ABSTRACT
Photosensitivity of flowering is the main yield limiting factor for soybean production in tropical areas. Our objective was to evaluate the variation of photosensitivity in the world soybean mini-core collections (GmWMC) under controlled environment. Ten and 13 h were selected as short- and long- photoperiods. The days from emergence to first flower open (DEF) were 20–49 days under 13 h, whereas 20–31 days under 10 h photoperiods. The variation in DEF under short photoperiod might be caused by juvenile growth phase or post-inductive phase, because 10 h was the photoperiod which induction phase of most genotypes were minimized. Index of photosensitivity of flowering (IPF) varied from 0.00 to 0.47 and correlated positively with DEF under short photoperiod. However, some genotypes were found having higher IPF but shorter DEF, or lower IPF but longer DEF. Results provided the valuable information for soybean production in tropical areas. 相似文献

The hydro-environmental aspects of a small, endangered wetland (Totsuru wetland), located in eastern Hokkaido, Japan, are investigated, and the efficacy of a proposed engineering strategy for restoring the wetland is examined. Taking 222Rn as a target tracer and building a static 222Rn mass balance equation, the share of groundwater-genetic surface water in yielding the entire surface water in the wetland is appraised. The share is also appraised by using a different approach based on a non-static, site-specific water balance model. Both approaches yielded nearly the same percentage share, and therefore, it is concluded that 70% or more of the surface water entering the Totsuru swamp through the natural rivers is groundwater genetic. Using the water balance model, it is deduced that the reverse flow from the Uenbetsu drainage river is not less than 30% of the entire surface water flow entering the swamp, and therefore contributes considerably to mitigating the shrinkage of the swamp. Finally, through numerical experiments, it is shown that raising the water level of the Uenbetsu drainage river with a constructed weir to enhance the reverse flow is efficacious for restoring the wetland. 相似文献

A sensitive and reproducible enzyme-linked immunosorbent assay (ELISA) using two
monoclonal antibodies directed against a synthetic peptide with an amino-acid sequence
related to the C-terminus of bovine myoglobin and the whole molecule of sodium dodecyl
sulphate (SDS)-denatured bovine myoglobin was adapted for detecting bovine myoglobin in
contaminated feeds. The ELISA employed bovine meat extract of a known myoglobin
concentration as a calibration standard and had an limit of detection (LOD) of 3.54
ng/ml and an limit of quantification (LOQ) of 11.0
ng/ml corresponding to 0.022% and 0.067% (wt/wt)
bovine meat-and-bone-meal (MBM) mixed in 20-fold-diluted feed extracts, respectively. A
cut-off threshold of 20.6 ng/ml bovine myoglobin was set
to simplify ELISA and facilitate quick assessment of test results without a tedious
calibration process. The ELISA was able to detect bovine MBM in artificially prepared
model feeds, mixed botanical feeds, mixed botanical feeds with skimmed milk, fish meal,
pork meal and pork/chicken meal at 0.1% (wt/wt). It was also able to detect sheep MBM in
test feeds, but showed no reactivity to swine MBM, chicken MBM, skimmed milk or gelatine
of bovine origin. The advantages of this method are the quick and easy extraction protocol
of proteins from test feeds, using 100 mM sodium sulphide and 0.6% sodium dodecyl sulphate
in the extraction solution and the effective detection of bovine and sheep MBM at 0.1%

In 2005 it was reported that the genetically modified (GM) maize strain or "event" called Bt10 had been distributed inadvertently in the United States over the previous 4 years. In order to ensure that grain for food and feed production did not contain trace amounts of Bt10 maize and complied with the applicable regulation, highly sensitive and specific detection of Bt10 maize was required. Accordingly, we developed a novel qualitative PCR system for specific detection of Bt10 maize. Moreover, we amply evaluated the performance characteristics of two PCR systems, our own and the one provided by the developer of Bt10, Syngenta Co. Ltd. It was confirmed that both of the qualitative PCR systems can specifically detect Bt10 maize, and the results of a single-laboratory examination suggested that the limit of detection was approximately less than 0.05% for both methods. To evaluate the reproducibility of the methods, we organized an interlaboratory study with the participation of 6 laboratories and analysis of 240 blind test samples. In this paper, we report, for the first time, the statistical analysis of the qualitative PCR data obtained from the interlaboratory study. The results of this analysis also revealed that there was no significant difference in the sensitivity between the two aforementioned methods and that the limit of detection of both the methods was less than 0.05%. Thus, we conclude that both of the methods are equally suitable for correct identification and sensitive detection of the unapproved GM maize Bt10 event in test samples. 相似文献

Plasma histamine concentrations (PHCs) were measured serially over 9 months or until death in 11 dogs with mast cell tumors (MCTs). Eight dogs had grossly visible disease and the other 3 dogs had microscopic disease. Initial PHCs in the dogs with gross disease were significantly higher than PHCs in healthy dogs (median, 0.73 ng/mL and 0.19 ng/mL respectively; P < .009), whereas initial PHCs in dogs with microscopic disease showed no difference from controls. Seven dogs subsequently had progressive increases in PHC, and developed hyperhistaminemia (median, 14.0 ng/mL; range, 5.11-30.1 ng/nL). These 7 dogs died from MCTs, and 1 had general weakness with rapid lysis of a large tumor burden after radiation therapy. PHCs of the other 4 dogs were less than 1 ng/mL during the study. These 4 dogs were still alive with adequate control of the tumor at the conclusion of the study. Four of the 11 dogs initially had gastrointestinal (G1) signs, which abated soon after administration of histamine-2 (H-2) blockers. No significant difference was found between PHCs in dogs with GI signs and those without GI signs (median, 0.86 ng/mL and 0.35 ng/mL. respectively). Thereafter, 7 dogs had serious GI complications for which H-2 blocker therapy was ineffective. PHCs in these 7 dogs were extremely high (median, 12.2 ng/mL; range, 3.42-30.1 ng/nL). Results of this study demonsrated that PHC was one factor related to disease progression, and indicated that marked hyperhistaminemia was associated with the GI signs refractory to H-2 blocker therapy in dogs with MCTs. 相似文献

The heating of the solar chromosphere and corona is a long-standing puzzle in solar physics. Hinode observations show the ubiquitous presence of chromospheric anemone jets outside sunspots in active regions. They are typically 3 to 7 arc seconds = 2000 to 5000 kilometers long and 0.2 to 0.4 arc second = 150 to 300 kilometers wide, and their velocity is 10 to 20 kilometers per second. These small jets have an inverted Y-shape, similar to the shape of x-ray anemone jets in the corona. These features imply that magnetic reconnection similar to that in the corona is occurring at a much smaller spatial scale throughout the chromosphere and suggest that the heating of the solar chromosphere and corona may be related to small-scale ubiquitous reconnection. 相似文献

Neoculin occurring in an edible tropical fruit is a heterodimeric protein which has both sweetness and a taste-modifying activity that converts sourness to sweetness. Both the primary and the overall tertiary structures of neoculin resemble those of monocot mannose-binding lectins. This study investigated differences in biochemical properties between neoculin and the lectins. Structural comparison between the mannose-binding sites of lectins and the corresponding regions of neoculin showed that there is at least one amino acid substitution at each site in neoculin, suggesting a reason for the lack of its mannose-binding ability. This was consistent with hemagglutination assay data demonstrating that neoculin had no detectable agglutinin activity. DNA microarray analysis indicated that neoculin had no significant influence on gene expression in Caco-2 cell, whereas kidney bean lectin (Phaseolus vulgaris agglutinin) greatly influenced various gene expressions. These data strongly suggest that neoculin has no lectin-like properties, encouraging its practical use in the food industry. 相似文献

Tomato plants (Lycopersicon. esculentum Mill cv. Momotaro) were cultured in nutrient solutions of varying concentrations or of varying nitrate (NO3) levels. The leaf blades and the main petioles including rachis were collected from the mixture of three successive leaves below each fruit truss at two growth stages. Comparisons were made between the nutrient concentration in the leaf blade and the petiole determined by plant sap analysis, and that in the leaf blade determined by the dry ashing method. Closely correlated relationships were found to exist between the phosphorus (P), calcium (Ca), and magnesium (Mg) concentrations in the leaf blade and the petiole determined by plant sap analysis, and those in the leaf blade determined by the dry ashing method at two growth stages. For the concentration of potassium (K), there was no correlation between results of plant sap analysis and those determined by the dry ashing method at either growth stage. 相似文献
