Anthracnose caused by species of Colletotrichum is considered one of the main postharvest diseases for avocado. In this study, Colletotrichum isolates associated with avocado anthracnose, collected in different states of Brazil, were evaluated through phylogenetic analysis, morphological characterization, and pathogenicity assays. Moreover, the events during pathogen infection of avocados were examined by scanning electron microscopy. To assess the genetic diversity of 54 Colletotrichum isolates, partial sequence analysis of the gene gapdh was performed. According to the generated groupings and the geographical origins of isolates, a subset of 14 strains was selected for performing multilocus phylogeny analysis (using sequences of gapdh, act, tub2, and ApMat). Two species previously described were identified: C. siamense belonging to the C. gloeosporioides species complex and Colletotrichum karstii belonging to the C. boninense species complex. All Colletotrichum strains evaluated caused typical symptoms of anthracnose in avocado fruits. Conidia of the most virulent strain germinated between 6 and 12 hr after inoculation (hai). Penetration through wounds occurred 48 hai, tissue colonization occurred between 144 and 240 hai, and sporulation took place at 240 hai via the production of an acervulus, conidiophores, and conidia. The findings shed light on the aetiology of avocado anthracnose in Brazil and provide a better understanding of the infection process of this pathogen, which may assist in the development of disease management strategies.  相似文献

Apple blotch (Marssonina coronaria) is a foliar disease of increasing importance globally. Methods to quantify the disease and knowledge about epidemiology are required for new studies on the disease. The objectives of this study were to develop a standard area diagram set (SADs) to assess apple blotch severity and to describe the temporal progress of the disease under field conditions on older and younger leaves of two apple cultivars. For the development of SADs, symptomatic leaves were collected and scanned to obtain the actual severity. Based on the pattern of the disease, a SADs was elaborated and validated. Leaf severities were estimated without and with SADs by 12 raters to validate the tool. After validation, the SADs was used to assess apple blotch in older and younger leaves of selected shoots during the vegetative cycle of cvs. Eva and Gala. SADs severities ranged from 0.2 to 96%. Accuracy, precision, and reliability of the estimates were significantly improved when the SADs was used. In the field, the onset of apple blotch was in late spring and greater increases in severity occurred during summer. Rates of disease progress ranged from 0.09 to 0.13. Epidemics were different in older and younger leaves at the end of the apple vegetative cycle for both cultivars tested. Our work provided a tool for apple blotch quantification and described the disease progress curve under subtropical conditions, which can contribute as basis for future studies.

It is difficult to use tissue culture assays to investigate adherence and other properties of Edwardsiella tarda because the organism is invasive and produces a potent hemolysin. We therefore relied on polymerase chain reaction (PCR) to determine the occurrence of genes for enterotoxins (LT-I, EAST-1), Shiga toxin (Stx-1, Stx-2), cytotoxic necrotizing factors (CNF-1, CNF-2), aerobactin, invasion plasmid of enteroinvasive Escherichia coli, EPEC adherence factor (EAF), intimin (Eae), enterohemolysin (EntHly) and hemolysin (Hly) in 53 isolates of E. tarda from humans and fish from several countries. All isolates were negative for all genes investigated by PCR. Adhesion to and invasion of HeLa cells were determined by using the unusually short incubation time of 1h or 30 min. All isolates adhered and invaded in these tests. Finally, a random amplified polymorphic DNA (RAPD) test distinguished, with a few exceptions, isolates of human and fish origin.  相似文献

The effect of coffee on cardiovascular diseases is still controversial. It is known that the process of decaffeination may influence the chemical constitution and, therefore, the biological effects of coffee. This study thus evaluated the effects of decaffeination on the levels of total phenols and chlorogenic acids in Coffea arabica L. samples, as well as the effects of ingesting both integral and decaffeinated coffee on the lipid profile and hemostatic and hematological parameters in normal and hyperlipidemic rats. Samples of integral and decaffeinated lyophilized coffee (Coffea arabica L., planted in Brazil) were used for chemical analysis (total phenols, chlorogenic acid and caffeine contents). For the bioassays, coffee beverages were prepared with non-lyophilized samples (10 % w/v) and were filtered and administered to animals by gavage (7.2 mL/kg/day) over 30 days. On the 31st day after beginning the treatment with coffee beverages, hyperlipidemia was induced to the animals by administering Triton WR-1339 (300 mg/kg body weight). On day 32, blood was taken to determine the lipid profile, platelet aggregation, prothrombin time, partially activated thromboplastin time and hemogram. The contents of both phenolic compounds and chlorogenic acid in the integral coffee beverage were significantly lower than those in the decaffeinated coffee beverage. The animals treated with Triton WR-1339 presented a mixed hyperlipidemia. Although the decaffeination process caused a relative increase in total phenols and chlorogenic acids, the coffee drinks were unable to change the lipid profile or the hemostatic and hematological parameters in the studied animals.  相似文献

Athletic horses often experience painful conditions of the musculoskeletal system, but their ability to compete can be prolonged using certain anesthetic agents. The present study investigated the ability of thermography to detect fetlock and middle carpal intrasynovial injections of bupivacaine hydrochloride in five mares. Saline injections were performed in the contralateral limbs. Thermographic evaluation was conducted at the dorsal and palmar aspects before (basal) and 15, 30, 60, 90, 120, and 1440 minutes after injection. The intrasynovial treatments resulted in increased limb temperature, with fetlock temperatures higher on the dorsal aspect at 15, 30, and 60 minutes and on the palmar aspect from 15 to 1440 minutes (P < .05) after the bupivacaine and saline injections. Increased carpal temperature was detected on the dorsal aspect at 60 and 90 minutes (P < .05). The present study demonstrates that thermography can be used to detect intrasynovial injections in horses.  相似文献

Standard area diagrams (SADs) to assess the severity of potato early blight (Alternaria grandis) on leaves of potato (Solanum tuberosum L.) were developed and validated. The proposed SADs include images of leaves with 10 distinct disease severities (0.1, 1, 3, 5, 10, 20, 40, 60, 80 and 100 %). The SADs were validated by 12 raters who had no previous experience in evaluating plant disease. Lin’s concordance correlation analysis of estimated vs. actual disease severity (based on image analysis) showed that precision and accuracy improved for most raters using the SADs, compared to assessments made without the SADs. The SADs improved accuracy (coefficient of bias, C b ?=?0.97 and 0.99, without and with SADs, respectively) and agreement (Lin’s concordance correlation coefficient, ρ c ?=?0.91 and 0.98 without and with SADs, respectively) of the estimates of severity. Severity estimates were more reliable when using SADs (coefficient of determination, R 2 ?=?0.80 unaided and R 2 ?=?0.95 with SADs, and the intra-class correlation ρ?=?0.86 without SADs and ρ?=?0.97 using the SADs). The SADs improved raters’ ability to accurately, precisely and reliably estimate potato early blight severity, and as such can be used to assess severity for several purposes, including breeding for resistance, fungicide screening, and pathotype characterization.  相似文献

Soil sulfur (S) partitioning among the various pools and changes in tropical pasture ecosystems remain poorly understood. Our study aimed to investigate the dynamics and distribution of soil S fractions in an 8‐year‐old signal grass (Brachiaria decumbens Stapf.) pasture fertilized with nitrogen (N) and S. A factorial combination of two N rates (0 and 600 kg N ha–1 y–1, as NH4NO3) and two S rates (0 and 60 kg S ha–1 y–1, as gypsum) were applied to signal grass pastures during 2 y. Cattle grazing was controlled during the experimental period. Organic S was the major S pool found in the tropical pasture soil, and represented 97% to 99% of total S content. Among the organic S fractions, residual S was the most abundant (42% to 67% of total S), followed by ester‐bonded S (19% to 42%), and C‐bonded S (11% to 19%). Plant‐available inorganic SO4‐S concentrations were very low, even for the treatments receiving S fertilizers. Low inorganic SO4‐S stocks suggest that S losses may play a major role in S dynamics of sandy tropical soils. Nitrogen and S additions affected forage yield, S plant uptake, and organic S fractions in the soil. Among the various soil fractions, residual S showed the greatest changes in response to N and S fertilization. Soil organic S increased in plots fertilized with S following the residual S fraction increment (16.6% to 34.8%). Soils cultivated without N and S fertilization showed a decrease in all soil organic S fractions.  相似文献

Many authors attribute the antioxidant activity of brewed coffee to its caffeine content. In addition, caffeine intake has been associated with increased performance during physical exercise. This study analyzed the in vivo effects of drinking caffeinated and decaffeinated instant coffee (8%, w/v) on oxidative stress and antioxidant enzyme activity in the anterior tibialis muscles of rats subjected to intense exercise. It was observed that exercise induced lipid peroxidation (estimated using malondialdehyde) and protein oxidation (evaluated by determining the formation of carbonyl groups) in the muscle (P < 0.05). Decaffeinated instant coffee and caffeine solution did not exhibit antioxidant activity in vivo. Caffeinated instant coffee beverage intake did not induce changes in superoxide dismutase and glutathione peroxidase activities but was able to diminish lipid and protein oxidation in the anterior tibialis muscles of rats after exercise (P < 0.05), contributing to a reduction in the oxidative stress triggered by exercise.  相似文献

The physiological responses of sugarcane (Saccharum officinarum L.) to oxidative stress induced by methyl viologen (paraquat) were examined with respect to photochemical activity, chlorophyll content, lipid peroxidation and superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities. Thirty-day-old sugarcane plants were sprayed with 0, 2, 4, 6 and 8 mM methyl viologen (MV). Chlorophyll fluorescence was measured after 18 h and biochemical analyses were performed after 24 and 48 h. Concentrations of MV above 2 mM caused significant damage to photosystem II (PSII) activity. Potential and effective quantum efficiency of PSII and apparent electron transport rate were greatly reduced or practically abolished. Both chlorophyll and soluble protein contents steadily decreased with MV concentrations above 2 mM after 24 h of exposure, which became more pronounced after 48 h, achieving a 3-fold decrease. Insoluble protein contents were little affected by MV. Oxidative stress induced by MV was evidenced by increases in lipid peroxidation. Specific activity of SOD increased, even after 48 h of exposure to the highest concentrations of MV, but total activity on a fresh weight basis did not change significantly. Nondenaturing PAGE assayed with H2O2 and KCN showed that treatment with MV did not change Cu/Zn-SOD and Mn-SOD isoform activities. In contrast, APX specific activity increased at 2 mM MV but then dropped at higher doses. Oxidative damage induced by MV was inversely related to APX activity. It is suggested that the major MV-induced oxidative damages in sugarcane leaves were related to excess H2O2, probably in chloroplasts, caused by an imbalance between SOD and APX activities, in which APX was a limiting step. Reduced photochemical activity allowed the early detection of the ensuing oxidative stress.  相似文献

The present study investigated the effects of isopulegol, a monoterpene alcohol, in PTZ-induced convulsions and verified possible involved mechanisms. Saline, isopulegol or diazepam were intraperitonealy injected 30 min before PTZ. The latency for development of convulsions and mortality, as well as the mortality protection percentage was recorded. For investigating the involvement of GABAergic system, flumazenil was utilized. The activity of antioxidant enzyme catalase as well as the levels of reduced glutathione and lipid peroxidation were measured in brain hippocampus. Similarly to diazepam, isopulegol significantly prolonged the latency for convulsions and mortality of mice. All animals were protected against mortality at higher dose of isopulegol. Flumazenil pretreatment decreased the prolongation of seizure latency induced by both diazepam and isopulegol, although it was not able to reverse the latency and protection percent for mortality. Isopulegol also significantly prevented PTZ-induced increase in lipid peroxidation, preserved catalase activity in normal levels, and prevented the PTZ-induced loss of GSH in hippocampus of mice. These results suggest that the anticonvulsant and bioprotective effects of isopulegol against PTZ-induced convulsions are possibly related to positive modulation of benzodiazepine-sensitive GABAA receptors and to antioxidant properties.  相似文献
