Black spot disease, which is caused by the Japanese pear pathotype of the filamentous fungus Alternaria alternata (Fries) Keissler, is one of the most harmful diseases in Japanese pear cultivation. We mapped a gene for susceptibility to black spot disease in the Japanese pear (Pyrus pyrifolia Nakai) cultivar ‘Kinchaku’ (Aki gene) at the top of linkage group 11, similar to the positions of the susceptibility genes Ani in ‘Osa Nijisseiki’ and Ana in ‘Nansui’. Using synteny-based marker enrichment, we developed novel apple SSR markers in the target region. We constructed a fine map of linkage group 11 of ‘Kinchaku’ and localized the Aki locus within a 1.5-cM genome region between SSR markers Mdo.chr11.28 and Mdo.chr11.34. Marker Mdo.chr11.30 co-segregated with Aki in all 621 F1 plantlets of a ‘Housui’ × ‘Kinchaku’ cross. The physical size of the Aki region, which includes three markers (Mdo.chr11.28, Mdo.chr11.30, and Mdo.chr11.34), was estimated to be 250 Kb in the ‘Golden Delicious’ apple genome and 107 Kb in the ‘Dangshansuli’ Chinese pear genome. Asian soybean rust (ASR) caused by Phakopsora pachyrhizi is one of the most serious soybean (Glycine max) diseases in tropical and subtropical areas. A soybean line, PI 587855, showed a resistance phenotype against ASR pathogens in Japan and South America at high frequency; however, little is known of the genetic control of this resistance and chromosomal location of the corresponding locus. Therefore, the aim of this study was to study the inheritance of PI 587855 resistance and map the corresponding locus with SSR markers aiming to use the linked markers in marker‐assisted selection. In the segregating population, resistance to ASR appeared to be controlled by a single dominant gene. The ASR resistance locus was mapped near to the chromosomal region where the resistant loci, Rpp1 and Rpp1‐b, were previously mapped. Comparative genetic mapping and disease reaction profiles of other seven lines carrying Rpp1 or Rpp1‐b to four Brazilian ASR isolates revealed that the resistance reaction exhibited by PI 587855 was similar to that of Rpp1‐b‐carrying varieties which have useful resistance to South American ASR strains. 相似文献

Lotus japonicus was used to study the distribution and interconnections of 15 elements in plant tissues, including essential and non-essential elements: boron (B), sodium (Na), magnesium (Mg), potassium (K), calcium (Ca), manganese (Mn), iron (Fe), nickel (Ni), copper (Cu), zinc (Zn), arsenic (As), strontium (Sr), molybdenum (Mo), cadmium (Cd) and cesium (Cs). Large amounts of B and Ca accumulated in plant leaves, while Fe, Na, Ni, As and Cd tended to mainly occur in the roots, and Mo was the only element to accumulate in the stems. The elemental compositions within plants were severely disturbed by treatment with toxic elements. Competition between element pairs in the same group (e.g. K and Cs; Ca and Sr) was not found. Iron, Cu and Zn accumulation were induced by Cd and Ni addition. When natural variants grew in a nutrition solution with subtoxic levels of As, Cd, Cs, Ni, Mo and Sr, intriguing relationships between the elements (such as Fe, As and K; Mg and Ni; Mn and Ca) were revealed using principal-component analysis. This study on the plant ionome offers detailed information of element interactions and indicates that chemically different elements might be closely linked in uptake or translocation systems. 相似文献

The body size of juvenile fish is often used as an index of growth rate, which in turn is influenced by local habitat conditions. We evaluated the size and origin of juvenile Chum salmon Oncorhynchus keta in the coastal areas of three regions (Atsuta, Shari, and Konbumori) of Hokkaido, northern Japan. The origin of the individuals in these communities differed between Konbumori and the other two regions. The former contained juveniles that originated from both the nearest stocked river and other rivers that were outside the area of interest. Conversely, the communities in Atsuta and Shari consisted exclusively of fish from the nearest stocked river. Moreover, the juveniles in Konbumori were larger than those in Atsuta and Shari. The results of our otolith analysis suggest that the larger size of the fish in Konbumori was due to the immigration of large individuals from natal rivers at distant locations. Thus, immigrants were likely to enlarge their body size composition in the area of interest. In summary, if the distance from a natal river is adjusted and daily growth is taken into account, body size can be used as an indicator of growth performance in coastal juvenile chum salmon. 相似文献

Elastomeric proteins occur in the shell ligament of bivalve molluscan shellfish. A newly identified protein named ligament protein-1, with a molecular mass of approximately 12 kDa, was extracted from the shell ligament of the scallop, Patinopecten yessoensis, with 70 % formic acid, and purified by gel filtration column chromatography and reverse-phase C18 column chromatography. Although the amino acid composition of this protein differs from that of the known shell ligament protein abductin, the partial amino acid sequences are similar to those of abductin. CD spectra and FT–IR spectra analyses showed that ligament protein-1 retains its secondary structures of α-helix, β-turn, and β-sheets. To the best of our knowledge, this is the first study on protein purification from the shell ligament of a bivalve molluscan shell. 相似文献

Amylose content is one of the most important factors influencing the physical and chemical properties of starch in rice. Analysis of 352 Vietnamese rice cultivars revealed a wide range of variation in apparent amylose content and the expression level of granule-bound starch synthase. On the basis of single-nucleotide polymorphisms (SNP) at the splicing donor site of the first intron and in the coding region of the granule-bound starch synthase I gene, Waxy gene, alleles can be classified into seven groups that reflect differences in apparent amylose content. The very low and low apparent amylose content levels were tightly associated with a G to T in the first intron whereas intermediate and high amylose was associated with a T genotype at SNP in exon 10. The correlation between the combination of T genotype at SNP in the first intron, C in exon 6, or C in exon 10 was predominant among low amylose rice varieties. Our analysis confirmed the existence of Wxop allele in Vietnamese rice germplasm. The results of this study suggest that the low amylose properties of Vietnamese local rice germplasm are attributable to spontaneous mutations at exons, and not at the splicing donor site. 相似文献

Although MRI has become widely used in small animal practice, little is known about the validity of advanced MRI techniques such as diffusion‐weighted imaging and diffusion tensor imaging. The aim of this retrospective analytical observational study was to investigate the characteristics of diffusion parameters, that is the apparent diffusion coefficient and fractional anisotropy, in dogs with a solitary intracranial meningioma or histiocytic sarcoma. Dogs were included based on the performance of diffusion MRI and histological confirmation. Statistical analyses were performed to compare apparent diffusion coefficient and fractional anisotropy for the two types of tumor in the intra‐ and peritumoral regions. Eleven cases with meningioma and six with histiocytic sarcoma satisfied the inclusion criteria. Significant differences in apparent diffusion coefficient value (× 10?3 mm2/s) between meningioma vs. histiocytic sarcoma were recognized in intratumoral small (1.07 vs. 0.76) and large (1.04 vs. 0.77) regions of interest, in the peritumoral margin (0.93 vs. 1.08), and in the T2 high region (1.21 vs. 1.41). Significant differences in fractional anisotropy values were found in the peritumoral margin (0.29 vs. 0.24) and the T2 high region (0.24 vs. 0.17). The current study identified differences in measurements of apparent diffusion coefficient and fractional anisotropy for meningioma and histiocytic sarcoma in a small sample of dogs. In addition, we observed that all cases of intracranial histiocytic sarcoma showed leptomeningeal enhancement and/or mass formation invading into the sulci in the contrast study. Future studies are needed to determine the sensitivity of these imaging characteristics for differentiating between these tumor types. 相似文献

Acid sulfate, peat, sandy podzolic, and saline soils are widely distributed inthe lowlands of Thailand and Malaysia. The nutrient concentrations in theleaves of plants grown in these type of soils were studied with the aim ofdeveloping a nutritional strategy for adapting to such problem soils. In sagoand oil palms that were well-adapted to peat soil, the N, P, and Kconcentrations were the same in the mature leaves, while the Ca, Mg, Na,and Fe concentrations were higher in the mature leaves of the oil palm thanof the sago palm. Melastoma malabathricum and Melaleuca cajuputi plantsthat were well-adapted to low pH soils, peat, and acid sulfate soils were alsostudied. It was observed that a high amount of Al accumulated in the M.marabathricum leaves, while Al did not accumulate in M. cajuputi leaves.M. cajuputi plants accumulated large amounts of Na in their leaves or stemsregardless of the exchangeable Na concentration in the soil, while M.malabathricum that was growing in saline-affected soils excluded Na.Positive relationships between macronutrients were recognized between Pand N, between K and N, and between P and K. Al showed antagonisticrelationships with P, K, Ca, Mg, Fe, Zn, Cu, and Na. Na also showedantagonistic relationships with P, K, Zn, Mn, Cu, and Al.
