In a switch-over experiment, eight male animals, four each of sheep and goats of local breeds with mean body weight of 26.
8 ± 2.0 and 30.0 ± 2.1 kg, were fed Dichanthium annulatum (DA) grass and four browse species viz. Helictris isora, Securengia virosa, Leucaena leucocephala (LL) and Hardwickia binnata (HB) in four feeding trials to assess their supplementary effect on activity of rumen enzymes. The sheep and goats were offered
DA grass with individual browse in 75:25 and 50:50 proportions, respectively, for more than 3 months during each feeding trial,
and rumen liquor samples were collected twice at 0 and 4 h post feeding after 60 and 90 days of feeding. Glutamate oxaloacetate
transaminase (GOT), glutamate pyruvate transaminase (GPT) and glutamate dehydrogenase (GDH) enzymes were determined in the
bacteria and protozoa fractions of rumen liquor, while cellulase enzyme activity was measured in mixed rumen liquor. LL and
HB had the highest and lowest contents of CP, while fibre contents were lower in early than later browse leaves. Supplementation
of browse leaves significantly (P < 0.05) affect the specific activity of GDH enzyme in bacteria fraction of rumen liquor of animal species, while GDH activity
was similar in protozoa fraction of rumen liquor of sheep and goats on all DA grass–browse-supplemented diets except DA–HB
(42.8 units/mg protein), where activity was significantly (P < 0.05) low. Specific activities of GOT and GPT enzymes in both bacteria and protozoa fractions of rumen liquor differ significantly
(P < 0.05) due to supplementation of browse leaves to DA grass. Browse leaves significantly (P < 0.05) affect the cellulase enzyme activity in animal rumen liquor, being highest on DA–LL (193.4) and lowest on DA–HB diet

Proteomic assessment of low-abundance leaf proteins is hindered by the large quantity of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) present within plant leaf tissues. In the present study, total proteins were extracted from wheat (Triticum aestivum L.) leaves by a conventional trichloroacetic acid (TCA)/acetone method and a protocol first developed in this work. Phytate/Ca2+ fractionation and TCA/acetone precipitation were combined to design an improved TCA/acetone method. The extracted proteins were analysed by two-dimensional gel electrophoresis (2-DE). The resulting 2-DE images were compared to reveal major differences. The results showed that large quantities of Rubisco were deleted from wheat leaf proteins prepared by the improved method. As many as (758±4) protein spots were detected from 2-DE images of protein extracts obtained by the improved method, 130 more than those detected by the TCA/acetone method. Further analysis indicated that more protein spots could be detected at regions of pI 4.00–4.99 and 6.50–7.00 in the improved method-based 2-DE images. Our findings indicated that the improved method is an efficient protein preparation protocol for separating low-abundance proteins in wheat leaf tissues by 2-DE analysis. Four Nili-Ravi buffalo calves (100 ± 4 kg) were used in 4 × 4 Latin Square Design to evaluate the influence of varying ruminally degradable protein (RDP) to ruminally undegradable protein (RUP) ratio on dry matter intake (DMI), digestibility and nitrogen (N) metabolism. Four experimental diets A, B, C and D were formulated to contain RDP:RUP of 70:30, 65:35, 60:40 and 55:45, respectively. The calves were fed ad libitum. Dry matter intake by calves fed C diet was higher (P < 0.05) than those fed D diet and lower (P < 0.05) than calves fed A diet, however, it was similar to those fed B diet. There was a linear decrease (P < 0.01) in DMI with decreasing the RDP to RUP ratio. Similar trend was noticed in crude protein (CP) intake. Neutral detergent fiber (NDF) intake was significantly different across all treatment. The decrease in CP and NDF intake was due to decreasing trend of DMI. Dry matter (DM) digestibility in calves fed A and B diets was higher (P < 0.05) than those fed C and D diets. A linear decrease (P < 0.01) in DM digestibility was observed with decreasing the RDP to RUP ratio. Crude protein digestibility remained unaltered across all treatments. Neutral detergent fiber digestibility was higher in calves fed A and B diets than those fed C and D diets. Higher NDF digestibility in calves fed A and B diets was due to higher level of dietary RDP that might resulted in higher ruminal ammonia concentration which stimulate activity of cellulytic bacteria and ultimately increased NDF digestibility. The N retention (g/d) was similar among the calves fed B, C and D diets, however, it was higher (P < 0.05) than those fed A diet. Decreasing the RDP to RUP ratio resulted in linear increase (P < 0.01) in N retention. The N retention, as percent of N intake was significantly different across all treatments. Decreasing RDP to RUP ratio resulted in linear increase (P < 0.01) in N retention, as percent of N intake. A similar trend was noticed in N retention, as percentage of N digestion. Blood urea nitrogen (BUN) concentration in calves fed B diet was higher (P < 0.05) than those fed D diet and was lower (P < 0.05) than those fed A diet, however, it was not different from calves fed C diet. Decreasing dietary RDP to RUP ratio resulted in linear decrease (P < 0.05) in BUN concentrations. The decrease in BUN concentration was because of decreasing level of dietary RDP. The N retention can be increased by decreasing RDP to RUP ratio in the diet of growing buffalo calves and diet containing RDP to RUP ratio 55:45 is considered optimum regarding N retention in buffalo calves. 相似文献
