During the 2003 to 2004 outbreak of avian influenza A (H5N1) virus in Asia, there were anecdotal reports of fatal infection in domestic cats, although this species is considered resistant to influenza. We experimentally inoculated cats with H5N1 virus intratracheally and by feeding them virus-infected chickens. The cats excreted virus, developed severe diffuse alveolar damage, and transmitted virus to sentinel cats. These results show that domestic cats are at risk of disease or death from H5N1 virus, can be infected by horizontal transmission, and may play a role in the epidemiology of this virus. 相似文献

Inundative biological control of Arceuthobium americanum occurring on Pinus contorta var. latifolia with the fungus Colletotrichum gloeosporioides was investigated. Isolates of C. gloeosporioides were collected throughout British Columbia, Canada, and one isolate was selected for assessment based on its growth and sporulation in culture. The fungus was formulated using the ‘Stabileze’ method and inoculated onto A. americanum under field conditions. It became established on some replicates and there was a higher incidence of C. gloeosporioides on treated replicates than controls. In some replicates, the treatment reduced fruit production, leading to a decrease in the reproductive capacity of the dwarf mistletoe plant; however, the efficacy was highly variable and not significant. 相似文献

Mycobacterium leprae induces T cell reactivity and protective immunity in the majority of exposed individuals, but the minority that develop leprosy exhibit various types of immunopathology. Thus, the definition of epitopes on M. leprae antigens that are recognized by T cells from different individuals might result in the development of an effective vaccine against leprosy. A sequence from the 65-kD protein of this organism was recognized by two HLA-DR2-restricted, M. leprae-specific helper T cell clones that were derived from a tuberculoid leprosy patient. Synthetic peptides were used to define this epitope as Leu-Gln-Ala-Ala-Pro-Ala-Leu-Asp-Lys-Leu. A similar peptide that was derived from the third hypervariable region of the HLA-DR2 chain, Glu-Gln-Ala-Arg-Ala-Ala-Val-Asp-Thr-Tyr, also activated the same clones. The unexpected cross-reactivity of this M. leprae-specific DR2-restricted T cell epitope with a DR2 peptide may have to be considered in the design of subunit vaccines against leprosy. 相似文献

An ultrastructural immunostaining method that uses silver-intensified gold was combined with another procedure that uses biotin peroxidase conjugates to allow simultaneous identification of two neurotransmitter-related antigens in the central nervous system. Tyrosine hydroxylase-immunoreactive neurons labeled with silver-intensified gold could be differentiated at both light and electron microscopic levels from glutamate decarboxylase-immunoreactive neurons labeled with peroxidase. Cross reactivity of the second group of immunoreagents with the first group was reduced by the heavy metal silver shell formed around the colloidal gold immunoglobulin complex. With this dual pre-embedding method, peroxidase-stained axons containing the inhibitory neurotransmitter gamma-aminobutyric acid were found to synapse directly on silver-stained dopamine neurons in the rat dorsomedial hypothalamus. This approach can be used in combination with a post-embedding immunocytochemical colloidal gold procedure, allowing ultrastructural identification of three neurotransmitter-related antigens in the same tissue section. 相似文献

Tyrosine phosphorylation, regulated by protein tyrosine phosphatases (PTPs) and kinases (PTKs), is important in signaling pathways underlying tumorigenesis. A mutational analysis of the tyrosine phosphatase gene superfamily in human cancers identified 83 somatic mutations in six PTPs (PTPRF, PTPRG, PTPRT, PTPN3, PTPN13, PTPN14), affecting 26% of colorectal cancers and a smaller fraction of lung, breast, and gastric cancers. Fifteen mutations were nonsense, frameshift, or splice-site alterations predicted to result in truncated proteins lacking phosphatase activity. Five missense mutations in the most commonly altered PTP (PTPRT) were biochemically examined and found to reduce phosphatase activity. Expression of wild-type but not a mutant PTPRT in human cancer cells inhibited cell growth. These observations suggest that the mutated tyrosine phosphatases are tumor suppressor genes, regulating cellular pathways that may be amenable to therapeutic intervention. 相似文献

The heart responds to diverse forms of stress by hypertrophic growth accompanied by fibrosis and eventual diminution of contractility, which results from down-regulation of alpha-myosin heavy chain (alphaMHC) and up-regulation of betaMHC, the primary contractile proteins of the heart. We found that a cardiac-specific microRNA (miR-208) encoded by an intron of the alphaMHC gene is required for cardiomyocyte hypertrophy, fibrosis, and expression of betaMHC in response to stress and hypothyroidism. Thus, the alphaMHC gene, in addition to encoding a major cardiac contractile protein, regulates cardiac growth and gene expression in response to stress and hormonal signaling through miR-208. 相似文献

Hyperspectral visible near infrared reflectance spectroscopy (VNIRRS) and geostatistical methods are considered for precision
soil mapping. This study evaluated whether VNIR or geostatistics, or their combined use, could provide efficient approaches
for assessing the soil spatially and associated reductions in sample size using soil samples from a 32 ha area (800 × 400 m)
in northern Turkey. Soil variables considered were CaCO3, organic matter, clay, sand and silt contents, pH, electrical conductivity, cation exchange capacity (CEC) and exchangeable
cations (Ca, Mg, Na and K). Cross-validation was used to compare the two approaches using all grid data (n = 512), systematic selections of 13, 25 and 50% of the data and random selections of 13 and 25% for calibration; the remaining
data were used for validation. Partial least squares regression (PLSR) analysis was used for calibrating soil properties from
first derivative VNIR reflectance spectra (VNIRRS), whereas ordinary-, co- and regression-kriging were used for spatial prediction.
The VNIRRS-PLSR method provided better prediction results than ordinary kriging for soil organic matter, clay and sand contents,
(R
2 values of 0.56–0.73, 0.79–0.85, 0.65–0.79, respectively) and smaller root mean squared errors of prediction (values of 2.7–4.1,
37.4–43, 46.9–61, respectively). The EC, pH, Na, K and silt content were predicted poorly by both approaches because either
the variables showed little variation or the data were not spatially correlated. Overall, the prediction accuracy of VNIRRS-PLSR
was not affected by sample size as much as it was for ordinary kriging. Cokriging (COK) and regression kriging (RK) were applied
to a combination of values predicted by VNIR reflectance spectroscopy and measured in the laboratory to improve the accuracy
of prediction of the soil properties. The results showed that both COK and RK with VNIRRS estimates improved the predictions
of soil variables compared to VNIRRS and OK. The combined use of VNIRRS and multivariate geostatistics results in better spatial
prediction of soil properties and enables a reduction in sampling and laboratory analyses. 相似文献

At a porphyry copper-gold deposit in Bajo de la Alumbrera, Argentina, silicate-melt inclusions coexist with hypersaline liquid- and vapor-rich inclusions in the earliest magmatic-hydrothermal quartz veins. Copper concentrations of the hypersaline liquid and vapor inclusions reached maxima of 10.0 weight % (wt %) and 4.5 wt %, respectively. These unusually copper-rich inclusions are considered to be the most primitive ore fluid found thus far.
