A sublethal dose (0.1 pmol per honeybee, i.e., approx. 0.9 pmol/g body wt) of the synthetic pyrethroid, deltamethrin, was injected intrathoracically into the hemocoel of emerging bees and the biochemical effects were determined over a period of 3 hr. The variations in hemolymph concentrations of trehalose, glucose, phospholipid, steroid, and diacylglycerol were limited, but large variations were observed for other lipids. Mono- and triacylglycerols increased dramatically 1.5 hr after injections; then a peak of free fatty acid appeared at the same time (3 hr) as the monoacylglycerols reached their maximum and the triacylglycerols started to decrease. Atypical forms of triacylglycerols accumulated from 1.5 hr to a maximum level 2 hr after injection. Then, after 3 hr, the concentrations of regular fatty acids, monoacylglycerols, and atypical triacylglycerols returned to normal while longer-chain fatty acids and different forms of ceride simultaneously appeared. The gut alkaline phosphatase parameters (VM, K, and Hill coefficient) also showed large variations 1.5 and 2 hr after injection, indicating an initial inhibition phase followed by superimposed induction and inhibition mechanisms. These data are discussed according to the hemolymph-lipid transport scheme from the biosynthetic to the utilization sites. Hormone release associated to mitochondrial alteration could be responsible for the observed effects. 相似文献

OBJECTIVE: To compare cardiovascular and systemic effects and analgesia during the postoperative period of epidural anesthesia performed with bupivacaine alone or with fentanyl or sufentanil in bitches maintained at a light plane of anesthesia with continuous infusion of propofol. STUDY DESIGN: Prospective randomized masked clinical trial. ANIMALS: 30 female dogs of various breeds. PROCEDURES: Dogs were allocated into 3 groups of 10 each. One group received fentanyl (2 microg/kg [0.91 microg/lb]) and bupivacaine (1 mg/kg [0.45 mg/lb]), 1 group received sufentanil (1 microg/kg) and bupivacaine (1 mg/kg), and 1 group received bupivacaine (1 mg/kg). All dogs received acepromazine (0.1 mg/kg [0.045 mg/lb]) and continuous infusion of propofol for sedation. The agents were administered into the lumbosacral space and diluted in saline (0.9% NaCl) solution to a total volume of 0.36 mL/kg (0.164 mL/lb). Cardiac and respiratory rates, arterial blood pressures, pH, and blood gases were evaluated. Analgesia, sedation level, serum cortisol concentrations, and plasma catecholamine concentrations were measured regularly for 6 hours. RESULTS: No important changes in cardiovascular, respiratory, or sedation variables were observed. Degree of analgesia in the postoperative period was higher in the sufentanil group, although use of fentanyl and bupivacaine also resulted in a sufficient level of analgesia. CONCLUSIONS AND CLINICAL RELEVANCE: Use of the 3 anesthetic techniques permitted ovariohysterectomy with sufficient analgesia and acceptable neuroendocrine modulation of pain with minimal adverse effects. 相似文献

Ivermectin is one of the most commonly used drugs in pharmacotherapy of parasitic diseases in domestic and wild animals caused
by parasitic nematodes and arthropods. However, ivermectin and other avermectins very often produce side-effects in hosts.
The most dominant clinical symptom of ivermectin toxicity in domestic and wild animals is CNS depression. In nematodes, the
target site of ivermectin’s action is glutamate-gated chloride-channel receptor and GABA receptor. The depressive effect of
ivermectin in mammals might include more than one mechanism; therefore, the anticonvulsive effect of ivermectin against convulsions
caused by lidocaine and strychnine was evaluated. Ivermectin antagonized lidocaine- and strychnine-induced convulsions in
rats, although these have different mechanisms. In the present study, the anticonvulsive ED50 of ivermectin for lidocaine-induced convulsions was 2.44 mg/kg (95% CL 1.67 to 3.57 mg/kg), whereas for convulsions induced
by strychnine it was higher at 4.25 mg/kg (95% CL 2.32 to 3.78 mg/kg). At the same time, both anticonvulsive doses are significantly
lower then the observed LD50 of ivermectin (18.20 mg/kg). Furthermore, flumazenil (0.1 and 0.2 mg/kg), an antagonist of benzodiazepine receptors, antagonizes
just one part of these anticonvulsive effects of ivermectin. Our results show the significant anticonvulsive properties of
ivermectin and support the findings that ivermectin in the CNS of mammals produces multiple inhibitory effects, probably through
participation in the function of GABA-sensitive and GABA-insensitive chloride channels. 相似文献

The broad spectrum, systemic fungicides tebuconazole and propiconazole are used to control rust in boronia (Boronia megastigma Nees). Gas chromatography combined with either a benchtop quadrupole mass spectrometer or a high-resolution mass spectrometer allowed for the monitoring of both pesticides in boronia leaves, flowers, and concrete. Field trials were established at two sites to determine the rate of dissipation of tebuconazole and propiconazole in boronia. At site 1, two application rates of 125 and 250 g active ingredient/hectare (ai/ha) tebuconazole were employed. Treatments were repeated 17 days later. At harvest, 286 days after the final application, tebuconazole was detected at levels of 0.06 +/- 0.05 and 0.5 +/- 0.1 [mg/kg +/- standard error, on a dry matter basis (DMB)] in the leaves collected from plots treated with 125 and 250 g ai/ha of tebuconazole, respectively. The oil produced from the flowers collected at the final harvest had residues of tebuconazole at levels of 0.06 +/- 0.03 and 0.10 +/- 0.08 mg/kg for the 125 and 250 g ai/ha application rates, respectively. Two repeat applications of 125 g ai/ha propiconazole were also used at site 1. Residues of propiconazole were detected at 0.09 +/- 0.03 mg/kg (DMB) 286 days after the final application. At site 2, treatments of 125 g ai/ha of tebuconazole were applied twice. At harvest, 279 days after the final application of tebuconazole, residues were recorded at 0.30 +/- 0.09 mg/kg in the leaves (DMB) while the oil produced had 0.20 +/- 0.07 mg/kg. 相似文献

The objective of this work was to evaluate rates for applications of water treatment sludge (WTS) as a nutrient source for grasses and leguminous plants cropped in a soil degraded by tin mining in the Amazon Region (Natural Forest of Jamari, Rondonia State, Brazil). The treatments consisted of three rates of nitrogen supplied by WTS (100, 150 and 200 mg kg−1 soil), five combinations of plants, two controls (absolute control, without fertilization; and chemical control, soil+lime+chemical fertilizers). Summary The occurrence of chromosomal interchanges was studied in F1 hyrids in two diallel schemes, including nine accessions of Triticum aestivum subsp. spelta, originating from Iran, Europe and North Africa, and the common wheat variety Chinese Spring as a reference. Seven accessions of subsp. spelta differed in one translocation from Chinese Spring. The European accession Grey differed in one or perhaps two interchanges from the reference variety, and Iran 417a showed a difference of three interchanges with Chinese Spring. In total six or perhaps seven interchanges were observed. Thus the number of translocation events in subsp. spelta appeared to be low. One translocation was unique and occurred only in the accession from North Africa. Also one accession from Europe, the landrace Schwarzwald, possibly carried a unique translocation. In both diallels an interchange with a relatively low coefficient of multivalent realisation at first meiotic metaphase was observed in most accessions. The two extra translocations in Iran 417a are considered to be the result of more recent translocation events, whereas the possible second translocation in accession Grey showed such a low coefficient of multivalent realisation that its existence could be doubted. 相似文献

We followed the contribution of released glucose to the formation of melanoidins in the maltose-glycine reaction by adding (14)C glucose to the maltose-glycine mixture, after it already had undergone some reaction. This approach allowed us to confirm the turnover of glucose in this reaction and hence the role of glucose in forming melanoidins. A comparison of the total amount of glucose converted into the melanoidins with the total concentration of melanoidins formed from maltose and glycine showed that the concentration of melanoidins originating from the released glucose was relatively small in comparison to the total melanoidins concentration. Hence, the parallel glucose-glycine reaction is considered to be only a minor pathway in the formation of maltose-glycine melanoidins. The incorporation of glucose into the nondialyzable melanoidins in the maltose-glycine reaction was in excellent agreement with the amount estimated from a kinetic model for the reaction of maltose with glycine. Understanding the mechanisms of phosphorus (P)-use efficiency (PUE) may contribute to enhancing crop P nutrition because species growth variability at low-P is well known. The experiment was carried out to evaluate the response of kale genotypes to different P supply in randomized block design in three replications. Low-P supply led to a decrease in most parameters, whereas an increase was recorded in root growth parameters. Genotypes differed in shoot dry weight (DW), leaf area, root length and area, and shoot and root P content and concentration. Root traits significantly positively correlated with PUE. Genotypes Red Russian (RR) and IJK 17 were superior in terms of shoot DW production at low P supply, and had the highest uptake efficiency. Genotypes IJK 17 and 81 had the highest P utilization efficiency, while Vates blue curled (VBC) showed the lowest PUE. Genotypes had similar shoot P content and concentration at low P supply, but large PUE differences, implying the importance of P utilization efficiency. 相似文献

Soils from field sites at Foulum (DK), Narbons (FR) and Varois (FR) planted with genetically modified maize expressing either the insecticidal Bacillus thuringiensis protein (Bt) or herbicide tolerance (HT), as described elsewhere in this volume, were analysed for nematodes, protozoa and microbial community structure. These analyses were mirrored in single-species testing and in mesocosm experiments, and were coordinated with field samples taken for microarthropods, enchytraeids and earthworms so allowing for cross-comparison and a better understanding of the results observed in the field. Over the first 2 years of the field experiments (in 2002 and 2003), the effect of Bt-maize was within the normal variation expected in these agricultural systems. Sampling in 2004 and 2005 was expanded to include the effects of tillage (i.e. reduced tillage versus conventional tillage) and also the use of HT-maize. Tillage had major effects regardless of soil type (Varois or Foulum), with reduced-tillage plots having a greater abundance of microfauna and a different microbial community structure (measured both by phospholipid fatty-acid analysis (PLFA) and by community-level physiological profiling (CLPP)) from conventionally tilled plots. Grass, as a contrasting cropping system to maize, also had an effect regardless of soil type and resulted in greater microfaunal abundance and an altered microbial community structure. Differences in crop management, which for the Bt-maize was removal of the insecticide used to control European corn borer and for HT-maize was a change in herbicide formulation, were only tested at single sites. There were differences in microbial community structure (CLPP but not PLFA) and sporadic increases in protozoan abundance under the Bt-crop management. The HT-maize cropping system, which covered a shorter period and only one site, showed little change from the conventional system other than an altered microbial community structure (as measured by PLFA only) at the final harvest. The Bt-trait had a minimal impact, with fewer amoebae at Foulum in May 2003, fewer nematodes at Foulum in May 2004 but more protozoa at Varois in October 2002 and an altered microbial community structure (PLFA) at Foulum in August 2005. These were not persistent effects and could not be distinguished from varietal effects. Based on the field evaluations of microfauna and microorganisms, we conclude that there were no soil ecological consequences for these communities associated with the use of Bt- or HT-maize in place of conventional varieties. Other land management options, such as tillage, crop type and pest management regime, had significantly larger effects on the biology of the soil than the type of maize grown. 相似文献
