The effects of artificial photoperiod regimes on reproductive patterns have been studied in several species, as have haematological parameters. However, information on how artificial photoperiods may affect blood components is scarce, especially under field conditions. We have assessed the effects of constant light [long day (LD) photoperiod: 24 h (light):0 h (dark)] on haematological parameters of cultured rainbow trout in Chile (Southern Hemisphere). In the initial stage (March up to June), two groups of fish were maintained under similar conditions and under the ambient (natural) photoperiod (NP). One group was then placed under the LD photoperiod regime for 2 months (June/July), following which it was returned to␣the NP (August–January); the control group remained under the NP throughout the experiment (March–January). All fish were assessed for haematological parameters and growth performance at various times during the experiment. During the initial stage, no differences were found between groups. However, at the end of the LD 24:0 period, the LD fish had increased haematocrit values and erythrocyte numbers and diminished mean corpuscular haemoglobin. After the LD fish had been returned to the NP, they developed secondary sexual characteristics and had a 40–44% decrease in the number of leukocytes.  相似文献

The rules of nucleic acid base-pairing have been used to construct nanoscale architectures and organize biomolecules, but little has been done to apply this technology in vivo. We designed and assembled multidimensional RNA structures and used them as scaffolds for the spatial organization of bacterial metabolism. Engineered RNA modules were assembled into discrete, one-dimensional, and two-dimensional scaffolds with distinct protein-docking sites and used to control the spatial organization of a hydrogen-producing pathway. We increased hydrogen output as a function of scaffold architecture. Rationally designed RNA assemblies can thus be used to construct functional architectures in vivo.  相似文献

We demonstrate enhanced generation of coherent light in the "water window" region of the soft x-ray spectrum at 4.4 nanometers, using quasi-phase-matched frequency conversion of ultrafast laser pulses. By periodically modulating the diameter of a gas-filled hollow waveguide, the phase mismatch normally present between the laser light and the generated soft x-ray light can be partially compensated. This makes it possible to use neon gas as the nonlinear medium to coherently convert light up to the water window, illustrating that techniques of nonlinear optics can be applied effectively in the soft x-ray region of the spectrum. These results advance the prospects for compact coherent soft x-ray sources for applications in biomicroscopy and in chemical spectroscopy.  相似文献

West Nile virus (WNV) is a zoonotic flavivirus that is transmitted by blood-suckling mosquitoes with birds serving as the primary vertebrate reservoir hosts (enzootic cycle). Some bird species like ravens, raptors and jays are highly susceptible and develop deadly encephalitis while others are infected subclinically only. Birds of prey are highly susceptible and show substantial mortality rates following infection. To investigate the WNV pathogenesis in falcons we inoculated twelve large falcons, 6 birds per group, subcutaneously with viruses belonging to two different lineages (lineage 1 strain NY 99 and lineage 2 strain Austria). Three different infection doses were utilized: low (approx. 500 TCID50), intermediate (approx. 4 log10 TCID50) and high (approx. 6 log10 TCID50). Clinical signs were monitored during the course of the experiments lasting 14 and 21 days. All falcons developed viremia for two weeks and shed virus for almost the same period of time. Using quantitative real-time RT-PCR WNV was detected in blood, in cloacal and oropharyngeal swabs and following euthanasia and necropsy of the animals in a variety of neuronal and extraneuronal organs. Antibodies to WNV were first time detected by ELISA and neutralization assay after 6 days post infection (dpi). Pathological findings consistently included splenomegaly, non-suppurative myocarditis, meningoencephalitis and vasculitis. By immunohistochemistry WNV-antigens were demonstrated intralesionally. These results impressively illustrate the devastating and possibly deadly effects of WNV infection in falcons, independent of the genetic lineage and dose of the challenge virus used. Due to the relatively high virus load and long duration of viremia falcons may also be considered competent WNV amplifying hosts, and thus may play a role in the transmission cycle of this zoonotic virus.  相似文献

Barley spot blotch (SB), caused by Cochliobolus sativus, is an important barley disease which causes extensive grain yield losses. These losses may not always correlate directly with the amount of diseased leaf area. Two barley cultivars, Quebracho (susceptible to SB) and Carumbé (with intermediate susceptibility to SB), were compared in field experiments in 2003, 2004 and 2006. Plots of each cultivar were either inoculated with C. sativus or protected with fungicide under field conditions to generate contrasting treatments: i) diseased, and ii) free of disease, respectively. SB severity over the growing season, photosynthetic rate on leaves with no visible symptoms and grain yield were assessed for each treatment and year. There was no treatment effect on cv. Carumbé, while cv. Quebracho showed a significant yield reduction, even though SB severity during the grain filling period was <10 %. This yield reduction was associated with a reduced photosynthetic rate at the beginning of the grain filling period in cv. Quebracho. A similar experiment was conducted under greenhouse conditions, adding a treatment without inoculum or fungicide. There were no differences in photosynthetic rate or grain yield per plant among treatments. These results suggest a distinct physiological response to SB infection among cultivars affecting leaf photosynthetic rate, and SB severity may not be the best estimator of yield losses caused by SB.  相似文献

Humans have used Caribbean island landscapes for millennia. Theconversion of wild lands to built-up lands or to agricultural lands in thesetropical countries follows predictable patterns. Conversion of moist forestlifezones and fertile flatlands is faster than conversion of wet and rain forestlife zones and low fertility steep lands. In Puerto Rico, these trends areleading to increased built-up areas, environmental surprises, and increaseddependence on external subsidies. Changes over the past 50 yr alsoinclude a reversal in deforestation and increase in forest patch size in spiteof increasing human population density. Present forests have different speciescomposition than the original ones but are indistinguishable in physiognomy andbasic function. The reversal of deforestation and forest fragmentation trends,if accompanied by an understanding of the forces that cause the reversal, canresult in the development of tools for landscape management. Tropical landscapemanagement requires understanding and application of natural resiliencemechanisms of ecosystems, greater use of ecological engineering approaches toinfrastructure development, enforcement of zoning laws, enlightened economicdevelopment policies, and an understanding and agreement of a conservationvision among all sectors of society. Mixing species in new combinations to formnew ecosystems is a necessary step in the development of future landscapes.Landscapes have always changed over time, but we sense,somewhat uneasily, that the processes now at work are more powerfully focusedand possibly irreversible.Our present assumption is that generally tropical lands aremuch more sensitive and less resilient than those in the temperatezones....the long-term world trend in land uses is a cascadingloss in capacity (i.e., degradation) in every broad category of landuse. Phagocyte numbers and activities were compared in milk from 2 groups of uninfected mammary-gland quarters from 3 cows each: 6 quarters with a high (> or = 200 000/mL) somatic cell concentration (SCC), analyzed as 4 individual quarters and 1 pooled sample; and 12 quarters with a low SCC (< 200 000/mL), analyzed as 6 paired samples. The concentrations and ability of macrophages and polymorphonuclear (PMN) cells to phagocytize fluorescent microspheres were determined by flow cytometry after exposure of the cells to the microspheres. The macrophages and PMNs contained 2 major subpopulations, characterized by low phagocytic (LP) or high phagocytic (HP) ability. The quarters with high SCCs had significantly lower percentages of HP cells than did the quarters with low SCCs (P < 0.01). Whether mammary-gland quarters or cows were the unit of analysis, the HP/LP ratio was negatively related to the SCC (P < 0.04), which explained more than 50% of the SCC variability. Thus, poor bovine mammary-gland phagocytic ability may be associated with high SCC. Longitudinal studies are suggested to further explore and characterize these relationships.  相似文献

We explored the hypothesis that the outcome of bacterial invasion (infection or no infection) may depend on immunologic factors when bacterial and environmental factors are kept constant. Leukocyte surface molecules (CD3, CD2, CD4, CD8, CD11b, and CD45r) were assessed before and 3 times after intramammary infusion of Staphylococcus aureus in 5 dairy cows. The somatic cell count (SCC/mL), bacterial count (colony-forming units [CFUs]/mL), ratio of milk phagocytes (mononuclear [Mphi] plus polymorphonuclear [PMN] cells) to lymphocytes (P/L index), and ratio of PMN to Mphi cells (PMN/Mphi index) were determined. Although all cows showed evidence of inflammation resulting from the infusion (the median P/L ratio was 11 times greater 1 d after infusion than before infusion), bacteria were not obtained from the milk of 2 cows. Threshold-like responses, resulting in bacterial counts that approached zero (indicating no infection) and SCCs of less than 500000/mL, were observed when the milk CD2+ lymphocyte proportion exceeded 73% (P < or = 0.007). At 1 d after infusion, 7 immune factors distinguished infected cows from those without infection with more than 95% confidence: compared with infected cows, uninfected cows had higher proportions of CD3+, CD2+, CD4+, and CD8+ T cells, higher densities of CD3 and CD2 molecules per cell, and a higher density of CD11b molecules on milk Mphi cells. At 7 d after infusion, the PMN/Mphi index was lower (94% confidence) in uninfected than in infected cows. At 14 d, the CD2, CD8, and CD45r marker densities were lower than those at 1 d (P < 0.02), findings compatible with memory function. Synergism was suggested by the combined effects of the proportions of CD3+, CD2+, and CD11b+ cells, which explained 75.5% of the bacterial-count variability (P < 0.001); alone, none of these markers predicted CFU variability. These results support further studies aimed at identifying cows capable (or incapable) of early bacterial clearance.  相似文献
