Insulin resistance is a major hallmark in the development of type II diabetes, which is characterized by the failure of insulin to promote glucose uptake in muscle and to suppress glucose production in liver. The serine-threonine kinase Akt (PKB) is a principal target of insulin signaling that inhibits hepatic glucose output when glucose is available from food. Here we show that TRB3, a mammalian homolog of Drosophila tribbles, functions as a negative modulator of Akt. TRB3 expression is induced in liver under fasting conditions, and TRB3 disrupts insulin signaling by binding directly to Akt and blocking activation of the kinase. Amounts of TRB3 RNA and protein were increased in livers of db/db diabetic mice compared with those in wild-type mice. Hepatic overexpression of TRB3 in amounts comparable to those in db/db mice promoted hyperglycemia and glucose intolerance. Our results suggest that, by interfering with Akt activation, TRB3 contributes to insulin resistance in individuals with susceptibility to type II diabetes.  相似文献

A series of monoazo disperse dyes with a photostablilizing o-hydroxycarbonyl moiety was incorporated resulted in the enhancement of the light fastness properties on polyester and nylon substrates as compared to the dyes that do not possess the photostablilizing moiety. The geometries of the azo and the hydrazone tautomeric forms of all dyes were optimized at B3LYP/6-311+G(d) and electrophilicity index was calculated for propensity of the moiety to absorb electrons. The values of absorption and stability trend of the dyes were in good agreement with the trend of experimental light fastness values. These disperse dyes possess excellent wash fastness and moderate to good sublimation fastness on hydrophobic substrates.  相似文献

本文对印度西部5种热带干落叶林中凋落物里生物元素的季节性变化和它们返回到新鲜叶中季节性变化进行研究。对5个树种:阿拉伯金合欢(Acacia nilotica L.)、金合欢(Acacia leucophloea Roxb.)、柚木(Tectonagrandis L.f.)、绒毛野独活(Miliusa tomentosa Roxb.)和紫矿(Butea mono-sperma Lain.)中凋落物的成熟叶、枝和再生部分的N、P、Ca、Mg、K和Na的浓度,以及老叶和幼叶中浓度间的关系进行调查。对这些树种的总干物质进行记录,并分析它们所含的N、P、Ca、Mg、K和Na的含量。其中阿拉伯金合欢叶子中N的含量最高,而金合欢的叶子里Ca和Mg含量最高。叶中含P最多的是阿托伯金合欢、金合欢和紫矿,而含P最少的是柚木和绒毛野独活。K和Na含量在这些树种中没有明显的区别。观察发现,N、P和K的浓度随季节变化大,但Ca和Mg则不然。唯有钾在所有树种中都是最稳定的,吸收N和P可以用于来年新叶的生长。从老叶和凋落物中吸收的营养会支持新叶的生长,并增加土壤的肥力。图1表4参30。  相似文献

Adipose tissue-derived stromal vascular fraction (AdSVF) comprises a heterogeneous cell population, including the multipotent mesenchymal stem cells, hematopoietic stem cells, immune cells, endothelial cells, fibroblasts, and pericytes. As such, multipotent adipose tissue-derived mesenchymal stem cells (AdMSCs), are one of the important components of AdSVF. Commonly used techniques to harvest AdSVF involve enzymatic or non-enzymatic methods. The enzymatic method is considered to be the gold standard technique due to its higher yield. The cellular components of AdSVF can be resuspended in normal saline, platelet-rich plasma, or phosphate-buffered saline to produce a ready-to-use solution. Freshly isolated AdSVF has exhibited promising osteogenic and vasculogenic capacity. AdSVF has already been proven to possess therapeutic potential for osteoarthritis management. It is also an attractive therapeutic option for enhancing wound healing. In addition, the combined use of AdSVF and platelet-rich plasma has an additive stimulatory effect in accelerating wound healing and can be considered an alternative to AdMSC treatment. It is also widely used for managing various orthopaedic conditions in clinical settings and has the potential for regenerating bone, cartilage, and tendons. Autologous AdSVF cells are used along with bone substitutes and other biological factors as an alternative to conventional bone grafting techniques owing to their promising osteogenic and vasculogenic capacity. It can also be used for treating osteonecrosis, meniscus tear, chondromalacia, and tendon injuries in veterinary practice. It has several advantages over in vitro expanded AdMSC, including precluding the need for culturing, reduced risk of cell contamination, and cost-effectiveness, making it ideal for clinical use.  相似文献

Lines of Brassica juncea with tetralocular (four-valved) pods were produced by interspecific hybridization between dilocular (two-valved) B. juncea var. Pusa Barani and tetralocular yellow-seeded turnip rape (Brassica rapa ssp. trilocularis) var. YID-1. Some selected lines were significantly superior to their parents for some agronomic traits, including seed yield and harvest index. Line 11 was the best among these and recorded 70% yield advantage in plot yield.  相似文献

During fasting, increased concentrations of circulating catecholamines promote the mobilization of lipid stores from adipose tissue in part by phosphorylating and inactivating acetyl-coenzyme A carboxylase (ACC), the rate-limiting enzyme in fatty acid synthesis. Here, we describe a parallel pathway, in which the pseudokinase Tribbles 3 (TRB3), whose abundance is increased during fasting, stimulates lipolysis by triggering the degradation of ACC in adipose tissue. TRB3 promoted ACC ubiquitination through an association with the E3 ubiquitin ligase constitutive photomorphogenic protein 1 (COP1). Indeed, adipocytes deficient in TRB3 accumulated larger amounts of ACC protein than did wild-type cells. Because transgenic mice expressing TRB3 in adipose tissue are protected from diet-induced obesity due to enhanced fatty acid oxidation, these results demonstrate how phosphorylation and ubiquitination pathways converge on a key regulator of lipid metabolism to maintain energy homeostasis.  相似文献

Cholinergic neurons are widespread, and pharmacological modulation of acetylcholine receptors affects numerous brain processes, but such modulation entails side effects due to limitations in specificity for receptor type and target cell. As a result, causal roles of cholinergic neurons in circuits have been unclear. We integrated optogenetics, freely moving mammalian behavior, in vivo electrophysiology, and slice physiology to probe the cholinergic interneurons of the nucleus accumbens by direct excitation or inhibition. Despite representing less than 1% of local neurons, these cholinergic cells have dominant control roles, exerting powerful modulation of circuit activity. Furthermore, these neurons could be activated by cocaine, and silencing this drug-induced activity during cocaine exposure (despite the fact that the manipulation of the cholinergic interneurons was not aversive by itself) blocked cocaine conditioning in freely moving mammals.  相似文献

A methodology to predict the draft requirements of combination tillage implements in any soil and operating conditions was developed. This methodology required the draft requirements of individual tillage implements in undisturbed soil condition and draft utilization ratio of the rear passive set of combination tillage implement, which is defined as the ratio of the drafts of the rear passive set operating in combination and individually. Laboratory experiments were conducted to measure the draft requirements of a reference tillage tool (single disk), three scale-model individual (moldboard plow, cultivator and disk gang) and two combination (moldboard plow with disk gang and cultivator with disk gang) tillage implements at different depths (5, 7.5 and 10 cm), speeds (1.2, 2.2, 3.2 and 4.2 km/h), wet bulk densities (in the range of 1.27–1.85 g/cm3) and cone index penetration resistance values (in the range of 445–1450 kPa) in soil bin filled with sandy clay loam soil. The average draft utilization ratio of the reference tillage tool obtained were analyzed by both orthogonal and multiple regression techniques to develop the regression equation considering soil properties, operating and tool parameters. The developed draft equation based on the above mentioned methodology was verified with the data obtained for the draft of scale-model and prototype combination tillage implements in the laboratory and field conditions, respectively. It was found that the developed equation predicted the draft of both combination tillage implements within an average absolute variation of 18.0 and 13.5%, respectively.  相似文献
