and heat inactivation were investigated on the activity of PPO purified from artichoke. The enzyme showed activity to 4-methylcatechol, pyrogallol, catechol, and L-dopa. No activity was detected toward L-tyrosine, resorsinol, and p-cresol. According to V(max)/K(m) values, 4-methylcatechol (1393 EU min(-1) mM(-1)) was the best substrate, followed by pyrogallol (1220 EU min(-1) mM(-1)), catechol (697 EU min(-1) mM(-1)), and L-dopa (102 EU min(-1) mM(-1)). The optimum pH values for PPO were 5.0, 8.0, and 7.0 using 4-methylcatechol, pyrogallol, and catechol as substrate, respectively. It was found that optimum temperatures were dependent on the substrates studied. The enzyme activity decreased due to heat denaturation of the enzyme with increasing temperature and inactivation time for 4-methylcatechol and pyrogallol substrates. However, all inactivation experiments for catechol showed that the activity of artichoke PPO increased with mild heating, reached a maximum, and then decreased with time. Finally, inhibition of artichoke PPO was investigated with inhibitors such as L-cysteine, EDTA, ascorbic acid, gallic acid, d,L-dithiothreitol, tropolone, glutathione, sodium azide, benzoic acid, salicylic acid, and 4-aminobenzoic acid using 4-methylcatechol, pyrogallol, and catechol as substrate. The presence of EDTA, 4-aminobenzoic acid, salicylic acid, gallic acid, and benzoic acid did not cause the inhibition of artichoke PPO. A competitive-type inhibition was obtained with sodium azide, L-cysteine, and d,L-dithiothreitol inhibitors using 4-methylcatechol as substrate; with L-cysteine, tropolone, d,L-dithiothreitol, ascorbic acid, and sodium azide inhibitors using pyrogallol as substrate; and with L-cysteine, tropolone, d,L-dithiotreitol, and ascorbic acid inhibitors using catechol as a substrate. A mixed-type inhibition was obtained with glutathione inhibitor using 4-methylcatechol as a substrate. A noncompetitive inhibition was obtained with tropolone and ascorbic acid inhibitors using 4-methylcatechol as substrate, with glutathione inhibitor using pyrogallol as substrate, and with glutathione and sodium azide inhibitors using catechol as substrate. From these results, it can be said that the most effective inhibitor for artichoke PPO is tropolone. Furthermore, it was found that the type of inhibition depended on the origin of the PPO studied and also on the substrate used.  相似文献

Inability to produce large quantities of high quality, relatively diseasefree potato seed is a major constraint to providing economic support to a highly important and rapidly expanding potato production industry in Pakistan. Studies were undertaken for 5 years to evaluate productivity of seed of cultivars Red Bad, Ajax, Atica, Patrones, Montana, Cosima, Desiree, and Cardinal grown at five environmentally diverse locations in Pakistan versus seed of the same cultivars imported from Europe. The productivity trials were conducted during the autumn and spring seasons at the Ayub Agricultural Research Institute, Faisalabad (Punjab Plains). The results from the 5-year test period indicate that seed of each of the cultivars produced in the high hills (Baluchistan, Northwest Frontier Province) and during autumn (September to December) in the Punjab Plains, the latter cold stored until the following September, are equally as productive as seed imported from Europe.  相似文献

This study was carried out to determine the prevalence of cavical myiasis caused by Oestrus ovis larvae in sheep of Kars province of north-eastern part of Turkey. From 30 to 35 sheep heads (total of 387) were examined every month regularly for O. ovis larvae during 12 months. Of 387 heads, 156 (%40.3) were infested with O. ovis larvae.. The prevalence of nasal myiasis was 54.3% in spring, 41% in summer, 28% in fall, and 38.9% in winter. The differences among seasons were significant statistically (P < 0.05). Infestation rate up to 1-years-old was 30.0%, 1 to 3 years-old 40.0% and older than 3 years old was 52.4%. The number of larvae made peak in spring months and went down in the months of fall. The mean number of larvae regarding examined animals was 1.8, and the mean according to infested animals was 4.5. Density of O.ovis larvae in infested sheep were changed from 1 to 31. Infestation rate in the morkaraman breed was higher (43.4%) comparing to the rate in the akkaraman breed (31.3%). The differences between sheep breed were also significant (p < 0.05). Sheep with dark colored head had higher infestation rate than that of sheep with light colored head (p < 0.05).  相似文献
