AIM:To investigate the kinetics of interleukin-6 (IL-6) protein and mRNA expressions in the co-culture of rat brain vascular endothelial cells(VECs) with fibrin. METHODS:The IL-6 concentration were measured by rat IL-6 ELISA kit and the IL-6 mRNA was measured by real-time PCR after development an in vitro model of in situ fibrin polymerization on rat brain vascular endothelial cells. RESULTS:Fibrin induced IL-6 expression. The IL-6 antigen concentration increased significantly in 1.0 g/L media group compared to the low dose fibrin group or control media group (P<0.01). Fibrin-induced IL-6 expression in rat brain VECs as early as 2 h post-fibrin stimulation, increased significantly at 8 h, keeping at high level till 24 h (P<0.05). Collagen did not induce IL-6 expression at 24 h. Real-time PCR analysis showed that IL-6 antigen increase in fibrin treated rat brain VECs was due to fibrin induced elevation of steady state mRNA expression. CONCLUSION:Fibrin is a potent vascular endothelial cell activator. It induces IL-6 expression in time and dose dependent fashions. 相似文献

采用硫酸阿米卡星可溶性粉40、50和70mg/L三个治疗浓度,对人工诱发的鸡大肠杆菌和沙门氏菌病进行治疗试验。通过15d试验,其0.05mL、0.1mL两个攻毒组,用40mg/L治疗鸡大肠杆菌和沙门氏菌病,平均死亡率为6.6%和3.3%;50mg/L和70mg/L治疗的死亡率均为0;对照组的平均死亡率分别为43.3%和60%。由此可见,治疗组鸡的死亡率之间虽有差异,但差异不显著(P〉0.05);与对照组相比较则差异极显著(P〈0.01)。鸡的增重结果也呈上述规律。说明硫酸阿米卡星可溶性粉对鸡大肠杆菌O2 株感染和鸡沙门氏菌C79—1感染,均具有良好的治疗效果。 相似文献

AIM:To establish and compare the methods for culturing neonatal rat cardiac fibroblasts culture. METHODS:Neonatal rat hearts were isolated by collagenase+trypsin or trypsin digestion. The cardiomyocytes and cardiac fibroblasts were isolated by different attachment techniques. The cellular morphology, purity and reactions to the reagent were tested. RESULTS:For morphology and purity, no difference between the 2 methods was observed, though more myocytes were harvested by the method of collagenase+trypsin digestion. For the cellular responses to reagent, the cardiac fibroblasts harvested with trypsin digestion had more potent proliferative ability than those with collagenase+trypsin digestion. CONCLUSION:There is no difference of cellular morphology and purity in the cardiac fibroblasts isolated by collagenase+trypsin digestion and trypsin digestion, but the fibroblasts with trypsin digestion have more potent proliferative ability. 相似文献

In order to analyze the effect of listeriolysin S (LLS) llsB gene deletion on the biological characteristics of Listeria monocytogenes (LM),this study used homologous recombination to construct the llsB gene deletion strain LM90-ΔllsB,and the biological characteristics of growth characteristics,median lethal dose (LD50) and organ-borne bacteria were studied in healthy Kunming male mice at 8 weeks old and weighing 40 g±5 g.The llsB gene deletion strain was successfully constructed,and the deletion strain had good genetic stability through continuous passage to 20 generations in vitro.Based on its growth curve examination,we found that the growth rate of the mutant strain was slightly higher than that of the parent strain.The results of mice infection test showed that the LD50 of the parent strain and the deletion strain were 106.17 and 106.50 CFU,respectively.Compared with the parent strain,the amount of bacteria load of the deletion strain in the liver and spleen of the mice was extremely significantly decreased (P<0.01),the results showed that the infection ability of the mutant strain on mice was obviously weakened.No Listeria monocytogenes was detected in the brain.The results suggested that llsB gene might have direct or indirect regulatory effect on some biological characteristics of LM90,and it would provide a theoretical basis for further understanding of the pathogenic mechanism of LLS,prevention and control of listeriosis. 相似文献

旨在探究风柜斗草提取物(Sarcopyramis napalensis Wall extract,SNE)对蛋氨酸胆碱缺乏(MCD)饮食诱导的肝脏脂质沉积的作用与机制。本研究将60只雄性C57BL/6小鼠随机分为6组,每组10只,设置MCS组(MCD饲料的对照饲料组)、MCS+风柜斗草提取物组(400 mg·(kg·bw)-1)、MCD组、MCD+风柜斗草提取物低、中、高剂量组(100、200、400 mg·(kg·bw)-1),造模及药物干预6周。进行肝组织切片HE染色、天狼猩红染色,检测血清碱性磷酸酶(AKP)、谷草转氨酶(AST)、谷丙转氨酶(ALT),肝组织谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活力水平及丙二醛(MDA)含量以评估肝脂质沉积、肝纤维化以及肝抗氧化情况。使用Western blot技术检测腺苷酸激活蛋白激酶(AMPK)蛋白的激活情况,使用qRT-PCR技术检测固醇调节元件结合蛋白-1C (SREBP-1c)、肝型脂肪酸结合蛋白(L-FABP)、脂肪酸转运蛋白-3(FATP-3)、脂肪酸转运蛋白-4(FATP-4)、细胞间黏附分子-l (ICAM-1)、血管细胞黏附分子-l (VCAM-1)、基质金属蛋白酶-2(MMP-2)、组织金属蛋白酶抑制剂-2(TIMP-2) mRNA表达水平,同时采用LC-MS/MS技术进行肝组织非靶向脂质组学分析。结果表明,给予200、400 mg·(kg·bw)-1剂量的风柜斗草提取物时均可显著改善MCD饲料饲喂的小鼠肝脂肪变性,减少炎症簇以及肝纤维化程度。给予风柜斗草提取物100、200、400 mg·(kg·bw)-1时均可显著降低MCD饲料饲喂的小鼠血清中ALT和AKP的活力以及肝组织中MDA的含量,显著提高小鼠肝中GSH-Px的活力。Western blot及qRT-PCR检测结果显示,风柜斗草提取物显著激活MCD饲料饲喂的小鼠肝组织AMPK;显著抑制MCD饲料饲喂的小鼠肝组织SREBP-1c、L-FABP、FATP-3、FATP-4、ICAM-1、VCAM-1、MMP-2、TIMP-2的mRNA表达水平。肝脂质组学检测结果显示,风柜斗草提取物显著降低MCD饲料饲喂的小鼠肝中甘油三酯(TG)、磷脂酰甘油(PG)以及神经酰胺(Cer)的相对含量;显著提高鞘磷脂(SM)以及辅酶Q9相对含量。结果提示,风柜斗草提取物可通过AMPK/SREBP-1c途径抑制TG合成,以及通过抑制FATP与L-FABP表达来减少肝对脂肪酸的摄取,从而改善MCD饲料饲喂的小鼠肝脂肪变性;通过提高MCD饲料饲喂的小鼠肝组织中SM和辅酶Q9相对含量以及降低Cer相对含量,改善肝细胞氧化应激损伤情况,进而改善MCD饮食诱导的小鼠肝炎症和纤维化,最终达到改善肝脂质沉积症的作用。 相似文献
