Cobia Rachycentron canadum juveniles (119.7 mm TL, weight 8.5 g) were reared for 10 wk at three salinity levels: 5 ppt, 15 ppt. and 30 ppt. Growth and survival were determined through biweekly sampling. Blood samples obtained at termination of the study were analyzed to determine hematocrit, blood osmolality, and total protein. Results indicated that the overall growth of fish was significantly affected by salinity. Mean (± SE) total length (TL) and weight of fish reared at a salinity of 30 ppt were 201.7 ± 2.6 mm and 47.6 ± 1.9 g, respectively, followed by fish reared at 15 ppt (182.2 ± 1.7 mm, 34.1 ± 1.6 g). and 5 ppt (168.3 ± 5.8 mm TL, 28.3 ± 2.3 g). Differences in specific growth rates among treatments for the 10-wk period were also significant. No differences were detected in mean survival among fish reared at salinities of 5, 15, and 30 ppt (84, 94, and 94%, respectively). However, fish reared at salinity 5 ppt appeared to be in poor health as skin lesions, fin erosion, and discoloration were evident. Analysis of blood revealed that, while no differences existed among treatments with respect to plasma total protein, fish reared at a salinity of 5 ppt exhibited significantly reduced hematocrit (25% vs. > 30%) and plasma osmolality values (318 vs. > 353 mmolkg) relative to fish reared at higher salinities. Cobia can tolerate exposure to low salinity environments for short periods of time without mortality; however, moderate to high salinities are required for sustained growth and health of this species. 相似文献

Theories for the chemical stability of foods cite the role of moisture content or water activity in reactant mobility, though mobility has been variously defined. One theory, based on plasticization by moisture, is limited by a lack of research directly linking the mobility of a matrix to the mobility and reactivity of small solute molecules in foods. A cross-polarization/magic angle spinning technique was developed to study glucose rotational mobility in the solid state over a range of water activities and in matrixes with different glass transition temperatures. Data analysis stressed the significance of separating molecular mobility from relaxation time. Results showed that, in a caseinate matrix, compared to a control, adding glycerol yielded the highest glucose mobility and lowest glass transition temperature (T(g)), while adding sorbitol also increased mobility and lowered T(g). Consequently, plasticization by either moisture or these humectants increases the mobility of small solute molecules such as glucose. 相似文献

The effects of transpiration rate on B uptake by roots and translocation to shoots of table beet (Beta vulgaris L. cv. Red Ace) plants were studied under conditions of environmental control. Plants grown under high or low relative humidities (RH) had low or high rates of transpiration, respectively. Dry weights and B contents of shoots and roots were higher among plants grown at 80% RH and B concentrations of roots were lower compared to plants grown at 30% RH. Shoot B concentrations were not affected by RH. Rates of growth and B accumulation in shoots were closely correlated for plants grown in 30% or 80% RH. These parameters were not closely correlated in roots.
The rate of B uptake per g root dry weight was greater in plants having higher transpiration rates; whereas the rate of B translocation to shoots per g dry weight was independent of transpiration rate. Thus we propose that B translocation to shoots is controlled mechanistically by rates of dry matter accumulation during stages of rapid growth, i.e. a sink effect. 相似文献

A method is reported for species quantification by exploiting single-nucleotide polymorphisms (SNPs). These single-base changes in DNA are particularly useful because they enable discrimination of closely related species and/or varieties. As a model, quantitative authentication studies were performed on coffee. These involved the determination of the percentage of Arabica and Robusta species based on a SNP in the chloroplastic trnL(UAA)-trnF(GAA) intraspacer region. Following polymerase chain reaction (PCR), the Robusta-specific and Arabica-specific fragments were subjected to 15 min extension reactions by DNA polymerase using species-specific primers carrying oligo(dA) tags. Biotin was incorporated into the extended strands. The products were captured in streptavidin-coated microtiter wells and quantified by using oligo(dT)-conjugated photoprotein aequorin. Aequorin was measured within 3 s via its characteristic flash-type bioluminescent reaction that was triggered by the addition of Ca(2+). Because of the close resemblance between the two DNA fragments, during PCR one species serves as an internal standard for the other. The percentage of the total luminescence signal obtained from a certain species was linearly related to the percent content of the sample with respect to this species. The method is accurate and reproducible. The microtiter well-based assay configuration allows high sample throughput and facilitates greatly the automation. 相似文献

The effective utilization of crop diversity held in genebanks depends on our knowledge of useful traits and available markers associated with the target traits. Target region amplification polymorphism (TRAP) was used to evaluate the genetic diversity and underlying relationships among 263 accessions of chickpea landraces maintained by the USDA-ARS Western Regional Plant Introduction Station in Pullman, WA, USA. Two-hundred sixty-two TRAP markers were amplified by eight primer combinations. Altogether, 110 (42 %) markers were polymorphic, the other 152 (58 %) displayed no variation. These polymorphic markers revealed important differences among the accessions, with an estimated, mean pair-wise genetic distance of 25.82 %, ranging from 2.8 to 50.0 %. Genetic distance analysis divided the accessions into two major groups, with 113 and 150 accessions each, and substantial association between molecular diversity and geographic origin was evident. Bayesian analysis of population structure revealed two groups (K = 2) with evidence for six sub-groups. Additionally, the population structure of a subset of 110 lines was determined (K = 3) for testing marker-trait associations (MTAs). Phenotypic traits included the concentrations of protein and nine mineral elements in the seeds. Two MTAs were significant (p < 0.01) for concentrations of Ca and K, and three MTAs were significant for Cu and Ni concentrations. The results indicate that this population is useful for genome-wide association studies on other economic traits given the level of genetic diversity uncovered and the marker-trait associations in seed minerals discovered. 相似文献

A catchment-wide study was conducted to investigate the geography of nutrient dynamics in the Androscoggin and Kennebec Rivers, Maine. While nutrient concentrations resembled relatively pristine waters, nitrogen (N) and phosphorus (P) exhibited dramatic spatial and temporal patterns. Though these rivers are underdeveloped, impacts of human activities upon nutrients are clear. Population was a good predictor for DIN and P concentrations for the Kennebec River, but a poor predictor in the Androscoggin River. Comparison of point-source discharges into each river indicated that the Androscoggin River is influenced by discharges from pulp and paper mills, a finding confirmed by reduced nutrient transport after the closure of two Androscoggin River mills. Nutrient ratio analysis demonstrated a dramatic shift towards Redfield ratios, with DIN:P ratios decreasing from about 200:1 in headwater reaches to below 16:1 near the river outlets. These results may aid in understanding blooms of toxic dynoflagellates in the Gulf of Maine. Ratios were also altered during a prolonged drought period. Results describe a simple relationship between increased nutrient input and human population within single watersheds, with implications for coastal zone ecosystems. 相似文献

Water availability is an important factor for irrigated agriculture in the Mediterranean countries, because it affects significantly crop production. Surface soil samples were collected in Merambelo area located in Eastern Crete and analyzed. High values for exchangeable potassium and magnesium values were recorded and can be attributed to intensive fertilization and soil heredity factors. Groundwater quality based on two sampling campaigns performed during two irrigation periods. Three different scenarios were taken into account regarding the height of water application by means of irrigation. The contribution and the effects of nutrients and micronutrients from irrigation water to the soil was calculated, on the basis of the mean determined concentrations and the volume of applied irrigation. Significant variation was recorded in most soil properties, and these differences may be attributed to different mobility of nutrients, erosion factors, fertilization practices, and hydraulic soil properties. 相似文献

Three studies were conducted to determine and confirm the effective dosage rate of ceftiofur crystalline-free acid sterile suspension (CCFA-SS, 200 mg ceftiofur equivalents [CE]/ml), a long-acting ceftiofur formulation, for control and treatment of bovine respiratory disease (BRD). In each study, CCFA-SS was administered once by subcutaneous (SC) injection in the middle third of the posterior aspect of the ear. Study 1 was conducted using an intratracheal challenge with Mannheimia (formerly Pasteurella) haemolytica and dosages ranging from 0 to 8.8 mg CE/kg to select a dosage for further field testing. In Study 2, a single dose of CCFA-SS at 0.0, 4.4, or 6.6 mg CE/kg was administered when uniform clinical signs of BRD were present in feedlot cattle. Study 3 was conducted in several feedlots to evaluate the efficacy, practicality, and safety of CCFA-SS at 4.4 or 6.6 mg CE/kg compared with a placebo control or tilmicosin for preemptive control of BRD. In Study 1, the effective dose was determined to be 5.35 mg CE/kg; therefore, 4.4 and 6.6 mg CE/kg were selected as the dosages for further field testing. Administration of CCFA-SS at 4.4 or 6.6 mg CE/kg improved treatment success compared with negative controls (P < or =.05 for both doses) in Study 2. In Study 3, a single administration of 4.4 or 6.6 mg CE/kg was comparable to tilmicosin (P <.001) and was significantly better than placebo (P <.001) for the control of BRD. Using the ear as an administration site was acceptable under field conditions and was well tolerated by all animals. These studies demonstrated that a single administration of CCFA-SS by SC injection in the middle third of the posterior aspect of the ear at 4.4 or 6.6 mg CE/kg is effective, safe, and practical for preemptive control and treatment of the bacterial component of BRD in feedlot cattle. Antioxidative activity of hydrolyzed protein prepared from alkali-solubilized catfish protein isolates was studied. The isolates were hydrolyzed to 5, 15, and 30% degree of hydrolysis using the protease enzyme, Protamex. Hydrolyzed protein was separated into hydrolysates and soluble supernatants, and both of these fractions were studied for their metal chelating ability, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, ferric reducing antioxidant power (FRAP), oxygen radical absorbance capacity (ORAC), and their ability to inhibit the formation of thiobarbituric acid reactive substances (TBARS) in washed tilapia muscle containing tilapia hemolysate. Both hydrolysates and supernatants were characterized using sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Results showed that DPPH radical scavenging ability and reducing power of catfish protein hydrolysates decreased, whereas the ORAC value, metal chelating ability, and ability to inhibit TBARS increased, with an increase in the degree of hydrolysis. Hydrolysate samples showed higher DPPH radical scavenging ability and Fe(3+) reducing ability, and supernatant samples had higher metal chelating ability. In general, low molecular weight (MW) peptides had high ORAC values and high metal chelating ability, and high MW peptides had a higher reducing power (FRAP) and were more effective in scavenging DPPH radicals. In a washed muscle model system, the ability of catfish protein hydrolysates and their corresponding supernatants to inhibit the formation of TBARS increased with an increase in the degree of hydrolysis. 相似文献

Greenhouse studies were conducted to evaluate the sensitivity of three commercial cultivars, eight experimental cultivars and common bermudagrass to clethodim, glufosinate and glyphosate. Each herbicide was applied at eight doses. Data were regressed on herbicide dose using a log-logistic curve (R2 = 0.56-0.95 for clethodim, R2 = 0.60-0.94 for glufosinate, and R2 = 0.70-0.96 for glyphosate). The herbicide rate that elicited a 50% plant response (I50) in the bermudagrass cultivars ranged from 0.04 to 0.19 kg ha(-1) clethodim, 0.19 to 1.33 kg ha(-1) glufosinate and 0.34 to 1.14 kg ha(-1) glyphosate. Relative to other cultivars, common bermudagrass was intermediate in its response to clethodim and among the most tolerant cultivars to glufosinate and glyphosate. TifSport was relatively tolerant to clethodim and glufosinate compared with other cultivars, but relatively sensitive to glyphosate. One cultivar, 94-437, was consistently among the most sensitive cultivars to each of the herbicides. While there were differential herbicide tolerances among the tested bermudagrass cultivars, there did not appear to be any naturally occurring herbicide resistance that could be commercially utilized. However, research indicated that breeding efforts should target herbicide resistance that is at least four times the registered use rate. Also, TifSport and Tifway have been identified as suitable representatives of triploid hybrid bermudagrass cultivars to be used to evaluate the success of turfgrass renovation programs. 相似文献
