The growth of rainbow trout fed diets containing 4, 10 and 25% chitin, over a 12-week period, was significantly depressed (P < 0.001) when compared with controls fed diets containing 25% starch. There was no difference in growth rate between control fish and those fed diets containing 10% N-acetyl glucosamine (GlcNAc). Isotopically labelled amino sugars were shown to be oxidized when injected intraperitoneally into rainbow trout. Relatively high levels were found of chitinase activity in the stomachs and of chitobiase in the intestines. These enzyme activities were similar in all the trout, irrespective of the amount of chitin in their diets, except that chitobiase in the intestines of fish fed diets containing GlcNAc showed higher levels of activity than the controls (P < 0.05). Chitin was not significantly digested when fed at 10 and 30% of the diet but the apparent digestibility of pre-cooked starch was 50% when fed at either 15 or 25%, on the basis of the inert indicator (Cr2O3) method. Chitinase and chitobiase activities were not reduced in fish fed diets containing 10% chitin and an antibiotic (Tribrissen®) although the alimentary microflora were completely eliminated. Enzyme activities were not enhanced when live chitinolytic bacteria (Vibrio alginolyticus) were incorporated into diets with 10% chitin; these bacteria were only recovered from the intestine. The evidence indicates an endogenous origin of chitinolytic enzymes in the trout gastro-intestinal tract. The presence of either antibiotic or bacteria in the diet had no effect on the digestibility of chitin.  相似文献

An ideal surface for implantable glucose sensors would be able to evade the events leading to chronic inflammation and fibrosis, thereby extending its utility in an in vivo environment. Nafion?, a perfluorinated ionomer, is the membrane material preferred for in situ glucose sensors. Unfortunately, the surface properties of Nafion? promote random protein adsorption and eventual foreign body encapsulation, thus leading to loss of glucose signal over time. Details of the techniques to render Nafion? nonprotein fouling are given in a previous article [T. I. Valdes et al., Biomaterials 29, 1356 (2008)]. Once random protein adsorption is prevented, a biologically active peptide can be covalently bonded to the treated Nafion? to induce cellular adhesion. Cellular responses to these novel decorated Nafion? surfaces are detailed here, including cell viability, cell spreading, and type I collagen synthesis. Normal human dermal fibroblasts (NHDFs) were cultured on control and modified Nafion? surfaces. Findings indicate that Nafion? modified with 10% 2-hydroxyethyl methacrylate and 90% tetraglyme created a nonfouling surface that was subsequently decorated with the YRGDS peptide. NHDFs were shown to have exhibited decreased type I collagen production in comparison to NHDF cells on unmodified Nafion? surfaces. Here, the authors report evidence that proves that optimizing conditions to prevent protein adsorption and enhance cellular adhesion may eliminate fibrous encapsulation of an implant.  相似文献

Quantifying the impacts of climate change on weed populations requires an understanding of the relative contributions of endogenous and exogenous factors on their numerical fluctuations. Here, we have used long‐term data (26 years) of seven weed species growing in a cereal–legume rotation from a locality in central Spain to determine the importance of endogenous (density dependence) and exogenous (tillage system, crop rotation, temperature and precipitation) factors. Density dependence was the main driver of the population dynamics studied, and it was exhibited more frequently under zero tillage (86% of the species) than under minimum tillage (57% of the species). Our results confirmed previous findings and provided stronger support for density dependence under zero tillage than under minimum tillage. Under the latter, temperature negatively affected the population growth rate of Descurainia sophia and positively Atriplex patula. We found no effect of either precipitation or crop rotation on population dynamics. Our findings could underpin an awareness campaign aimed at farmers to prevent them from drawing unwarranted conclusions regarding the efficacy of the particular control method used in a given year.  相似文献

Queensland fruit fly, Bactrocera tryoni (Froggatt), is the most serious pest of the native tephritid species in Australia and a significant market access impediment for fruit commodities from any area where this species is endemic. An area-wide management (AWM) program was implemented in the Central Burnett district of Queensland with the aim of improving fruit fly control and enhancing market access opportunities for citrus and other fruits produced in the district. The primary control measures adopted in the AWM system included bait spraying of commercial and non-commercial hosts and the year-round installation of male annihilation technology (MAT) carriers in both orchards and town areas. The MAT carrier used consisted of a dental wick impregnated with 1 ml cue-lure [4-(4-acetoxyphenol)-2-butanone] and 1 ml Malathion 500 EC in a plastic cup. The application of these control measures from 2003 to 2007 resulted in overall suppression of fruit fly populations across the entire district. Male trap catches at the peak activity time were reduced by 95% and overall fruit fly infestation in untreated backyard fruit of town areas reduced from 60.8% to 21.8%. Our results demonstrate remarkable improvement in fruit fly control and economic benefit to the Central Burnett horticulture. Therefore, commercial growers are continuing the AWM program as a long-term, industry funded activity, to provide an additional layer of phytosanitary security for market access of fruit commodities from this district.  相似文献

The regional scale variability of the bacterial community inhabiting the rhizosphere was studied with soil collected from maize fields located in the Santo Domingo Valley (SDV; Baja California Sur, Mexico), a semi-arid agricultural ecosystem of approximately 200 km2. The bacterial community structure was visualized by single-strand conformation polymorphism (SSCP) profiles of PCR-amplified partial 16S rRNA genes of directly extracted rhizosphere soil DNA. SSCP profiles of different SDV sites and an external field site in Germany were evaluated for their similarities and the contributing bacteria were characterized by DNA sequence analyses. SSCP profiles from each site were significantly different from the others, as revealed by permutation of pairwise similarities (P < 0.05). In comparison to the German site, SSCP profiles from SDV were more similar to each other despite contrasting soil salinity levels. Correspondence analysis revealed that among SDV sites, salinity levels, soil organic carbon and calcium (Ca2+) were most influential on the bacterial community structure. Depending on the phylogenetic group analyzed (Bacteria, Alphaproteobacteria, Pseudomonas), the importance of these soil variables varied. Interestingly, the East–West direction also revealed an effect, suggesting that future explorations of bacterial diversity patterns should also consider landscape topography in search of explaining patterns of bacterial diversity in soils.  相似文献
