Summary A linkage map for watermelon (Citrullus lanatus) was constructed on the basis of RADP, ribosomal DNA restriction fragment length polymorphism (RFLP), isozyme, and morphological markers using F1BC1. A segregating population of 78 individuals was the result of a backcross of a cultivated inbred line (H-7; Citrullus lanatus; 2n=22) and a wild form (SA-1; C. lanatus; 2n=22), in which the latter was the recurrent (male) parent. A total of 69 RAPD, one RFLP, one isozyme, and three morphological markers was found to segregate in the BC1 population. Linkage analysis revealed that 62 loci could be mapped to 11 linkage groups that extended more than 524 centimorgans (cM), while 12 loci segregated independently of all other markers. The locus for exocarp color was linked to two RAPD markers within a region of 5 cM on linkage group 4. The locus for flesh color was linked to a RAPD marker within a region of 30 cM on linkage group 6. The isozyme marker GOT was located on the linkage group 1. Linkage group 2 contained a locus for ribosomal DNA within 5 cM of a RAPD marker. Half of the RAPD markers on the linkage group 7 displayed severely distorted segregation. The construction of linkage map using molecular markers is necessary for the breeding of watermelon to introduce useful gene of wild watermelon efficiently. Reversible decrease in CO2 fixation has been reported in rice plants exposed to low concentrations of SO2 (Matsuoka et al., 1969). Alpha hydroxy sulfonate is thought to form in leaves by an addition-reaction between plant aldehyde and SO2, and to inhibit the process of the photosynthesis. However, the identification of this compound in the leaves has not been successful. This report deals with the results of the radiochemical experiments to examine the occurrence of glyoxylate bisulfite, a-hydroxy sulfonate forms of glyoxylic acid in rice plant leaves exposed to radioactive sulfur dioxide. In plants exposed to SO2, sufficient amounts of glyoxylate bisulfite could be formed and thereby inhibit the progress of the path from glicolic to glyoxylic acid.  相似文献

Effects of daily feeding frequency, water temperature, and stocking density on the growth of tiger puffer, Takifugu rubripes, fry were examined to develop effective techniques to produce tiger puffer in a closed recirculation system. Fish of 4, 14, and 180 g in initial body weight were fed commercial pellet diets once to five times a day to apparent satiation each by hand for 8 or 12 wk at 20 C. Daily feeding frequency did not affect the growth of 14‐ and 180‐g‐size fish. However, the daily feed consumption and weight gain of the 4‐g‐size fish fed three and five times daily were significantly higher than those of fish fed once daily (P < 0.05). Fish of 4 and 50 g in initial body weight were reared with the pellet diet at 15–30 C for 8 wk. The weight gain of fish increased with increasing water temperature up to 25 C and decreased drastically at 30 C for both sizes. Similar trends were observed for feed efficiency, although 4‐g fish had highest efficiency at 20 C. Effects of stocking density on growth were examined with fish of 8, 13, and 100 g in initial body weight. Fish were reared with the pellet diet for 8 or 16 wk at 20 C. Fish were placed in floating net cages in the culture tank, and the stocking density was determined based on the total weight of fish and volume of the net cage. Fish of 8 g in body weight grew up to 35–36 g during the 8‐wk rearing period independent of the stocking density of 8, 15, and 31 kg/m3 at the end of rearing. Final biomass per cage reached 32, 60, and 115 kg/m3 for 13‐g‐size fish, and 10, 18, and 35 kg/m3 for 100‐g‐size fish, and the growth of the fish tended to decrease with increasing stocking density for both sizes.  相似文献

The study was performed to determine whether orally administered KP102 (also known as GHRP-2) stimulates GH release in adult goats, and how the orally administered KP102 passes through the digestive tract and stimulates GH release in ruminant animals. Five mg/kg body weight (BW) of KP102 dissolved in 9 ml of saline were administered into the oral cavity, rumen, omasum and duodenum of adult goats, and GH release after administration of KP102 was examined. The GH levels were significantly elevated at 20 min after administration of KP102 into the oral cavity, and plasma concentrations of GH remained significantly elevated until 60 min (P < 0.05). The GH levels after administration of KP102 into the abomasum were variable. However, the GH level tended to increase within 30 min after administration, and were significantly higher than those of controls after 120 to 150 min (P < 0.05). The GH levels after administration of KP102 into the duodenum were significantly elevated at 40 min after administration, and plasma concentrations of GH remained significantly elevated until 140 min (P < 0.05). The administration of KP102 into the rumen failed to stimulate GH release. The GH response curves (AUC) produced after administration of KP102 into the abomasum or duodenum were 2.2-fold greater than those for after administration into the oral cavity (P < 0.05). The oral administration of 5 mg/kg BW of KP102 in the powder state, not dissolved in 9 ml of saline, failed to stimulate GH release. These results suggested that orally administered KP102 dissolved in saline transiently stimulates GH release in adult goats, and this phenomenon might be due to small amounts of the peptides entering directly into the abomasum with liquid bypassing the rumen.  相似文献

ABSTRACT:   The installation depth of the hook in longline fishing gear has previously been measured with micro depth loggers. Research that assumes the catenary shape of longline fishing gear by a simulation based on these data has been done. However, it was not known whether the branch line was tangled with the main line or flowed with the current. In this research, an ultrasonic positioning system generally used to investigate the underwater behavior of marine organisms, and a buoy with a communications satellite, have been used, and the 3-dimensional underwater shape of tuna longline fishing gear was measured. It was possible to monitor changes in fishing gear in real time. The possibility of high precision measurement was suggested with future technical improvement.  相似文献

The aim of the present study was to clarify the effect of melatonin (MEL) on the salsolinol (SAL)‐induced release of prolactin (PRL) in goats. Female goats were kept at 20°C with 16 h of light, 8 h of darkness, and orally administered saline or MEL for 5 weeks. A single intravenous (i.v.) injection of saline (controls), SAL, thyrotropin‐releasing hormone (TRH) or a dopamine receptor antagonist, sulpiride, was given to the goats 3 weeks after the first oral administrations of saline or MEL, and the responses were compared. The mean basal plasma PRL concentrations in the control group were higher for the saline treatments than MEL treatments (P < 0.05). SAL as well as TRH and sulpiride stimulated the release of PRL promptly after each injection in both the saline‐ and MEL‐treated groups (P < 0.05). The area under the response curve of PRL for the 60‐min period after the i.v. injection of SAL, TRH and sulpiride in the saline‐treated group was greater than each corresponding value in the MEL‐treated group (P < 0.05). These results show that daily exposure to MEL under a long day length reduces the PRL‐releasing response to SAL as well as TRH and sulpiride in goats.  相似文献

The effects of vasoactive intestinal polypeptide (VIP) and growth hormone releasing factor (GRF:hpGRF(1–29)-NH2) on the release of growth hormone (GH) from anterior pituitaries from cows were examined by using an in vitro superfusion system. The pituitaries were excised randomly from cycling cows, dissected to obtain medial portions, and minced to obtain cubes with approximate dimensions of 1.5mm on a side. For each perifusion setup, 5 pieces of pituitary tissues were chambered and flushed with modified KRB solution saturated with 95% O2-5% CO2 at 38C. Perifusion with media containing 10−6 and 10−7M VIP for 30 min induced a significant release of GH during the treatments (P<0.05). VIP (10−8M) increased GH levels significantly (P<0.05), but to a minor degree. Perifusion with the media containing 10−6, 10−7 and 10−8M GRF for 30 min markedly increased the GH concentration and the effects continued up to 90 min after termination of the perifusion of the peptide (P<0.05, P<0.01). The GH releasing effects of GRF could be seen at doses as low as 10−11M GRF (P<0.05, P<0.01).

These findings indicate that the GH releasing effect of VIP is less potent that that of GRF in cows.  相似文献

We examined the distribution of the orexin‐like peptides in the pituitary and median eminence of the flat‐tailed house gecko (Hemidactylus platyurus) using immunohistochemistry. Orexin‐B‐like, but not orexin‐A‐like, immunoreactivity was detected in the pituitary, specifically in the pars intermedia, and these cells corresponded to alpha‐melanocyte‐stimulating hormone (αMSH)‐producing cells. Orexin‐B and αMSH secreted from pars intermedia may modulate secretion of adenohypophyseal cells in the pars distalis. In the median eminence, orexin‐B‐immunoreactive puncta and fibres were observed, and these structures corresponded to gonadotropin‐releasing hormone (GnRH)‐immunoreactive puncta and fibres. Orexin‐B secreted from GnRH‐containing neurons in the hypothalamus may affect thyrotropin‐releasing hormone‐containing neurons resulting in modulation of αMSH secretion of melanotrophs in the pars intermedia.  相似文献

This study aimed to develop an in vitro model for the analysis of the bovine endometrium. Immunofluorescent staining revealed that the hetero‐spheroids and the cultured explants showed almost similar structure in the localization of bovine endometrial epithelial cells and endometrial stromal cells, except the glandular‐like structure of the epithelial cells inside the explants. Gelatin zymography revealed that the hetero‐spheroids did not express matrix metalloproteinases (MMPs) after 4 days of culture, but strong MMP expressions were observed in the cultured explants until 7 days of culture. Additionally, expression of progesterone receptor (PR), estrogen receptor alpha (ERα), type I interferon receptor 1 (IFNAR1) and 2 (IFNAR2) messenger RNA was observed both in the homo‐ and hetero‐spheroids. The expression of oxytocin receptor (OTR) mRNA in E2 and E2+P4 (1,3,5(10)‐Estratrien‐3, 17β‐diol + 4‐Pregnen‐3, 20‐dinone) treated groups were significantly (P < 0.05) higher than that of the control group of spheroids. In case of cultured explants, the expression of PR and OTR mRNA were significantly (P < 0.05) higher in E2 treated groups compared to the control groups. Hepatocyte growth factor (HGF) mRNA expression was also higher in P4 treated groups at 10 days in culture (P < 0.05). In a nutshell, the in vitro model developed in this study for the analysis of the endometrium may provide a new platform for extensive research on bovine endometrial function.  相似文献
