SUMMARYWe investigated the effects of dry-storage of Musa velutina seeds on seed and embryo germination. Seeds were collected immediately after harvest and stored in dry vermiculite at 25°C, in the dark, for 0, 1, 2, or 4 months. The germinability of dry-stored seeds was assessed by incubation in vermiculite at 25°C for 4 months, with 75% [(w/w); –0.01 MPa] moisture content to promote embryo development in viable seeds. Seeds that had not been dry-stored and stored in vermiculite at 75% (w/w) moisture content for 4 months showed 90% germination by 20 d after sowing, but no seeds that had been dry-stored for longer than 1 month germinated. Seeds were also dry-stored at 25°C, in the dark, in Petri dishes for 0, 1, 2, 3, or 4 weeks after harvest and the embryos removed from these seeds were cultured on MS medium. All embryos that had been isolated from seeds at harvest (i.e., 0 weeks in dry-storage) germinated during embryo culture. However, embryos removed from seeds that had been dry-stored for 1 week showed a substantial decrease in both size and germinability. Seeds were also stored at different moisture contents for 4 months after harvest. No seeds that had been stored at 0% moisture content (i.e., dry) germinated, but > 85% of seeds stored at ≥ 25% (w/w) moisture contents germinated. Higher moisture contents increased the uniformity of seed germination, showing that M. velutina seeds rapidly lose their germinability through dehydration, indicating that they are sensitive to desiccation. 相似文献

Our previous studies have found that (±)-(E)-12-hydroxyoctadec-10-enoic acid (HOEA) isolated from the red alga Tricleocarpa jejuensis showed cytotoxic effects on various living organisms including harmful microalgae, Gram-positive bacteria, and mammalian tumor cells. Since natural products with apoptosis-inducing ability can be promising anti-cancer agents, in this study, we investigated the cytotoxic mechanism of HOEA on U937 cells focusing on apoptosis induction. HOEA showed much stronger cytotoxic and cytolytic effects on U937 cells than elaidic acid, which has similar structure but no 12-hydroxy group, suggesting that hydroxy group is important for the cytotoxicity of HOEA. HOEA induced apoptotic nuclear morphological changes, DNA fragmentation, and decrease in mitochondrial membrane potential. Furthermore, time-dependent increase in annexin V+/PI+ cell population in HOEA-treated U937 cells was detected. Among the apoptosis-related reagents, caspase-family inhibitor almost completely inhibited HOEA-induced DNA fragmentation. In the analyses using specific caspase-substrates, extremely high cleavage activity toward caspase-3/7/8 substrate was observed in HOEA-treated U937 cells, and weak activities of caspase-1 and -3 were detected. Analyses using specific caspase inhibitors suggested that caspase-3 and caspase-8 might be predominantly responsible for the cleavage activity. Activation of these caspases were also confirmed by western blotting in which significant levels of cleaved forms of caspase 3, caspase 8, and PARP were detected in HOEA-treated U937 cells. Our results suggest that HOEA is capable of inducing apoptosis in U937 cells in which caspase-3 and caspase-8 might play important roles. Since the cytotoxic effect of HOEA is not strictly specific to tumor cells, development of appropriate drug delivery system for selective tumor targeting is necessary for the clinical applications to reduce the possible side effects. 相似文献

The pituitary gland is present in all vertebrates, from agnathans (jawless vertebrates) to mammals, but not in invertebrates. Reproduction in gnathostomes (jawed vertebrates) is controlled by two pituitary gonadotropins (GTHs), luteinizing hormone and follicle-stimulating hormone, which are part of the pituitary glycoprotein hormone (GPH) family. Hagfishes, which lack both jaws and vertebrae, are considered the most primitive vertebrate known, living or extinct. Accordingly, they are of particular importance in understanding the evolution of the pituitary GPHs and their functions related to vertebrate reproduction. Nevertheless, key elements of the reproductive endocrine system in hagfish have yet to be elucidated. Our current report has revealed the first identification of a functional GPH composed of two subunits that possess gonadotropic action at the pituitary of brown hagfish. It seems most likely that an ancestral GPH gave rise to only one GTH in hagfish pituitary and that multiplicity of GPHs arose later during the early evolution of gnathostomes. The daggertooth pike conger, Muraenesox cinereus (Forsskål), has become an important fish resource in the western Seto Inland Sea, Japan, since the 1990s. However, introducing sustainable fisheries resource management for this species is difficult in this region because stock assessments have not been performed, and official fisheries statistics for this stock were discontinued after 2007. This study used existing limited data sets to compile the first report for fisheries resource management for this M. cinereus stock. Yield‐per‐recruit analyses showed that increasing fishing pressure above current levels would provide only a minimal increase in expected catch levels. Hence, the current harvest level is considered to represent the upper limit of fishing pressure. Age composition in a given year could potentially be used to forecast landing abundance for the following 2 years. This study provides a basis for establishing effective fisheries resource management strategies for M. cinereus. 相似文献

For recovery of the declining population of pen shells in the wild, the production of pen shell juveniles for transplantation or aquaculture is underway in Japan. For more stable juvenile production, artificial fertilization methods for pen shells are needed, but methods to induce oocyte maturation (meiosis resumption) used in other bivalves, which make oocytes fertilizable, were ineffective for pen shells. Here, we report evidence showing that retinoic acid (RA) has strong activity in inducing oocyte maturation and activating sperm motility in pen shells. Treatment of fully developed oocytes with 1.0 μM all-trans-RA (at-RA) induced germinal vesicle breakdown, a typical morphological sign of oocyte maturation, but 1.0 μM at-retinol and at-retinal, 2 mM ammonia, and 1.0 μM serotonin were ineffective. Treatment with at-RA for 30 min was sufficient for oocyte maturation and was more potent than its isomers, 9-cis- and 13-cis-RA. Parallel results were obtained for sperm motility activation. Oocyte responsiveness to at-RA increased during the final stage of ovary development. Artificial fertilization was successful only with the oocytes treated with at-RA, and fertilized eggs developed to D-shaped (veliger) larvae without apparent morphological abnormalities. Long-term simulation using the distributed hydro-environmental watershed model is efficacious for assessing irrigation impacts
on hydrological cycle in detail and for implementing watershed management successfully. In this article, the previously developed
hydro-environmental watershed model (HEWM-1) is improved in the water exchange process caused by surface water-groundwater
interaction via drainage canals and/or underdrains. The time-varying stream flow in canals is described by the complete one-dimensional
shallow water equations in a newly introduced submodel, the open channel flow submodel. This submodel coordinates with the
other submodels: the tank, soil moisture and groundwater flow submodels which are interlinked in a cascade manner. The improved
model (HEWM-2) is applied to an agricultural watershed covering an area from an alluvial fan onto a nearly level alluvial
plain, to be validated. The simulation by HEWM-2 is informative for identifying whether any drainage canal is gaining or losing
water in relation to groundwater level. It could thus provide useful information for conserving a complex network of drainage

Indigestible oligosaccharides have been shown to normalize blood glucose and insulin concentration thereby promoting good health and preventing diseases, such as diabetes. Transglucosidase (TG, α-glucosidase, enzyme code (EC) 3.2.1.20) is an enzyme capable of converting starch to oligosaccharides, such as iso-malto-oligosaccharides from maltose, via the action of amylase. The aim of this study was to evaluate whether oral administration of TG with maltose or dextrin is capable of reducing post-prandial serum glucose concentration in experimentally streptozotocin (STZ)-induced diabetic dogs fed on a high-fiber diet. Five healthy and five STZ-induced diabetic dogs were employed in this study. TG supplementation with dextrin or maltose had no detrimental effect in healthy dogs. In fact, TG and dextrin exhibited a flatlined serum glucose pattern, while reducing mean post-prandial serum insulin and glucose concentration as compared to control diet alone. When TG supplementation was tested in STZ-induced diabetic dogs under the context of a high fiber diet, a 13.8% and 23.9% reduction in mean glucose concentration for TG with maltose and dextrin, respectively was observed. Moreover, TG with dextrin resulted in a 13% lower mean post-prandial glucose concentration than TG with maltose, suggesting that dextrin may be a more efficient substrate than maltose when used at the same concentration (1 g/kg). Our results indicate that TG supplementation with diet can lead to lower postprandial glucose levels versus diet alone. However, the efficacy of TG supplementation may depend on the type of diet it is supplemented with. As such, TG administration may be useful for preventing the progression of diabetes mellitus and in its management in dogs. 相似文献

A feeding trail was conducted to evaluate the effect of lowering dietary fishmeal (FM) levels while increasing levels of dehulled soybean meal (SBM) on growth, nutrient utilization and body composition of juvenile kuruma shrimp, Marsupenaeus japonicas. Five experimental diets were formulated to be isoenergetic, isolipidic and isonitrogenous with decreasing FM levels from 40 to 16% while increasing SBM from 0 to 33% respectively. Quadruplicate groups of shrimp (initial wt = 1.5 g) were fed the test diets for 56 days under the flow‐through system. There were no significant differences in final weight (g) and specific growth rate (SGR, % day) among shrimp fed FM40, FM34, FM28 and FM22 diets respectively. Growth parameters significantly decreased in shrimp when fed FM16 diet, which was the lowest level of FM. Feed intake was positively correlated with the SGR of shrimp, and the lowest one was found in shrimp fed FM16 diet. Protein gain and retention, whole body lipid, arginine and methionine significantly decreased in FM16 fed group. Thus, it is concluded that dietary FM could be reduced down to 22% with SBM without compromising growth, nutrient utilization and retention, and whole body composition of kuruma shrimp. 相似文献

BACKGROUND: Dibenzoylhydrazine analogues have been developed successfully as a new group of insect growth regulators, called ecdysone agonists or moulting accelerating compounds. A notable feature is their high activity against lepidopteran insects, raising the question as to whether species‐specific analogues can be isolated. In this study, the specificity of ecdysone agonists was addressed through a comparative analysis in two important lepidopterans, the silkworm Bombyx mori L. and the cotton leafworm Spodoptera littoralis (Boisd.). RESULTS: When collections of non‐steroidal ecdysone agonists containing different mother structures (dibenzoylhydrazine, acylaminoketone, tetrahydroquinoline) were tested, in vitro reporter assays showed minor differences using cell lines derived from both species. However, when compounds with high ecdysone agonist activity were examined in toxicity assays, larvicidal activity differed considerably. Of note was the identification of three dibenzoylhydrazine analogues with > 100‐fold higher activity against Bombyx than against Spodoptera larvae. CONCLUSION: The present study demonstrated that species‐specific ecdysone‐agonist‐based insecticides can be developed, but their species specificity is not based on differences in the activation of the ecdysone receptor but rather on unidentified in vivo parameters such as permeability of the cuticle, uptake/excretion by the gut or metabolic detoxification.
