A simple capillary electrophoretic method is described for the simultaneous determination of soluble oxalate and nitrate in forage grasses. Grass samples were ground and extracted with water. The extracts were filtered and submitted to capillary electrophoresis. Electrophoresis was performed in a 75 microm x 50 cm fused silica capillary with 30 mM sodium sulfate containing an electroosmotic flow modifier under constant voltage at -8 kV. Separated oxalate and nitrate were detected with direct UV absorption at 214 nm. The present method can be used for routine monitoring of the concentration of soluble oxalate and nitrate in grasses.  相似文献

In order to analyze the behavior and phytoxicity of iodine in soil, the chemical forms of soil iodine must be identified. Therefore, a method for quantitative speciation of iodine in soil was proposed. Iodine extracted from soil samples with tetrametBPyIammonium hydroxide (TMAH) was separated into humic and fnalvic acid fractions at pH4 1.5 after the addition of ascorbic acid into the TMAH extract to reduce iodate into iodide. Since the iodide in the TMAH extract was recovered in the fdvic acid fraction by this procedure, iodine contained in the haamic acid fraction was considered to be organically bound. Podine in the fulvic acid fraction was separated into organic iodine bound to fnlvic acids and the total inorganic iodine. Furthermore, iodine soluble from soil in 0.1 mol L-1 potassium chloride was assumed to correspond to the amount of total iodide in soil, and from the difference in the concentration of total inorganic iodine and soluble iodide, the amount of iodate was calculated. By the application of this method, iodine in soil was separated into four fractions: organic iodine bound to humic acids, organic iodine bound to fulvic acids, iodate, and iodide. In order to understand the present status of heavy metal contamination and the risk associated with human consumption, commercial freshwater eels in Vietnam were examined. The concentration of eight elements (Cr, Mn, Co, Cu, Zn, Sr, Cd, and Pb) was determined in muscle tissue of tropical eel Anguilla marmorata collected in four provinces, Quang Tri, Quang Ngai, Binh Dinh, and Phu Yen, from the central part of Vietnam. Concentrations of Cu, Zn, Cd, and Sr were found to be highest, whereas Co concentration showed the lowest at Quang Tri. Cr was recorded to be lowest in Quang Ngai, while the amounts of Mn and Pb did not vary among locations. Although the metal concentrations in the muscle of A. marmorata did not represent a risk for human consumption, elevated Cd concentrations in Quang Tri related to agriculture activities suggest that further investigations should be undertaken to clarify the potential risk to the eel population.  相似文献

To elucidate the structural changes in pink (P), white (W), and red (R) muscles during storage in ice, we measured the breaking strength and changes in pericellular connective tissues of cultured carp. The breaking strength just after killing was highest in R muscle (1.00 ± 0.20 N), lowest in W muscle (0.37 ± 0.07 N), and intermediate (0.84 ± 0.12 N) in P muscle. During the storage period, the breaking strength decreased first in R muscle, then in P muscle, followed by W muscle. The diameter of muscle fibers was greater in W muscle (113 ± 15 μm) than in P muscle (72 ± 3 μm) and R muscle (48 ± 2 μm). Destruction of the honeycomb structure of the pericellular connective tissue occurred most rapidly in W muscle and most slowly in R muscle. These results suggest that the interposing of P muscle fibers in the dorsal ordinary muscle contributes to the acceleration of post-mortem tenderization in fish.  相似文献

Since 2006, a commercial grower in Japan has noted a leaf blight symptom on potted cosmos plants grown in a field. In August 2012, a Pseudomonas-like bacterium was isolated from the symptomatic leaves and found to cause the same symptom on cosmos seedlings after inoculation. On the basis of bacteriological and phylogenetic analyses, the causative bacterium was identified as Pseudomonas cichorii. This is the first report of bacterial leaf blight on cosmos caused by P. cichorii in Japan.  相似文献

The aim of this study was to ultrasonographically evaluate curd formation in 29 pre-ruminant calves that were fed a clotting milk replacer. Abomasal curd was absent in 8/29 calves at 2 h after feeding. In these eight calves, abomasal contents were observed as an anechoic image with small echogenic spots (five calves), or as an echogenic image with an unclear outline (three calves), but there was no echogenic image with a clear outline corresponding to curd that was visualised in the other 21 calves. The curd was not observed until 7 h after feeding in the eight calves. Our analysis also indicated that the absence of curd formation in the pre-ruminant calves did not have a significant impact on the appearance, appetite and vigour of pre-ruminant calves or on their blood parameters, including serum triglyceride, blood urea nitrogen and glucose concentrations. The study provided the first evidence that the abomasum of some calves does not form curd despite ingestion of a clotting milk replacer.  相似文献
