Restaurant patrons, particularly in upscale locations, increasingly prefer entrées developed from portions cut from fillets larger than those currently sold by the US catfish, Ictalurus punctatus, industry. A production study evaluated the feasibility of producing the necessary size (1.6 kg) of catfish. Twelve 0.1‐ha earthen ponds were stocked with 0.363‐kg channel catfish at 2500 (low); 5000 (medium); or 7500 (high) fish/ha, with four replicates. Mean individual weight at harvest exceeded the mean target weight (1.6 kg) at the two lower densities, and the minimum target weight (1.36 kg) at the highest density. Percentages of fish (by weight) that did not meet the minimum weight required were: 1, 5, and 18%, in the low, medium, and high density treatments, respectively. Yields were significantly greater at higher densities (P < 0.05). Costs of production at the two higher densities were 4 to 7% higher than in the traditional multiple‐batch system, but increased to 51% at the lowest density. Results demonstrated the biological feasibility of producing channel catfish of a mean weight of 1.6 kg. Economic feasibility depends on (1) the price premium paid and (2) development of a market for fish sizes between current acceptable maximum sizes accepted and 1.36 kg.  相似文献

Gonadotropin (GTH) hormones are glycoprotein which stimulates gonadal maturation in vertebrates. Follicle stimulating hormone is involved in initiation of gametogenesis and regulation of gonadal growth. FSHβ has been cloned and characterized from the brain of Catla catla. The FSHβ full‐length of cDNA sequence of 523 bp comprised 3, 394 and 128 bp of 5′‐UTR, open reading frame (ORF) 3′‐UTR respectively. The coding region of C. catla FSHβ encoded a peptide of 130 amino acids. Phylogenetic analysis of C. catla FSHβ deduced amino acid sequence showed high similarity with Gobiocypris rarus followed by goldfish, Carassius auratus. The qPCR result shows that FSHβ mRNA is mainly expressed in pituitary while moderate and low expression was observed in testis and ovary respectively. Chitosan‐nanoconjugated kisspeptin‐10 (CK‐10) of particle size 125 nm, polydispersity index of 0.335 to 0.65 and zeta potential of ?34.95 mV were synthesized and evaluated at against naked kisspeptin‐10 for their reproductive hormonal profile. Treatment of fish with CK‐10 showed controlled and sustained surge of the reproductive hormones (FSH & LH) with peak at 12 h. The hormone levels of naked kisspeptin‐10 treated fish decline after 6 h. The sustained release of this CK‐10 will help in reducing maturation age, synchronization of ovulation and spawning in fish. This is the first report on use of chitosan‐nanoconjugated kisspeptin‐10 (CK‐10) for reproduction in fish.  相似文献

To evaluate different crosses and purebreds ducks in respect to various economic traits and to estimate different crossbreeding genetic parameters, a 3?×?3 complete diallel cross involving indigenous duck (DD), Khaki Campbell (KK) and White Pekin (WW) were used to produce three purebreds (DD, KK, WW) three crossbreds (DK, DW, KW) and three reciprocals (KD, WD, WK). A total of 609 ducklings produced were reared on deep litter and the females (316 in number) were evaluated for growing and laying period body weight along with the production performance traits. Different crossbreeding genetic parameters were estimated for different traits. All the traits in respect to body weight gain during growing and laying period and different production traits including laying house mortality rate showed significant (p?≤?0.05) difference between different genetic groups. In general, crossbreds perform better than the purebreds for most of the traits studied. General combining ability (GCA), specific combining ability (SCA) and reciprocal effect (RE) were significant (p?≤?0.01) for body weight and production traits. Egg weight showed significant (p?≤?0.01) difference in respect to GCA, SCA and RE for all the ages of measurement except RE for 30th week egg weight. Laying period mortality rate was only significant (p?≤?0.05) for SCA. Most of the crossbreds recorded heterosis rate in desirable direction for majority of the traits. Overall results revealed that the crossbreds perform well in respect to different traits than the purebreds and may be used to take advantage of heterosis. DW performs well in respect to majority of the traits measured and is of importance for commercial exploitation. Further, pure line selection with development of specialised sire and dam line followed by crossing may be of importance to enhance the performances in the crosses.  相似文献

Purines are synthesized de novo in 10 chemical steps that are catalyzed by six enzymes in eukaryotes. Studies in vitro have provided little evidence of anticipated protein-protein interactions that would enable substrate channeling and regulation of the metabolic flux. We applied fluorescence microscopy to HeLa cells and discovered that all six enzymes colocalize to form clusters in the cellular cytoplasm. The association and dissociation of these enzyme clusters can be regulated dynamically, by either changing the purine levels of or adding exogenous agents to the culture media. Collectively, the data provide strong evidence for the formation of a multi-enzyme complex, the "purinosome," to carry out de novo purine biosynthesis in cells.  相似文献

Various methods have been applied for the enzymatic preparation of diacylglycerols that are used as dietary oils for weight reduction in obesity and related disorders. Interesterification of rapeseed oil triacylglycerols with commercial preparations of monoacylglycerols, such as Monomuls 90-O18, Mulgaprime 90, and Nutrisoft 55, catalyzed by immobilized lipase from Rhizomucor miehei (Lipozyme RM IM) in vacuo at 60 degrees C led to extensive (from 60 to 75%) formation of diacylglycerols. Esterification of rapeseed oil fatty acids with Nutrisoft, catalyzed by Lipozyme RM in vacuo at 60 degrees C, also led to extensive (from 60 to 70%) formation of diacylglycerols. Esterification of rapeseed oil fatty acids with glycerol in vacuo at 60 degrees C, catalyzed by Lipozyme RM and lipases from Thermomyces lanuginosus (Lipozyme TL IM) and Candida antarctica (lipase B, Novozym 435), also provided diacylglycerols, however, to a lower extent (40-45%). High-throughput deep-sequencing technology and bioinformatics analysis of the small RNA(sRNA) population isolated from plants allows universal virus detection and complete virome reconstruction for a given sample. In the present sRNA deep-sequencing analysis of virus-infected wheat samples in the Czech Republic, samples were firstly tested for barley yellow dwarf viruses(BYDVs), wheat streak mosaic virus(WSMV) and wheat dwarf virus(WDV) using ELISA, RT-PCR and PCR. Subsequent sRNA sequencing of these samples yielded more than ~60 million single-end 50-bp reads with high confidence for nine field samples of wheat. Overall, 16.5% of reads were virus-specific and 83.5% were mapped to the host. More 21-nt reads(~7.7 E+06 reads) were found than 24-nt(~6.20 E+06 reads) or 22-nt(~4.30 E+06 reads) reads. De novo assembly of the high-quality contigs revealed the presence of three earlier reported viruses in the Czech Republic: BYDVs(31.48%), WSMV(24.23%) and WDV(26.66%). We also showed the presence of cereal yellow dwarf virus(14.33%; two species CYDV-RPS and CYDV-RPV(family Luteoviridae/Polerovirus) and wheat yellow dwarf virus(WYDV, 3.30%; Luteoviridae). Phylogenetic analysis showed CYDV and WYDV grouped separately from BYDVs. Furthermore, several recombination breakpoints were found among the groups of yellow dwarf viruses(BYDVs, CYDV, and WYDV). Using RNA deep sequencing, we confirmed the presence of the three known viruses(BYDVs, WSMV, and WDV) and the first record of two species of CYDV and WYDV in wheat in the Czech Republic.  相似文献

The present study compared the efficiency of transgenic (TG) cloned embryo production by somatic cell nuclear transfer (SCNT) with fetal-derived fibroblast cells (FFCs) which were transfected with pEGFP-N1 to in vitro-fertilized (IVF), parthenogenetic and SCNT counterparts by evaluating the rates of cleavage and blastocyst formation, apoptosis rate at different developmental stages, cell number, ploidy and gene expression in blastocysts. In SCNT and TG embryos, the rates of cleavage and blastocyst formation were significantly lower (p < 0.05) than those of IVF controls, but it did not differ between SCNT and TG embryos. In IVF control, 86.7% embryos displayed diploid chromosomal complements and the rates were significantly (p < 0.05) higher than those of SCNT and TG embryos. Most TG embryos (79%) with FFCs expressed the gene by both PCR and under fluorescence microscopy. The expression of apoptosis by TUNEL was first detected at six to eight cell stages in all embryos of IVF, SCNT and TG groups, but the expression rate at each developmental stages was significantly higher (p < 0.05) in SCNT and TG embryos than in IVF counterparts. The expression rate in inner cell mass (ICM) of TG embryos was significantly higher (p < 0.05) than in SCNT and IVF embryos. These results indicate that the high occurrence of apoptosis observed in SCNT and TG embryos compared with IVF counterparts might influence the developmental competence. Moreover, the SCNT embryos derived using non-transfected donor cells exhibited a lower apoptosis expression in ICM cells than in TG embryos derived using pEGP-N1-transfected donor cells suggesting a possible role of negative gene effect in TG embryos.  相似文献

Summary Response of dwarf wheat (Triticum aestivum L. em Thell) to three different levels of water stress at three growth stages — seeding to maximum tillering, maximum tillering to flowering and flowering to maturity, was studied under field conditions for two seasons. At each of these three stages, plants were subject to three ratios of irrigation water to cumulative pan evaporation (IW/CPE) –0.45, 0.60 and 0.75. During the remaining stages the plants were irrigated with an IW/CPE ratio of 0.9. Thus mild, moderate and severe stress treatments were compared with a no-stress control. At all stages moderate and severe water stress decreased plant height, leaf area, ear number, 1000-grain weight, grain yield and water-use efficiency. In stage 3 the effect of water stress on straw yield was not marked. Wheat was most sensitive to water stress during stage 1 when the reduction in grain yield was caused by a reduction in numbers of ears and grains per ear. In stage 2, grain yield reduction was due to fewer grains perear and a lower 1000-grain weight. On rewatering, mild stressed plants showed recovery of plant height, tiller number and in consequence, yield. Results indicate that under the conditions of this study the wheat crop should be irrigated at a IW/CPE ratio of 0.75 when water resources are limited. With an unlimited water supply the ratio may be increased to 1.2 in stage 2 to maximise the yield.  相似文献

The objective of the present study was to investigate the curative and antioxidative efficacy of ivermectin and ivermectin + vitamin E-selenium, and the influence of these agents on oxidative stress parameters in canines infested by Sarcoptes scabiei. Twenty two sarcoptic mites infested dogs and nine healthy dogs of 6 months to 2 years of age were divided into three groups. Group I comprised of healthy dogs (n = 9) whereas animals in group II (n = 11) and III (n = 11) were positive for scabies. Group II animals were treated with only 1% ivermectin @ 0.2 mg/kg SC whereas group III were additionally treated with Vitamin E and selenium (tocopherol 50 mg + Se 1.5 mg/ml) @0.5 ml/20 kg IM at weekly intervals for three times. Blood samples were collected on day 0 and 28 post therapy. The values for hemato-biochemical parameters and activities of antioxidant enzymes were significantly decreased (P < 0.05) whereas level of lipid peroxidation was significantly increased in all the infested dogs in comparison to the healthy dogs on day 0 which approached normalcy by day 28 post therapy. The dogs of group III showed better clinical recovery in comparison to group II at the end of therapy. Thus, administration of vitamin E and selenium in addition to standard therapy can alleviate these alterations hastening the clinical recovery of diseased dogs and can be recommended as an adjunct therapy with miticides for canine sarcoptic mange.  相似文献
