Among marine organisms, sponges are the richest sources of pharmacologically-active compounds. Stemming from a previous lead discovery program that gathered a comprehensive library of organic extracts of marine sponges from the off-shore region of Portugal, crude extracts of Erylus cf. deficiens collected in the Gorringe Bank (Atlantic Ocean) were tested in the innovative high throughput screening (HTS) assay for inhibitors of indoleamine 2,3-dioxygenase (IDO) and showed activity. Bioassay guided fractionation of the dichloromethane extract led to the isolation of four new glycolipids, named erylusamide A–D. The structures of the isolated compounds were established by 1D and 2D nuclear magnetic resonance (NMR) spectroscopy, high-resolution electrospray ionization mass spectrometry (HR-ESI-MS) and chemical derivatization. The metabolites shared a pentasaccharide moiety constituted by unusual highly acetylated d-glucose moieties as well as d-xylose and d-galactose. The aglycones were unprecedented long chain dihydroxyketo amides. Erylusamides A, B and D differ in the length of the hydrocarbon chain, while erylusamide C is a structural isomer of erylusamide B.  相似文献

Purpose

The main objective of the present study was to evaluate the toxicity of two reference chemicals, Carbendazim and Phenmedipham, for the compostworm Eisenia andrei (effects of Carbendazim) and the potworm Enchytraeus crypticus (effects of Phenmedipham) in 12 Mediterranean soils with contrasting soil properties. The observed toxicity was also compared to that obtained for OECD standard soil, used as a control.

Materials and methods

The soils were selected to be representative for the Mediterranean region and to cover a broad range of soil properties. The evaluated endpoints were avoidance behavior and reproduction. Soils were also assembled in two groups according to their pedological properties.

Results and discussion

Toxicity benchmarks (AC50s) obtained for E. andrei avoidance behavior in carbendazim-contaminated soils were generally higher for sandy soils with low pH. The toxic effects on the reproduction of the compostworms were similar in the six tested soils, indicating a low influence of soil properties. The avoidance response of E. crypticus towards Phenmedipham was generally highly variable in all tested soils. Even though, a higher toxicity was observed for more acidic soils. The EC50s for reproduction of the latter species varied by a factor of 9 and Phenmedipham toxicity also tended to be increasing in soils with lower pH, except for the soils with extreme organic matter content (0.6 and 5.8%).

Conclusions

A soil effect on chemical toxicity was clearly confirmed, highlighting the influence that test soils can have in site-specific ecological risk assessment. Despite some relationships between soil properties and toxicity were outlined, a clear and statistically significant prediction of chemical toxicity could not be established. The range of soil properties was probably narrow to give clearer and more consistent insights on their influence. For the four groups of tests, the toxicity observed for OECD soil was either similar, lower, or generally higher if compared with Mediterranean soils. Moreover, it did represent neither the organic matter content found in Mediterranean soils nor their textural classes.  相似文献

The effects of Trametes hirsuta laccase and Pentopan Mono BG xylanase and their combination on oat, wheat, and mixed oat-wheat doughs and the corresponding breads were investigated. Laccase treatment decreased the content of water-extractable arabinoxylan (WEAX) in oat dough due to oxidative cross-linking of feruloylated arabinoxylans. Laccase treatment also increased the proportion of water-soluble polysaccharides (WSNSP) apparently due to the beta-glucanase side activity present in the laccase preparation. As a result of the laccase treatment, the firmness of fresh oat bread was increased. Xylanase treatment doubled the content of WEAX in oat dough and slightly increased the amount of WSNSP. Increased stiffness of the dough and firmness of the fresh bread were detected, probably because of the increased WEAX content, which decreased the amount of water available for beta-glucan. The combination of laccase and xylanase produced slight hydrolysis of beta-glucan by the beta-glucanase side activity of laccase and enhanced the availability of AX for xylanase with concomitant reduction of the amount and molar mass of WSNSP. Subsequently, the volume of oat bread was increased. Laccase treatment tightened wheat dough, probably due to cross-linking of WEAX to higher molecular weight. In oat-wheat dough, laccase slightly increased the proportion of WSNSP between medium to low molecular weight and increased the specific volume of the bread. Xylanase increased the contents of WEAX and WSNSP between medium to low molecular weight in oat-wheat dough, which increased the softness of the dough, as well as the specific volume and softness of the bread. The results thus indicate that a combination of laccase and xylanase was beneficial for the textures of both oat and oat-wheat breads.  相似文献

Understanding how cool stars produce magnetic fields within their interiors is crucial for predicting the impact of such fields, such as the activity cycle of the Sun. In this respect, studying fully convective stars enables us to investigate the role of convective zones in magnetic field generation. We produced a magnetic map of a rapidly rotating, very-low-mass, fully convective dwarf through tomographic imaging from time series of spectropolarimetric data. Our results, which demonstrate that fully convective stars are able to trigger axisymmetric large-scale poloidal fields without differential rotation, challenge existing theoretical models of field generation in cool stars.  相似文献

Early season leaf growth depends largely on nitrogen (N) provided by remobilization from storage, and many studies have tested the effect of N availability to roots on the amount of N provided for new leaf development by remobilization. Although it is well known that the light regime experienced by a leaf influences the amount of N per unit leaf area (LA), the effect of the local light regime on the amount of N derived either directly from root uptake or from remobilization for early season leaf growth has never been tested at an intra- canopy scale. The objective of this study was to quantify the relative importance of (1) N availability to roots, (2) local light regime experienced by the foliage (at the shoot scale) and (3) leaf rank along the shoot, on the total amount of N allocated to leaves and on the proportions of N provided by remobilization and root uptake. To quantify the importance of N uptake and remobilization as sources of leaf N, potted hybrid walnut trees (Juglans nigra L. x regia L.) were grown outdoors in sand and fed with a labeled ((15)N) nutrient solution. By removing the apical bud, the trees were manipulated to produce only two shoots. The experimental design had two factors: (1) high (HN; 8 mol N m(-3)) and low (LN; 2 mol N m(-3)) N availability; and (2) high (HL; 90% of incident photosynthetically active photon flux (PPF)) and low (LL; 10% of incident PPF) light. Total leaf N per tree was unaffected by either N availability or irradiance. The HN treatment increased the amount of leaf N derived from root uptake at the whole-tree scale (typically around 8 and 2% in the HN and LN treatments, respectively). Nitrogen allocation within foliage of individual trees was controlled by the local light regime, which strongly affected individual leaf characteristics as leaf mass per unit LA and area- based amount of leaf (N(a)). Decreasing the light availability to a branch decreased the amount of N allocated to it, benefiting the less shaded branches. In contrast, shading of the lower branch did not affect the fraction of total leaf N remobilized for either the lower, shaded branch or the upper, unshaded branch. The relevance of these findings for tree growth modeling is discussed.  相似文献

Pulp and paper industries face serious environmental challenges, especially with regard to the conservation of water resources. Chemical thermal mechanical pulping (CTMP) is a process of pulping that combines chemical and mechanical pulping. This reduces the volume of water used in the process. But on the other hand, CTMP generates an effluent with high concentration of organic matter and is difficult to treat. This study evaluated the efficiency in the combination of physicochemical pretreatment by coagulation-flocculation-sedimentation (CFS) process and advanced oxidation process (AOP) by Fenton in sequence to treat CTMP effluent of a Brazilian industry. At first, the best treatment conditions for this type of effluent were determined. To evaluate the efficiency, pH, chemical oxygen demand, biochemical oxygen demand, total organic carbon, lignin contents, color, total phenolic contents, turbidity, and solids were measured before and after treatment. The acute toxicity on Daphnia magna was also determined. The treatment with CFS showed better results in the removal of solids and Fenton in the removal of recalcitrant compounds, such as lignin, demonstrating the need to use them in sequence. Combining CFS and Fenton to treat CTMP effluent allowed to achieve a removal efficiency of 95% for TOC, 61% for COD, and 76% for lignin contents.  相似文献
