十种常用农药与球孢白僵菌的生物学相容性

球孢白僵菌孢子粉与10种常用农药相容性的测定结果显示,随着孢子浓度上升,所试农药对孢子的抑制作用均有不同程度的增强。在1/10田间常规使用浓度下,百菌清和代森锰锌均能抑制或杀死孢子(萌发率<1％)。除阿维菌素外,所有杀虫剂均与白僵菌孢子相容,在常规使用浓度的10倍稀释液中孢子萌发率达90％以上。吡虫啉、蚜虱灵、灭多威和氟虫腈与孢子的相容性最好,其中吡虫啉和蚜虱灵对孢子萌发率的影响不明显随药剂浓度的变化而变化,即使在田间常规使用浓度下孢子萌发率也在95％以上,而阿维菌素与白僵菌的相容性极差。因此,应用白僵菌制剂防治害虫,选择生物学相容性好的农药以低剂量与白僵菌制剂混用,既可使菌剂增效,又可大幅度降低化学药剂用量。     

变叶海棠种群多样性的形成与分化研究

 采用杂种指数和形象化散点图法, 探明了变叶海棠以兼性无融合生殖方式与陇东海棠和花叶海棠产生渗入杂交变异, 又以无融合生殖方式保持新产生变异的遗传, 经过数千万年的世代繁衍,形成现今这样极其复杂多样的渗入杂交群体, 该群体为3 个种的杂交复合体。由于杂种的全部或部分能育子代中发生的基因重组和增加基因的突变, 丰富了物种的基因库, 从而深入的揭示了变叶海棠具有多种高抗性的遗传机理。在变叶海棠居群内保护或栽植适量的陇东海棠和花叶海棠, 对于保持和促进变叶海棠多样性的形成与分化具有重要意义。     

入世后我国甜樱桃面临的机遇与挑战及发展对策

甜樱桃作为一种早熟、优质、保健果品,而且种植效益较高,在国内国际都正处于发展时期。根据世界甜樱桃生产和贸易情况,指出了我国在甜樱桃科学技术与生产上与世界先进甜樱桃生产国的差距,分析了我国加入WTO之后,我国甜樱桃面临的机遇和挑战,并且机遇大于挑战。如果有适当的科技投入,我国甜樱桃产业将可克服现在科技滞后的状态,发挥我国劳动力低廉和国土辽阔的优势,迅速扩大其生产总量,提高品质,扩大出口,增加农民的收入,2015年可望形成32.4～43.2亿元的社会总产值。     

Preparation of gfp-bcl-XL-contained recombinant adenovirus vector by the homologous recombination in bacteria

AIM: To prepare gfp-bcl-X_L-contained recombinant adenovirus(rAd-gfp-bcl-X_L).METHODS: Bcl-X_L gene was amplified from pEGFP-C₃-bcl-X_L, subcloned into shuttle plasmid and formed transfer plasmid of pAdTrack-CMV-bcl-X_L. Then pAdTrack-CMV-bcl-X_L was linealinzed with PmeI and co-transformed into BJ5183 bacteria with adenovirus genomic plasmid of pAdEasy-1. The identified recombinant adenovirus plasmid was digested with PacI and transfected into 293 cells to package recombinant adenovirus particles. The target gene was detected by PCR.RESULTS: There were about 35% positive recombinant bacterial clones after the co-transformation of pAdTrack-CMV-bcl-X_L and pAdEasy-1 into BJ5183. Recombinant adenovirus particle were produced and further amplified after the transfection of pAdEasy-1-gfp-bcl-X_L into 293 cells. PCR test indicated that the recombinant Ad contained bcl-X_L gene. The titer of the purified rAd-gfp-bcl-X_L was 6.5×10¹² PFU/L. CONCLUSIONS: The homologous recombination in bacteria is a convenient and high efficient method to prepare rAd-gfp-bcl-X_L. This affords a good gene transfer vector for the gene therapy in human’s diseases.     

Effects of PD98059 on the differentiation from mesenchymal stem cells to osteoblasts

AIM: To investigate the effects of PD98059 on the differentiation from mesenchymal stem cells to osteoblasts.METHODS: hMSC were separated from human marrow and expanded in cuture medium. hMSC were induced with dexamethasone, β-glycerophosphate, vitamin C which acted as osteoblast differentiation inducer. PD98059 was added into the osteoblasts induction medium. The cells were assayed with cell morphology, alkaline phosphatase (AP) activity and calcium deposition. RESULTS: The isolated cultured MSC comprised a single phenotypic population and displayed a fibroblast-like morphology. After induced with osteoblasts induction medium, the cells showed changes in cell morphology from spindle-shape to cuboidal and polygonal. The AP activity increased gradually and reached the peak in 12 days, then decreased. Many scattered tangerice calcium nodes were observed. PD 98059 significantly inhibited AP activity and calcium deposition in a dose-dependent manner. A striking observation of the present study was that a few adipocytes appeared in cultures that were treated with PD 98059 and osteogenic differentiation medium. CONCLUSION: These results indicated that osteogenic diferentiation from the hMSCs was related to the activation of the ERK.     

Changes of intracellular potassium activity and membrane potential in guinea pig ventricular myocardium during superfusion with low sodium solutions and effects of low pH

AIM:To investigate the changes in intracellular potassium activity(aⁱK) and membrane potential(V_m) induced by low external sodium infusion (Low [Na]_o) and to detect the mechanisms involved and the relationship between aⁱK and V_m. METHODS:aⁱK and V_m were measured in infusion with different sodium concentrations using methods of convenient and ion selective microelectrodes in guinea pig ventricular myocardium. RESULTS:Low [Na]_o resulted in a decrease in aⁱK and an increase in V_m in a Na⁺ concentration-dependent manner.At the same time,contraction and resting potential increased, and action potential duration decreased markedly,but action potential amplitude was not affected. A change of the pH from 7.4 to 7.0 in low [Na]_o solution reduced the decrease in aⁱK, but did not affect the increase in V_m.CONCLUSION:A better linear relationship appeared between the changes in aⁱk and [Na]_o or in V_m and [Na]_o,while during each low [Na]_o the change in both aⁱK or V_m may reach a new balance level.     

早熟优质长豇豆新品种‘鄂豇豆2 号’

 ‘鄂豇豆2号’是经有性杂交选育而成的早熟蔓生豇豆新品种。该品种荚浅绿色, 长60 cm,单荚质量18~ 24 g。早熟, 荚商品性状优良, 产量高, 对光周期反应不敏感, 田间疫病、煤霉病、病毒病发病率低, 适宜于春、夏、秋各季栽培, 已在全国大部分省份推广种植成功。     

杜鹃花新品种‘朝晖’和‘红晕’

 ‘朝晖’杜鹃花是由大白花杜鹃和马缨花杂交选育而成, 叶面灰绿色, 顶生圆锥伞房花序, 有花10～12 朵, 花冠紫红色。‘红晕’杜鹃花是由露珠杜鹃和马缨花杂交选育而成, 叶浅绿色, 花冠漏斗状钟形, 花冠上部粉红色, 下部乳黄色, 有紫红色斑点。     

Comparison of two sampling methods for leafhoppers (Homoptera, Cicadellidae) associated with sesame in the east Mediterranean region of Turkey

Yellow sticky-traps and a modified Johnson-Taylor suction-trap were used to index the relative population dynamic of leafhoppers associated with sesame in the east Mediterranean region of Turkey. Comparison of the obtained results signified important differences between both traps. 32 leafhoppers species were caught in the suction-trap while only 18 species were determined on yellow sticky-traps. For most of the leafhopper species, relatively more males were determined on the yellow-traps than in the suction-trap, indicating a higher activity of male leafhoppers during daytime hours. A close relationship between the seasonal flight pattern as indexed by sticky-traps and by suction-trap was only observed forAsymmetrasca decedens (Paoli) andEmpoasca decipiens Paoli. For most other leafhopper species no or only a very poor correlation for the relative population dynamic was determined between both traps. Important leafhopper vector species, e.g.,Circulifer haematoceps (Mulsant et Rey) andOrosius orientalis (Matsumura) were better represented in yellow sticky-trap catches than they were in the suction trap.

Zusammenfassung  Die relative Populationsdynamik der mit Sesam assozüerten Zwergzikaden wurde mit beleimten Gelbtafeln und einer stationären Johnson-Taylor-Saugfalle an der südöstlichen Mittelmeerküste der Türkei untersucht. Beim Vergleich beider Methoden zeigten sich deutliche Unterschiede zwischen den Fallentypen. In der Saugfalle konnten 32 Zwergzikadenarten erfaßt werden, während dies auf den Gelbtafeln nur 18 Arten waren. Die meisten Zwergzikadenarten zeigten auf den Gelbtafeln ein stark in Richtung der Männchen verschobenes Geschlechterverhältnis, was eine erhöhte Flugaktivität der Männchen während des Tageslichts vermuten läßt. Auch der Verlauf der saisonalen Flugaktivität war für die meisten Zwergzikadenarten zwischen den beiden Fallentypen sehr unterschiedlich, und es konnten zwischen Saug- und Gelbfalle keine deutlichen Korrelationen hergestellt werden. Nur für die beiden zusammen erfaßten ArtenAsymmetrasca decedens (Paoli) undEmpoasca decipiens Paoli wurde ein enger Zusammenhang zwischen der mit der Saugfalle und den gelben Leimtafeln erfaßten relativen Populationsdynamik festgestellt. Wichtige Vektorenarten unter den Zwergzikaden, z. B.Circulifer haematoceps (Mulsant et Rey) undOrosius orientalis (Matsumura) waren auf den Gelbtafeln besser repräsentiert als in der Saugfalle.
