本文简化了黄瓜DNA提取程序并探讨了黄瓜SSR反应程序及体系.反应体系25 μL:10×buffer 2.5 μL,2.5 mmol/L MgCl2 2.0 μL,2 mmol/L dNTP 2.5 μL,gDNA模板(10 ng/μL)2 μL,forward primer(10 pmol/μL)0.5 μL,reverse primer(10 pmol/μL)0.5 μL,ddH2O 14 μL,Taq DNA聚合酶(1 U/μL)1 μL.反应程序:95℃预变性5 min,94℃变性30 sec,49℃退火1 min,72℃延伸90 sec,循环次数35,72℃延伸5 min.     

An Overview on Fiberoptic Sensing Technology in Smart Civil Structure

The paper discuss the concept of fiberoptic smart civil structure,and analysis the principle of non destructive evaluation of civil structure with the help of embeded fiberoptic sensor in sensing the inner state and integrity of the structure.The typical experiment and development of fiberoptic sensing system in the health monitoring of dam,highway,bridge and buildings are overriewed.     

p38MAPK信号通路选择性调节FSH对甾体生成 的诱导作用研究

利用乙烯雌酚（DES）处理23日龄SD大鼠，分离卵巢颗粒细胞(GC)进行无血清培养。结果表明，FSH (50ng/m1) 处理GC，可迅速激活有丝分裂原蛋白激酶（p38 MAPK），FSH处理5min便可观察到磷酸化的p38 MAPK；FSH处理30min，磷酸化的p38 MAPK水平达到最高;向培养液中加入H89(蛋白激酶A抑制剂，10μM)，则显著抑制了FSH对p38 MAPK的激活作用，提示这种激活作用依赖于蛋白激酶A(PKA)。用 SB203580(p38MAPK抑制剂，20μM)抑制p38 MAPK激活，则进一步提高了FSH对孕酮和甾体生成快速调节蛋白（StAR）的诱导作用，同时降低了FSH对雌激素生成的促进作用(p<0.01)。RT-PCR结果显示：抑制p38 MAPK活性后，FSH对StAR mRNA刺激作用明显增强，但对细胞色素P450芳香化酶（P450arom）mRNA的诱导作用却减弱了(p<0.05)。激光共聚焦和蛋白印迹结果显示：在GC中，StAR蛋白主要分布在线粒体中；与对照组相比，FSH显著提高了StAR的荧光强度和蛋白水平；抑制p38MAPK活性则增强了FSH对StAR蛋白表达的诱导作用。     

ANALYSES FOR DEFECTS OF 18G_2ANb STEEL SPHERICAL TANKS OF NATURAL GAS

Calculations and analyses are made with the CVDA, IIW, JWES standards for the strength and welding defects of two spherical tanks of natural gas in CHONG QING. The results of the analyses indecate that though there are some problems with the welding quality in these tanks, they may still be used with the desingned presure. Now the formal working of these two spherical thanks has verified of the validity the calculations and analyses.     

A Globally Convergent Algorithm for Nonlinear Programming

A new globally convergent algorithm was presented for a continuous differen-tiable nonlinear programming by defining a measure function deviating from the Kuhn-Tucker point. With this algorithm which can be used to get the optimum solution of the problem,the optimal lagrangian multiplier corresponding the optimal solution of the problem was also obtained. A new iterative algorithm for quadratic programming is obtained when applying the general algorithm to quadratic programming. Finally,a numerical example was given.     

Influence of the Flood-Control-Projects on the Hydrodynamic State of the Rivers Confluence in Chongqing

Though the Flood-Control-Projects along Yantze River and Jialing River in Chongqing may well improve the urban traffic and construction,it may change the river hydrodynamic characters.There may be two consequences,one is to reduce the size of cross section and the other is to change the river shape,such as the curvature radius.The phenomena of the local backwater and velocity variety are obvious,which result in some problems for the navigation: sediment,transport and so on.In this paper, the study object is the Flood-Control-Projects and Yangtse River and Jialing River,and the aim is to study the influence of the Flood-Control-Projects on the hydrodynamic state of the two rivers.Using the numerical technique,with the two rivers hydrodynamic model the flow field's change of river after completion of Flood-Control-Projects is simulated and the reasons caused by the change are analyzed.     

粉花石斛组织培养快速繁殖

采用种子培养(胚培养)和茎节培养直接诱导成苗的方式进行了粉花石斛组织培养快速繁育试验.试验结果表明,无论是以种子还是以茎节作为外植体,均能诱导成苗.种子培养的最适培养基为N6 6-BA 5 NAA 0.4;茎节培养的最适培养基为I/2MS 6-BA 0.9 NAA 0.2;适宜的生根培养基分别为:N6 10%香蕉汁、1/2MS.移栽最适基质配方为腐叶土:珍珠岩为1:1.     

湿式多盘制动器的研究现状及展望

分别对国内外在湿式多盘制动器摩擦副间对流换热机理、摩擦衬片压力分布规律、摩擦偶件温度场与应力场的分析、摩擦衬盘与对偶钢盘的摩擦机理、制动噪声、制动过程的影响因素、整体散热特性及其试验研究等方面的研究现状进行了综述和评价，并对湿式多盘制动器的某些研究内容提出了新的见解，最后展望了湿式多盘制动器的研究方向及其发展动态。

概述了普通齿轮转子泵的结构及工作原理．介绍了标准型(三行星轮)复合齿轮转子泵的工作原理，分析了该泵的力学特性，推出了平均流量和瞬时流量公式，并对该泵的脉动率进行了分析，得出当工作轮的齿数为非3的倍数时，该泵的流量脉动将显著降低的结论。当泵的工作轮及腔体用特殊材料(如不锈钢或陶瓷)制作或处理时，该泵完全可以用在纯水液压系统。
